Project description:Wheat, an important human food crop, is very sensitive to temperature changes. Thermosensitive sterile lines (TMS) is a natural material for exploring the effects of anther development and temperature changes on male fertility. In order to study the possible molecular mechanism of regulating protein activity during anther fertility alteration, proteomics analysis based on high resolution mass spectrometry were used to study the binuclear anther of wheat YS3038.

Project description:Durum wheat (Triticum turgidum subsp. durum) is widely grown for pasta production, and more recently, is gaining additional interest due to its resilience to warm, dry climates and its use as an experimental model for wheat research. To enable further research into endosperm development and storage reserve synthesis, we generated a high-quality transcriptomics dataset from developing endosperms of durum variety Kronos, to complement the extensive mutant resources available for this variety. Endosperms were dissected from grains harvested at eight timepoints during grain development (6 to 30 days post anthesis (dpa)), then RNA sequencing was used to profile the transcriptome at each stage. The largest changes in gene expression profile were observed between the earlier timepoints, prior to 15 dpa. We detected a total of 29,925 genes that were significantly differentially expressed between at least two timepoints, and clustering analysis revealed nine distinct expression patterns. Overall, we provide a valuable resource for studying endosperm development in this increasingly important crop species.

Project description:The analysis of gene expression during wheat development: Gene expression measurements were carried out on a developmental tissue series for wild-type wheat (cv. Chinese Spring) using the Affymetrix Wheat GeneChip. Thirteen tissues at defined developmental stages were chosen to match the barley (cv. Morex) tissue series of Druka et al. 2006 that used the Affymetrix Barley1 GeneChip. Three replicates of: root tissue at two different developmental stages, leaf, crown, caryopsis, anther, pistil, inflorescence, bracts, mesocotyl, endosperm, embryo and coleoptiles were hybridised. Project description:The RNA sequencing analysis was undertaken to investigate the transcriptomic changes in adult wheat inoculated with Puccinia graminis f. sp. tritici the causal agent of stem rust disease. The project firstly aims to compare gene expression in one susceptible wheat line with two wheat lines exhibiting adult plant resistance to the stem rust. Secondly, the project aims to examine the temporal changes in gene expression in wheat after inoculation. Wheat plants was grown until maturity under greenhouse conditions. Plants were inoculated with Puccinia graminis f. sp. tritici and the flag leaf sheath sampled for RNA sequencing. The project aims to give essential insight into the adult plant resistance response in wheat to Puccinia graminis f. sp. tritici inoculation.

Project description:In the current project, we aimed to discover how the barley anther proteome changes, both quantitatively and qualitatively, during meiotic prophase I when the important process of homologous recombination is taking place. Taking advantage of a recently published barley anther transcriptomic dataset (BAnTr) and a newly developed sensitive mass spectrometry-based approach to analyse barley anther proteome, we have conducted high-resolution mass spectrometry analysis of barley anthers, collected at 6 time points and representing their development from pre-meiosis to metaphase. Each time point was carefully staged using immunocytology. We identified >6400 proteins, including 82 known and putative meiotic proteins, and were able to quantify dynamic variation in the proteome during different stages of barley anther development.

Project description:Resistance mechanisms of wheat to Fusarium graminearum are still poorly understood. Multiple reports have investigated transcriptomic differences between resistant and susceptible genotypes. To investigate the genetic determinants underlying the two major contributor QTL Fhb1 and Qfhs.ifa-5A, we generated a set of near-isogenic lines, that we have used for transcriptomic studies using the Affymetrix wheat GeneChip®: 4 near-isogenic lines (BC5F2) derived from a cross of CM82036 and cv Remus (susceptible recurrent parent) and the resistant parent CM82036 have been inoculated with Fusarium spore suspension or water as control at anthesis. Samples have been taken at 3 timepoints 8, 24 and 72 hours after inoculation. Five wheat heads per condition were pooled into one sample, that has been used for RNA preparation. Including 3 replicates a total of 90 microarrays have been hybridized. Our results identify a wide variety of genes (631 transcripts) that are differentially regulated for Fusarium graminearum in all genotypes after 72 h and 339 genes that are upregulated only in lines without Qfhs.ifa.5A. Few genes were found differentially regulated for Fusarium and QTL.
