Project description:Understanding of plant adaptation to abiotic stresses has implications in plant breeding, especially in the context of climate change. MicroRNAs (miRNAs) and short interfering RNAs play a crucial role in gene regulation. Here, wheat plants were exposed to one of the following stresses: continuous light, heat or ultraviolet radiations over five consecutive days and, leaf tissues from three biological replicates were harvested at 0, 1, 2, 3, 7 and 10 days after treatment (DAT). A total of 72 small RNA libraries were sequenced on the Illumina platform generating ~524 million reads corresponding to ~129 million distinct tags from which 232 conserved miRNAs were identified. The expression levels of 1, 2 and 79 miRNAs were affected by ultraviolet radiations, continuous light and heat, respectively. Approximately 55% of the differentially expressed miRNAs were downregulated at 0 and 1 DAT including miR398, miR528 and miR156 that control mRNAs involved in activation of signal transduction pathways and flowering. Other putative targets included histone variants and methyltransferases. These results suggest a temporal miRNA-guided post-transcriptional regulation that enables wheat to respond to abiotic stresses, particularly heat. Designing novel wheat breeding strategies such as regulatory gene-based marker assisted selection depends on accurate identification of stress induced miRNAs.

Project description:Plants regulate growth and development in different ambient environments through light signaling pathways and heat stress signaling pathways, both of which are very important for plant adaptation to the environment, but whether there is an interaction between the two is still unclear. In this article, we reported that the blue light receptor Cryptochrome CRY1 phosphorylation will be inhibited under high temperature stress, and CRY1 activity will decrease; at this time, CRY1 could not interact with E3 ubiquitin ligase COP1(CONSTITUTIVELY PHOTOMORPHOGENIC 1) and COP1 activity increased. Its substrate HY5(ELONGATED HYPOCOTYL 5）is continuously degraded under heat stress. Through further transcriptome analysis, we found that the substrates of HY5 have a class of HSFAs（HEAT SHOCK FACTOR）HSFA2/HSFA7As/HSFA7Bs transcription factors, which are rapidly induced under heat stress conditions and activate downstream HSPs (HEAT SHOCK PROTEIN)expression.Under normal conditions, HY5 protein inhibits HSFA2/HSFA7As/HSFA7Bs. Under heat stress, HY5 protein is degraded, which releases the expression of HSFAs, thereby enhancing plant heat tolerance. In this paper, we put forward the hypothesis that the phosphorylation of CRY1 seems to be a switch in the ambient environment , and through the regulation of phosphorylation level, the light and heat stress signal are transmitted downward, forming an light-heat signal interactive regulation pathway to regulate the growth and development of plants.

Project description:Plants transcriptome react to environment temperture changes profoundly. In Arabidopsis seedlings, genes response to temperature fluctuations to adopt the ever-changing ambient envrionment. We used microarrays to detail the global programme of gene expression underlying heat stress response progress in Arabidopsis. Ten-day-old Arabidopsis seedlings were selected for RNA extraction and hybridization on Affymetrix microarrays. We sought to explore the heat stress response in transcriptome, thus we treat the plants with heat stress. While in order to identify the interaction between light and temperature signaling pathways in plant , we treat Arabidopsis with heat stress under both light and dark conditions. To that end, our plant tissues are grouped as: HS-LIGHT, HS-DARK,CONTROL-LIGHT,CONTROL-DARK.

Project description:Considering global climate changes, incidences of combined drought and heat stress are likely to increase in the future and will considerably influence plant-pathogen interactions. Until now, little is known about plants exposed to simultaneously occurring abiotic and biotic stresses. To shed some light on molecular plant responses to multiple stress factors, a versatile multi-factorial test system, allowing simultaneous application of heat, drought and virus stress, was developed. Comparative analysis of single, double and triple stress responses by transcriptome and metabolome analysis revealed that gene expression under multi-factorial stress is not predictable from single stress treatments. Hierarchical cluster and principal component analysis identified heat as the major stress factor clearly separating heat-stressed from non-heat stressed plants. We identified 11 genes differentially regulated in all stress combinations as well as 23 genes specifically-regulated under triple stress. Furthermore, we showed that virus treated plants displayed enhanced expression of defense genes, which was abolished in plants additionally subjected to heat and drought stress. Triple stress also reduced expression of genes involved in the R-mediated disease response and increased the cytoplasmic protein response which was not seen under single stress conditions. These observations suggested that abiotic stress factors significantly altered TuMV-specific signaling networks which lead to a deactivation of defense responses and a higher susceptibility of plants. Collectively, our transcriptome and metabolome data provide a powerful resource to study plant responses during multi-factorial stress and allows identifying metabolic processes and functional networks involved in tripartite interactions of plants with their environment.

Stress induced gene expression in Arabidopsis leaves was measured after exposure to single and combined abiotic and biotic stress. Plants were grown on soil for 21 days till virus infection. Eight days later controlled drought stress was applied. At the end of the treatments heat was applied for three days. Four biological replicates have been hybridized for each treatment. Furthermore, Arabidopsis plants were exposed to a single severe heat stress (37°C day/33°C night) to mimic the severity of the triple stress experiment.
