Project description:Karst rocky desertification (KRD) is a type of land deterioration, resulting in the degraded soil and a delicate ecosystem. Previous studies focused on the influence of KRD on the animals and plants, the impact of KRD on microorganisms, especially soil fungi remains to be discovered. This study reveals the change in the soil fungal community in response to KRD progression in southwest China. Illumina HiSeq was used to survey the soil fungal community. Results showed that the soil fungal community in the severe KRD (SKRD) was noticeably different from that in other KRD areas. Statistical analyses suggested that soil TN was the primary factor associated with the fungal community, followed by pH. Phylum Ascomycota was significantly abundant in non-degraded soils; whereas Basidiomycota predominated in SKRD. The ratio of Ascomycota/Basidiomycota significantly decreased along with KRD progression, which might be used as an indicator of KRD severity. Phylum Basidiomycota was sensitive to changes in all the soil properties but AP. Genus Sebacina might have the potential to promote vegetation and land restoration in KRD areas. This study fills a gap of knowledge on changes in soil fungal communities in accordance with KRD progression.

Project description:Vegetation restoration has become a common practice in karst rocky desertification (KRD) areas of southwestern China. The bacteria, which have made a connection between soil and plants, have been an important role in regulating the succession and restoration of karst vegetation. However, it is still unclear how soil bacterial communities and soil properties respond to natural vegetation restoration processes in karst areas. To address this gap, we investigated the soil nutrients, enzyme activity, and soil bacterial community among various plant communities, including farmland (FL), land with herbs only (SSI), herb-and-shrub land (SSII), woody thickets (SSIII), coniferous forest (SSIV), coniferous and broad-leaved mixed forest (SSV), and evergreen broad-leaved forest (SSVI). Our results found that SSII had the highest levels of soil organic matter, total nitrogen, available phosphorus, available nitrogen, sucrase, and β-glucosidase among all the plant communities. These results indicated that herb-and-shrub land have contributed to the rapid restoration of vegetation in KRD regions. FL exhibited the lowest levels of soil nutrients and enzyme activities, while showing the highest bacterial richness and diversity among all the plant communities. This suggested that appropriate human intervention can increase bacterial diversity and richness in the area. The predominant bacterial phylum also varied among the different plant communities, with Actinobacteria being the most abundant in SSI, SSII, SSIII, and SSIV, while Proteobacteria were the most abundant in SSV and SSVI. Furthermore, PCoA analysis demonstrated significant changes in the soil bacterial community structure, with SSI, SSII, SSIII, and SSIV had shared similar structures, while SSV and SSVI had comparable structures. As for soil characteristics, total phosphorus (TP) and total potassium (TK) were the primary factors affecting the soil bacterial community. SSV and SSVI had the most complex bacterial networks and were more stable than other groups. The genera Ktedonobacter, norank_f_Anaerolineaceae, and Vicinamibacter had the highest betweenness centrality scores and were identified as keystone genera in the co-occurrence network in KRD areas. In summary, our results have demonstrated that herb-and-shrub can promote community succession and increase soil nutrient levels in KRD regions.

Project description:Soil bacteria play an important role in regulating the process of vegetation restoration in karst ecosystems. However, the effects of vegetation restoration for different cultivated pastures on soil bacterial communities in the karst rocky desertification regions remain unclear. Therefore, we hypothesized that mixed pasture is the most effective for soil bacterial communities among different vegetation restorations. In this study, we systematically studied the soil properties and soil bacterial communities in four vegetation restoration modes [i.e., Dactylis glomerata pasture (DG), Lolium perenne pasture (LP), Lolium perenne + Trifolium repens mixed pasture (LT), and natural grassland (NG)] by using 16S rDNA Illumina sequencing, combined with six soil indicators and data models. We found that the vegetation restoration of cultivated pastures can improve the soil nutrient content compared with the natural grassland, especially LT treatment. LT treatment significantly increased the MBC content and Shannon index. The vegetation restoration of cultivated pastures significantly increased the relative abundance of Proteobacteria, but LT treatment significantly decreased the relative abundance of Acidobacteria. Soil pH and MBC significantly correlated with the alpha diversity of soil bacterial. Soil pH and SOC were the main factors that can affect the soil bacterial community. FAPROTAX analysis showed LT treatment significantly decreased the relative abundance of aerobic chemoheterotrophs. The results showed that the bacterial communities were highly beneficial to soil restoration in the LT treatment, and it confirmed our hypothesis. This finding provides a scientific reference for the restoration of degraded ecosystems in karst rocky desertification areas.

Project description:Microorganisms play critical roles in belowground ecosystems, and karst rocky desertification (KRD) control affects edaphic properties and vegetation coverage. However, the relationship between KRD control and soil bacterial communities remains unclear. 16S rRNA gene next-generation sequencing was used to investigate soil bacterial community structure, composition, diversity, and co-occurrence network from five ecological types in KRD control area. Moreover, soil physical-chemical properties and soil stoichiometry characteristics of carbon, nitrogen and phosphorus were analyzed. Soil N and P co-limitation decreased in the contribution of the promotion of KRD control on edaphic properties. Though soil bacterial communities appeared strongly associated with soil pH, soil calcium, soil phosphorus and plant richness, the key factor to determine their compositions was the latter via changed edaphic properties. The co-occurrence network analysis indicated that soil bacterial network complexity in natural ecosystem was higher than that in additional management ecosystem. Candidatus Udaeobacter, Chthoniobacterales, and Pedosphaeraceae were recognized as the key taxa maintaining karst soil ecosystems in KRD control area. Our results indicate that natural recovery is the suitable way for restoration and rehabilitation of degraded ecosystems, and thus contribute to the ongoing endeavor to appraise the interactions among soil-plant ecological networks.

Project description:Karst rocky desertification is one of the main causes of habitat fragmentation in Southwest China. Guizhou Province is located in the center of the karst area in southern China and is a typical karst ecological environment fragile area. Many studies have shown that habitat fragmentation is the main factor leading to the loss of biodiversity and species extinction, and it is also one of the important factors that threaten the survival of natural organisms. This study initially explored the habitat fragmentation degree, species diversity, and genetic diversity of leafhoppers in three typical karst areas in Guizhou. The study was combined with the general situation of the study area, understanding the main factors affecting habitat fragmentation, and putting forward reasonable protection suggestions for species resources. Based on satellite imagery, field survey collection, molecular sequencing data, and related index measurement methods, we measured the habitat fragmentation degree, species diversity index, and genetic diversity index of Erythroneurine leafhoppers of Shibing Yuntai Mountain Nature Reserve, Bijie Salaxi Demonstration Zone, Zhenfeng-Huajiang Demonstration Zone. Moreover, we compared the differences in the three study areas, carried out correlation analysis with relevant environmental factors, and discussed the main factors that formed the results. The results of the study show that the species diversity and genetic diversity of Erythroneurine leafhoppers in the study areas are affected by habitat fragmentation, and the weaker the degree of habitat fragmentation in the region, the higher the species diversity and genetic diversity, which is specifically manifested in species, quantity, and gene-flow. Understanding the status of biodiversity in karst areas is conducive to the sustainable development of biological resources. In order to better protect the diversity of such insects and their host plants and other biological diversity, combined with the background of the research area, we propose corresponding protection measures for reference.

Project description:Vegetation restoration has been widely used in karst rocky desertification (KRD) areas of southwestern China, but the response of microbial community to revegetation has not been well characterized. We investigated the diversity, structure, and co-occurrence patterns of bacterial communities in soils of five vegetation types (grassland, shrubbery, secondary forest, pure plantation and mixed plantation) in KRD area using high-throughput sequencing of the 16S rRNA gene. Bray-Curtis dissimilarity analysis revealed that 15 bacterial community samples were clustered into five groups that corresponded very well to the five vegetation types. Shannon diversity was positively correlated with pH and Ca2+ content but negatively correlated with organic carbon, total nitrogen, and soil moisture. Redundancy analysis indicated that soil pH, Ca2+ content, organic carbon, total nitrogen, and soil moisture jointly influenced bacterial community structure. Co-occurrence network analysis revealed non-random assembly patterns of bacterial composition in the soils. Bryobacter, GR-WP33-30, and Rhizomicrobium were identified as keystone genera in co-occurrence network. These results indicate that diverse soil physicochemical properties and potential interactions among taxa during vegetation restoration may jointly affect the bacterial community structure in KRD regions.

Project description:Rocky desertification is the most serious ecological disaster in karst areas. Comprehensive control of rocky desertification plays an important role in promoting the economic development of karst areas. Studying the stoichiometric characteristics of mosses and soil can provide a powerful reference for the ecological restoration and evaluation of ecosystems experiencing rocky desertification. Soil and mosses were collected from sites representing different stages of ecological restoration (bare rock, grassland, shrubland, and secondary forest), and the contents of carbon (C), nitrogen (N), and phosphorus (P) were detected for ecological stoichiometric analysis. The results indicate that in different restoration stages following karst rocky desertification, the contents of soil organic carbon (SOC), total nitrogen (TN), and total phosphorus (TP) and the stoichiometric ratios in the shrub habitat are higher than those in the bare rock, grassland, and secondary forest habitats. However, the TP and available P contents were low at all stages (0.06 g/kg and 0.62 mg/kg, respectively). The N and P contents and stoichiometric ratios in the mosses showed no significant differences among the succession stages. The C contents in the mosses had a significant positive correlation with SOC and TN and TP content, and the P content had a significant positive correlation with the soil available P. However, there was a significant negative correlation between the C: N and C:P ratios of the bryophytes and soil C: N. In summary, during the process of natural restoration of karst rocky desertification areas, SOC and soil TN contents accumulate with each succession stage. Soil nutrients are higher in shrub habitats than in other succession stages. Mosses have a strong effect on improving soil nutrients in rocky desertification areas.

Project description:In karst rocky desertification areas, bryophytes coexist with algae, bacteria, and fungi on exposed calcareous rocks to form a bryophyte crust, which plays an irreplaceable role in the restoration of karst degraded ecosystems. We investigated the biodiversity of crust bryophytes in karst rocky desertification areas from Guizhou Province, China. A total of 145 species in 22 families and 56 genera were identified. According to frequency and coverage, seven candidate dominant mosses were screened out, and five drought-resistant indexes of them were measured. Hypnum leptothallum, Racopilum cuspidigerum, and Hyophila involuta have high drought adaptability. We explored the interactions between two dominant mosses (H. leptothallum, H. involuta) and the structure of microbial communities in three karst rocky desertification types. Microbial diversity and function analysis showed that both moss species and karst rocky desertification types affect microbial communities. Moss species much more strongly affected the diversity and changed the community composition of these microbial groups. Bacteria were more sensitive in the microbiome as their communities changed strongly between mosses and drought resistance factors. Moreover, several species of fungi and bacteria could be significantly associated with three drought-resistant indexes: Pro (free proline content), SOD (superoxide dismutase activity), and POD (peroxidase activity), which were closely related to the drought adaptability of mosses. Our results enforced the potential role of moss-associated microbes that are important components involved in the related biological processes when bryophytes adapted to arid habitats, or as one kind of promoters in the distribution pattern of early mosses succession in karst rocky desertification areas.

Project description:The major phylogenetic pattern of the cyprinid tribe Labeonini has been revealed by previous molecular studies; however, the relationships within a clade that mainly inhabits the karst regions, which we refer to as the "karst group", in southwest China remain unresolved due to the low taxon sampling. This group includes more than 50% of the genera and species of Labeonini in China. Moreover, more than 90% of the genera of this group are endemic to China. In addition, some new genera and species of Labeonini have been discovered from these karst regions, but their taxonomic validity and phylogenetic position have not been examined. In this contribution, partial sequences of four nuclear (exon 3 of recombination activating protein 1, rhodopsin, early growth response protein 2B gene and interphotoreceptor retinoid binding protein gene) and three mitochondrial genes (cytochrome b, cytochrome oxidase subunit I and 16S ribosomal RNA) from 36 ingroup taxa and 25 outgroup taxa were analyzed to provide a hypothesis of the phylogenetic relationships within the labeonins of the karst regions in China. We propose that the monophyly of Parasinilabeo, Ptychidio, Rectoris and Semilabeo are supported. A new genus, Prolixicheilus, is erected for Pseudogyrinocheilus longisulcus. Cophecheilus bamen is the sister to Prolixicheilus longisulcus. Ptychidio, Pseudocrossocheilus, Semilabeo, Rectoris and Stenorynchoacrum are closely related with high support values. Sinocrossocheilus, Pseudogyrinocheilus, Paraqianlabeo, Hongshuia, Discogobio and Discocheilus form a clade together with high support. Considering molecular results and morphological differences, Parasinilabeo longicorpus and Ptychidio macrops might be the synonyms of Parasinilabeo assimilis and Ptychidio jordani respectively. Comprehensive taxonomic revisions of the two genera Parasinilabeo and Ptychidio may be necessary.

Project description:Phosphorus in the soil accessible to plants can easily be combined with calcium ion, the content of which is high in karst rocky desertification (KRD) regions, thereby resulting in a low utilization efficiency of phosphorus. The application of phosphate-solubilizing bacteria (PSB) from the KRD region would facilitate enhanced phosphate availability in the soil. In the present study, the strains belonging to Acinetobacter, Paraburkholderia, and Pseudomonas with efficient phosphate-solubilizing ability were isolated from fruit tree rhizosphere soils in KRD regions. Particularly, Acinetobacter sp. Ac-14 had a sustained and stable phosphate-solubilizing ability (439-448 mg/L, 48-120 h). Calcium carbonate decreased the phosphate-solubilizing ability in liquid medium; however, it did not affect the solubilization index in agar-solidified medium. When cocultivated with Arabidopsis thaliana seedling, Ac-14 increased the number of lateral roots, fresh weight, and chlorophyll content of the seedlings. Metabolomics analysis revealed that Ac-14 could produce 23 types of organic acids, majorly including gluconic acid and D-(-)-quinic acid. Expression of Ac-14 glucose dehydrogenase gene (gcd) conferred Pseudomonas sp. Ps-12 with a sustained and stable phosphate-solubilizing ability, suggesting that the production of gluconic acid is an important mechanism that confers phosphate solubilization in bacteria. Moreover, Ac-14 could also produce indole acetic acid and ammonia. Collectively, the isolated Ac-14 from KRD regions possess an efficient phosphate-solubilizing ability and plant growth-promoting effect which could be exploited for enhancing phosphorus availability in KRD regions. This study holds significance for the improvement of soil fertility and agricultural sustainable development in phosphorus-deficient KRD regions.
