Project description:NILs were created for the detailed genetic mapping of a QTL (gFW9.1) in tomato. Residual heterozygocity in recombinant inbred lines from the cross S. lycopersicum var. cerasiforme (parental accession E9) × S. pimpinellifolium line (parental accession L5) was used for the generation of a pair of NILs of which line E9 had DNA from parent E9 on a large section of chromosome 9 (5-56 Mb), whereas line L5 had parent L5 DNA. These two lines were tested as rootstocks grafted to a common scion (cv. Boludo) in multi-stress conditions - low phosphorus and drought stress compared to high phosphorus and well-watered conditions. RNA for transcriptomic analysis was prepared from root tissues. Secondary use of data: In a study aiming to identify genes that respond to multiple abiotic stresses, microarray data obtained from different plant species and under different stresses was analysed. A number of conserved stress-responsive genes were identified whose expression was differentially regulated in tomato roots in response to one or several stresses. 10 of these genes were validated as reliable biomarkers whose expression levels are related to different signalling pathways involved in adaptive stress responses. This dataset comprises part of the full stress dataset and is for plant roots harvested after growth under low phosphorus and drought conditions, compared to plant roots harvested after growth under sufficient phosphorus and well watered conditions.

Project description:Plants and rhizosphere microbes rely closely on each other, with plants supplying carbon to bacteria in root exudates, and bacteria mobilizing soil-bound phosphate for plant nutrition. When the phosphate supply becomes limiting for plant growth, the composition of root exudation changes, affecting rhizosphere microbial communities and microbially-mediated nutrient fluxes. To evaluate how plant phosphate deprivation affects rhizosphere bacteria, Lolium perenne seedlings were root-inoculated with Pseudomonas aeruginosa 7NR, and grown in axenic microcosms under different phosphate regimes (330 uM vs 3-6 uM phosphate). The effect of biological nutrient limitation was examined by DNA microarray studies of rhizobacterial gene expression.

Project description:Possitive effects of plant growth promoting bacteria (PGPB) inoculation on plant growth and development are dependent on interaction between bacterial strains and plant roots, which are usually the bacterial niche. Furthermore, phytohormones are key regulators of plant physiology. Ethylene is essential in plant growth and development and in response to drought. Plant sensibility to ethylene is involved in plant response to PGPB strain inoculation and plant growth promotion. We used microarrays to detail the global programme of gene expression underlying plant interaction with two different PGPB strains (isolated from arid soils in southern Spain) regarding to plant sentitivity to ethylene by tomato ethylene receptor 3 (SlETR3).

Project description:Nitrogen (N) and phosphorus (P) are the most limiting factors for plant growth. Some microorganisms improve the uptake and availability of N and P, minimizing chemical fertilizer dependence. It has been published that the RD64 strain, a Sinorhizobium meliloti 1021 strain engineered to overproduce indole-3-acetic acid (IAA), showed improved nitrogen fixation ability as compared to the wild type 1021 strain. We present here data showing that RD64 is also highly effective in mobilizing P from insoluble sources such as phosphate rock (PR). Under P-limiting conditions, the higher P-mobilizing activity of RD64, as compared to the 1021 wild type strain, is connected with the up-regulation of genes coding for the high-affinity P transport system, the induction of acid phosphatase activity and the increased secretion into the growth media of malic, succinic and fumaric acids. Medicago truncatula plants nodulated by RD64 (Mt-RD64), when grown under P-deficient conditions, released higher amounts of another P-solubilizing organic acid, the 2-hydroxyglutaric acid, as compared to the plants nodulated by the wild-type strain (Mt-1021). It has already been shown that Mt-RD64 plants exhibited a higher dry weight production as compared to Mt-1021 plants. Here we report that also P-starved Mt-RD64 plants show a significant increase both in shoot and root fresh weight when compared to P-starved Mt-1021 plants. We discuss how, in a rhizobium-legume model system, a balanced interplay of different factors linked to the bacterial IAA over-production rather than IAA production per se stimulates plant growth under stressful environmental conditions, and in particular, under P-starvation. Two-conditions experiment: untreated vs. IND-treated cells. Biological replicates: 6 untreated control, 6 treated samples, independently grown and harvested. One replicate per array.
