Project description:Cancer associated fibroblasts (CAFs) have been shown to plays crucial roles in cancer progression. Although increasing evidence demonstrates that CAFs have important roles in modulating the aggressive phenotypes of cancer cells, their effects on the tumor vasculature remain underexplored. We co-cultured TIME human microvascular endothelial cells (MECs) with either primary human ovarian CAFs or normal ovarian fibroblasts (NFs) to evaluate the effects of CAFs on phenotypes of endothelial cells.

Thus, RNA-seq was carried to analyze the transcriptome differences among fibroblasts cultured alone and fibroblasts co-cultured with MSCs to find detailed mechanisms about how fibroblasts had functions on MSCs. We then performed gene expression profiling analysis using data obtained from RNA-seq of fibroblasts cultured alone or co-cultured with MSCs.

Project description:Patients with Lymphangioleiomyomatosis (LAM) develop nodules of cells within the lung parenchyma. These lesions contain both TSC2-/- LAM cells, and wild type fibroblasts. We investigated the effect of TSC2-/- cells on the transcriptional profile of both normal fibroblasts and fibroblasts isolated from LAM patient tissue (LAFs) using Affymetrix Gene Chip arrays, to identify upregulated pathways which could contribute to LAM pathology. We used Affymetrix microarrays to detail the changes in the global profile of gene expression in fibroblasts following co-culture with 621-101 cells, and identified upregulation of several chemokines, indicating that the fibroblasts had adopted an inflammatory phenotype.
