Project description:Arabidopsis Affymetrix ATH1 GeneChips were used to compare the mRNA profiles of root tissues of the transgenic plants overexpressing 4D09 effector gene from the cyst nematode Heterodera schachtii and the wild-type (C24). Also, Arabidopsis Affymetrix ATH1 GeneChips were used to compare the mRNA profiles of root tissues of the transgenic plants overexpressing 14-3-3Ɛ gene from Arabidopsis and the wild-type (Col-0). Wild-type (Arabidopsis thaliana, ecotypes C24 and Col-0 ), and the transgenic plants overexpressing 4D09 effector gene or overexpressing 14-3-3Ɛ gene from Arabidopsis were grown in vertical culture dishes on modified Knop’s medium for 2 weeks and then root tissues were collected for RNA extraction. ****[PLEXdb(http://www.plexdb.org) has submitted this series at GEO on behalf of the original contributor, Tarek Hewezi. Project description:In this experiment we evaluate the transcriptional response to beet cyst nematode infection in wild-type (Col-0) and siz1 a knocked-down Arabidopsis thaliana (siz1-2). The data indicates that the mutation in siz1-2 leads to a strong transcriptional regulation in the plant, mostly of genes related to plant defense. The interaction of the mutation and infection by H. schachtii results in differential regulation of a small subset of genes. Samples were taken from whole root systems of mock or H. schahtii-infected 21 day-old seedlings (7 days post-infection). Each sample contained at least 18 plants and 3 biological replicates of each sample were collected.

We have used RNAseq to study the gene expression profile in the roots and shoots of the pho1 mutant, two independent transgenic lines of pho1 expressing the EXS domain of PHO1 (lines EXS32 and EXS34) as well as a pho1 mutant transformed with the full length PHO1 gene (complemented line).

Project description:In this experiment we evaluate the transcriptional response to beet cyst nematode infection in wild-type (Col-0) and a upl3 knocked-down Arabidopsis thaliana. The data indicates that the upl3 knock-down alone does not induce a strong transcriptional regulation in the plant. However, the interaction of the mutation and infection by H. schachtii results in a strong transcriptional differential regulation, likely involving the plant’s immune responses. Samples were taken from whole root systems of 14 day-old seedlings (0 days post-infection) and mock or H. schahtii-infected 21 day-old seedlings (7 days post-infection). Each sample contained at least 18 plants and 4 biological replicates of each sample were collected.

RNA-seq analysis of Arabidopsis thaliana wild-type roots and type-A arr3,4,5,6,7,8,9,15 mutant roots non-infected and infected with Heterodera schachtii nematodes

Project description:Plant-parasitic cyst nematodes induce the formation of hypermetabolic feeding sites, termed syncytia, as their sole source of nutrients. The formation of the syncytium is orchestrated by the nematode in part by modulation of phytohormone responses, including cytokinin. In response to infection by the nematode H. schachtii, cytokinin signaling is transiently induced at the site of infection and in the developing syncytium. Arabidopsis lines with reduced cytokinin sensitivity show reduced susceptibility to nematode infection, indicating that cytokinin signaling is required for optimal nematode development. Furthermore, lines with increased cytokinin sensitivity also exhibit reduced nematode susceptibility. To ascertain why cytokinin hypersensitivity reduces nematode parasitism, we examined the transcriptomes in wild-type and a cytokinin-hypersensitive type-A arr Arabidopsis mutant in response to H. schachtii infection. Genes involved in the response to biotic stress and defense response were elevated in the type-A arr mutant in the absence of nematodes and were hyper-induced following H. schachtii infection, which suggests that the Arabidopsis type-A arr mutants impede nematode development because they are primed to respond to pathogen infection. These results suggest that cytokinin signaling is required for optimal H. schachtii parasitism of Arabidopsis, but that elevated cytokinin signaling triggers a heightened immune response to nematode infection.
