Project description:We used adult male Sprague-Dawley rats (280-329 g body weight). Controls were naïve rats. Ischemic rats were subjected to 1-hour occlusion of the right middle cerebral artery and 16h reperfusion. The goal was to identify the changes in the gene expression profile of PV after brain ischemia.

Project description:Immune response following CNS disease and injury comprises three different compartments, parenchymal, perivascular and blood involving specialized immune cells. Microglia are the major immune cell residing in the CNS parenchymal compartment but together with other macrophages which are residing in the non-parenchymal structures such as perivascular spaces, leptomeninges and choroid plexus constitute the total macrophage population of the CNS. While the homeostatic functions of these specialized macrophages in not very well understood, evidence shows that these macrophages at the blood-brain interface might involve immune-surveillance and establish a gate way for the recruitment of peripheral immune cells in to the CNS in response to the pathological stimuli. In humans and rats these macrophages identified exclusively by the high levels of scavenger receptor CD163

Perivascular adipose tissue inflammation in ischemic heart disease

Project description:We performed microarray analyses comparing perivascular adipose tissue gene expression of thoracic aortic vs. left internal mammary artery in human subjects with coronary artery disease. 300 genes were significantly upregulated in aortic fat, with at least 63 genes involved in inflammation and atherosclerosis. qPCR validation demonstrated increased expression of WNT-5A related genes, and CCL-2, CCL-8, JNK, and IL-6 gene expression.

Meningeal cells contribute to postnatal cortical neurogenesis.

Project description:Postnatal brain neurogenesis in mammals is believed to be restricted to rare germinative remnants of the neuroepithelium. In this study, we discovered that, in the postnatal brain, a subset of embryonically derived progenitors is present in meningeal substructures. These cells migrate from the meningeal substructures to the retrosplenial and visual motor cortex and differentiate into (electrically) functional integrated neurons. Lineage tracing analysis revealed that this subset of neural progenitors originate largely from PDGFR+ cells. PDGFR-derived cells differentiate mostly into Satb2+ neurons in cortical layers I-IV. Thus, a reservoir of embryonically derived progenitors in the meninges contributes to postnatal cerebral cortical neurogenesis.

Project description:All tissue resident macrophages of the central nervous system (CNS), including parenchymal microglia as well as CNS-associated macrophages (CAMs) such as meningeal (mMF) and perivascular macrophages (pvMF) are part of the CNS endogenous innate immune system that acts as the first line of defense during infections or trauma. It has been suggested that microglia and all CAM subsets are derived from prenatal cellular sources in the yolk sac that were defined as early erythromyeloid progenitors. However, the precise ontogenetic relationships, the underlying transcriptional programs and the molecular signals that drive the development of distinct CAMs subsets in situ are poorly understood. Using novel fate mapping systems, single-cell profiling and cell-specific mutants, we show that only mMF and microglia share a common prenatal progenitor. In contrast, pvMF originate from perinatal mMF only after birth in an integrin-dependent manner. Furthermore, the establishment of pvMFcritically requires the presence of vascular smooth muscle cells. In summary, our data reveal a novel precisely timed process in distinct anatomical niches for the establishment of CNS macrophage subsets.

Single-cell atlas reveals meningeal leukocyte heterogeneity in the developing mouse brain

Project description:The meninges are important for brain development and pathology. Using single-cell RNA-sequencing, we have generated the first comprehensive transcriptional atlas of neonatal mouse meningeal leukocytes under normal conditions and after perinatal brain injury. We identified almost all known leukocyte subtypes and found differences between neonatal and adult border-associated macrophages, thus highlighting that neonatal border-associated macrophages are functionally immature with regards to immune responses compared to their adult counterparts. We also identified novel meningeal microglia-like populations that appear to participate in white matter development. Early after hypoxic-ischemic insult, neutrophil numbers increased and they exhibited increased granulopoiesis, suggesting that the meninges are an important site of immune cell expansion with implications for the initiation of inflammatory cascades after neonatal brain injury. Our study provides a single-cell-resolution view of the importance of meningeal leukocytes at different stages of development in health and disease. DOI:10.1101/gad.348190.120
