Project description:Plants sense light and temperature changes to regulate flowering time. The expression of the florigen gene, FLOWERING LOCUS T (FT), peaks in the morning during spring, a different pattern than we observe in the lab. Providing our lab growth conditions with a red/far-red light ratio similar to open field conditions and average natural temperature oscillation is sufficient to mimic the FT expression and flowering time in natural long days. Here, we use RNA-seq to identify and understand the molecular differences between natural growth conditions, conventional lab growth conditions, and supplemented lab growth conditions that mimic natural conditions. Project description:Plants sense light and temperature changes to regulate flowering time. Here, we show that expression of the Arabidopsis florigen gene, FLOWERING LOCUS T (FT), peaks in the morning during spring, a different pattern than we observe in the laboratory. Providing our laboratory growth conditions with a red/far-red light ratio similar to open-field conditions and daily temperature oscillation is sufficient to mimic the FT expression and flowering time in natural long days. Under the adjusted growth conditions, key light signalling components, such as phytochrome A and EARLY FLOWERING 3, play important roles in morning FT expression. These conditions stabilize CONSTANS protein, a major FT activator, in the morning, which is probably a critical mechanism for photoperiodic flowering in nature. Refining the parameters of our standard growth conditions to more precisely mimic plant responses in nature can provide a powerful method for improving our understanding of seasonal response.

Project description:Although flowering in rice has been extensively investigated, few studies focused on genetic interactions. Flowering evaluation of two recombinant inbred line (RIL) populations involving photo-insensitive rice cultivars, Bengal and Cypress, and a weedy rice accession, PSRR-1, under natural long-day (LD) conditions, revealed six to ten quantitative trait loci (QTLs) and a major QTL interaction. In addition to the validation of several previously cloned genes using an introgression lines (IL) population of PSRR-1, a few novel QTLs were also discovered. Analysis of the marker profiles of the advanced backcross lines revealed that Hd1 allele of PSRR-1 was responsible for the photoperiodic response in the near-isogenic lines (NILs) developed in both cultivar backgrounds. Based on the phenotypic and genotypic data of the NILs, and NIL mapping population and the transcript abundance of key flowering pathway genes, we conclude that Hd1 and its interaction with a novel gene other than Ghd7 play an important role in controlling flowering under LD conditions. Our study demonstrates the important role of genetic interaction that regulates flowering time in rice and the need for further investigation to exploit it for breeding adaptable rice varieties.

<i>GhSWEET42</i> Regulates Flowering Time under Long-Day Conditions in <i>Arabidopsis thaliana</i>.

Project description:Flowering in plants is pivotal for initiating and advancing reproductive processes, impacting regional adaptation and crop yield. Despite numerous cloned and identified flowering time genes, research in cotton remains sparse. This study identified GhSWEET42 as a key determinant of the flowering time in cotton, demonstrating that its heterologous expression in Arabidopsis accelerated flowering under LD conditions compared to WT. Transgenic plants exhibited upregulated expression of the flowering inducers AtFT, AtSOC1, AtGI, and AtFKF1, alongside downregulated expression of the repressors AtTSF, AtFLC, and AtRGL2, correlating with the earlier flowering phenotype. GhSWEET42 showed a constitutive expression pattern, with elevated levels in the leaves, petals, and flower buds, and was notably higher in early-maturing cotton varieties. Subcellular localization assays confirmed GhSWEET42's presence on the cell membrane. Transcriptome analysis between WT and GhSWEET42-overexpressing Arabidopsis plants revealed 2393 differentially expressed genes (DEGs), spanning 221 biological processes, 93 molecular functions, and 37 cellular components according to Gene Ontology (GO) enrichment analysis. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis categorized the DEGs into metabolism and environmental information processing. These findings enhance the understanding of GhSWEET42's function and provide a foundation for elucidating the molecular mechanisms governing flowering time regulation in cotton.

SIN3 LIKE genes mediate long-day induction of flowering but inhibit the floral transition in short days through histone deacetylation in Arabidopsis

Project description:We show that in Arabidopsis SIN3 LIKE (SNL)family genes encoding a scoffold protein for assembly of histone deacetylase complex, directly regulate the expression of an FT activator and three FT repressors to regulate the transition to flowering in short days and long days, respectively. Under inductive long days, SNLs including SIN3 LIKE 1(SNL1) to SNL5, function in partial redundancy to repress the expression of three AP2 family transcription factors that repress FT expression, and thus mediate long-day induction of FT expression and promote the transitiion to flowering. In contrast, under non-inductive short days SNLs act to inhibit the floral transition, partly through direct repression of a MADS box transcriptional factor that promotes FT expression. Thus, our results reveal that SNLs, through histone deacetylation, play a novel dual role for the control of flowering in the long-day plant Arabidopsis: inhibiting flowering when the day length is shorter and promoting the floral transition when days become longer than a threshold length.

Two DELLA-interacting proteins bHLH48 and bHLH60 regulate flowering under long-day conditions in Arabidopsis thaliana.

Project description:Gibberellin (GA) regulates many developmental transitions in the plant life cycle. Although great progress has been made, the GA signaling pathways have not been fully elucidated. Identifying and characterizing new targets of DELLA proteins is an effective approach to reveal the complicated GA signaling networks. In this study, two novel DELLA-interacting transcription factors, bHLH48 and bHLH60, were identified. Their overexpression caused plants to flower early under long-day conditions, whereas their functional repression resulted in the opposite result. The constitutive expression of bHLH48 and bHLH60 upregulated the transcription of the FLOWERING LOCUS T (FT) gene. Chromatin immunoprecipitation experiments confirmed that bHLH48 bound to the promoter of FT and that GA promoted the DNA-binding activity of bHLH48. Genetic analyses indicated that the early flowering phenotype of plants overexpressing bHLH48 and bHLH60 depended on FT and that the overexpression of bHLH48 and bHLH60 could rescue the late-flowering phenotypes of RGL1 overexpressing plants. Transient expression assays suggested that RGL1 inhibited the transcription activation ability of bHLH48 and bHLH60. Taken together, this study confirmed that bHLH48 and bHLH60 positively regulate GA-mediated flowering.
