Project description:Transcriptional profiling of SOX11-expressing mouse epidermis (K14-rtTA;TRE-Sox11-FLAG) compared to control (K14-rtTA) epidermis at postnatal day 4 (P4). The littermate pairs were injected with Dox for 12 h before their epidermis was harvested. Goal was to identify the gene expression profile of postnatal epidermis changed by SOX11 induced expression.

Gene expression signatures in mouse epidermis with transglutaminase 1 deficiency

Project description:The gene expression profile of 19.5 dpc Tgm1–/– mouse epidermis was compared with that of wild-type mice in duplicate samples, which were analyzed using microarrays. The expression of 2,502 entities was increased more than 2-fold in Tgm1–/– epidermis vs wild-type epidermis, and 1,403 of those entities corresponded to protein-coding genes. Gene ontology (GO) analysis of those entities revealed that the top 15.5% of 90 GO terms with corrected P < 0.05 were related to defense or immune responses. The entities for 177 annotated transcripts showed more than a 5-fold increase in expression in Tgm1–/– epidermis vs wild-type epidermis and those were also subjected to GO analysis. Interestingly, 24 genes from those entities were sorted into the category of defense response (P<2.78E-11; corrected P<1.50E-07).

Project description:Analysis of the transcriptional response to aneuploidy in mouse epidermis. In this study we measured the transcriptional response to aneuploidy by aboragting the spindle checkpoint in mouse epidermis. We found that, whereas spindle checkpoint inactivation in the epidermis is tolerated, but results in metabolic deranged cells, SAC abrogation kills bulge stem cells Mad2; K14-Cre mice were sacrificed at indicated timepoints and epidermis was separated from dermis using overnight trypsin. RNA was isolated and expression patterns were compared between K14-Cre; Mad2f/f and Cfre negative animals for all timepoints.

Constitutive activation of Stat3 in mouse epidermis

Project description:Potential Stat3 target genes involved in hair follicle formation were examined using mouse whole-genome microarray analysis. Global gene expression patterns in the epidermis of transgenic mice with keratinocytes expressing a constitutively active Stat3BK5.Stat3C mice (Sano et al., 2005) were compared with the pattern in the epidermis of wild-type mice. Of approximately 130 differentially expressed genes detected by microarray, approximately 70 genes were significantly up-regulated in epidermis by constitutive activation of Stat3. Thirty three of these genes were hair follicle related genes. These genes included trichohyalin, hair keratins, and keratin-associated proteins that are expressed in the hair shaft and inner root sheath as essential structural components for hair growth (Winter et al., 1997). To confirm the microarray results, five genes whose expression was up-regulated in the epidermis of BK5.Stat3C mice were chosen for quantitative RT-PCR analysis. Consistent with the microarray data, the expression of Tchh, Krtap16-8, Krtap6-1, Krt33a, and Krt25d was significantly increased in the epidermis of BK5.Stat3C mice compared to wild-type mice. In addition, Krtap16-1, which was not identified by microarray analysis but is localized on mouse chromosome 16, was highly expressed in the epidermis of BK5.Stat3C mice in comparison to wild-type (Figure 1f).
