Project description:To reveal the possible molecular mechanism underpinning the male sterility induced by a sulfonyflurea herbicide tribenuron-methyl (TBM), a comparative transcriptome analysis between rapeseed plants exposed to a TBM solution of 0.2 mg/L concentration and the control (spray with water) was conducted by using Illumina digital gene expression tag profiling technology (DGE). A transcriptomic analysis of the flower buds of TBM treatment identified 208 up-regulated and 164 down-regulated differential expression genes in comparison with the control. It was showed by the genetic network that the main groups of GO terms including response to chemical and hormone stimulus (response to stress), developmental process (pollen wall and pollen exine formation), lipid metabolism, and cellular amino acid derivative biosynthesis process were significantly influenced by the TBM treatment. The cytological and transcriptomic alterations in the treated plants were in accordance with reduction of chlorophyll, photosynthetic rate, and elevation of ethylene release.

Project description:To reveal the possible molecular mechanism underpinning the male sterility induced by a sulfonyflurea herbicide amidosulfuron, a comparative transcriptome analysis between rapeseed under amidosulfuron folia spray and the contrast was conducted by using Illumina digital gene expression tag profiling technology (DGE). A total of 54 up-regulated and 119 down-regulated DETs (differentially expressed transcripts) were annotated by 357 GOs and 32 KEGG pathways, indicating that the reaction to amidosulfuron exposure is complicated in rapeseed. Some interesting GOs included (1) amino acid metabolite, (2) energy metabolite, (3) carbohydrate, lipid, protein metabolite, (4) cell division, growth and death, (5) stimulus and defense response, (6) chlorophyll and mitochondria, (7) hormone, (8) oxidoreductase, and (9) flavonoid biosynthesis. Expression of most genes in cell division, growth and death category, and flavonoid biosynthesis were down-regulated. Ethylene mediated signaling pathway was up-regulated, while auxin mediated signaling pathway was down-regulated. Stimulus and defense response were induced.

When the air temperature reaches to above the optimal range (higher than 30/26˚C; day/night), only a small number of flower buds would develop into mature flowers and produce a reduced number of pollen. This project used the iodoTMT proteomics analysis method to identify heat-induced proteomes in these tomato flower buds.

Project description:Papaya (Carica papaya) is a trioecious species, with female, male, and hermaphrodite plants. Because of sex segregation, selecting hermaphroditic plants is vital for orchard establishment due to their higher commercial value. In addition to the costly sexing step, environmental stresses can result in abnormal flower development. However, molecular mechanisms that regulate sex differentiation in papaya are still poorly known. Thus, this study aimed to identify proteins associated with sex development in female and hermaphrodite flowers of papaya through comparative proteomic analysis. Proteins from flower buds at the early and late developmental stages of three papaya genotypes (UENF-CALIMAN 01, JS12, and Sunrise Solo 72/12) were studied via proteomic analysis via the combination of the shotgun method and nanoESI-HDMSE technology. In buds at an early stage of development, 496 proteins exhibited significantly different abundances between sexes for the SS72/12 genotype, 139 for the JS12 genotype, and 165 for the UC-01 genotype. At the final stage of development, there were 181 for SS72/12, 113 for JS12, and 125 for UC-01. The large group of differentially accumulated proteins (DAPs) between the sexes was related to metabolism, as shown by the observation of only the proteins that exhibited the same pattern of accumulation in the three genotypes. Specifically, carbohydrate metabolism proteins were up-regulated in hermaphrodite flower buds early in development, while those linked to monosaccharide and amino acid metabolism increased during late development. Enrichment of sporopollenin and phenylpropanoid biosynthesis pathways characterizes hermaphrodite samples across developmental stages, with predicted protein interactions highlighting the crucial role of phenylpropanoids in sporopollenin biosynthesis for pollen wall formation. Most of the DAPs played key roles in pectin, cellulose, and lignin synthesis and were essential for cell wall formation and male flower structure development, notably in the pollen coat. These findings suggest that hermaphrodite flowers require more energy for development, likely due to complex pollen wall formation. Overall, these insights illuminate the molecular mechanisms of papaya floral development, revealing complex regulatory networks and energetic demands in the formation of male reproductive structures.

Project description:Oil rapeseed (Brassica napus L.) is a typical winter biennial plant, with high cold tolerance during vegetative stage. In recent years, more and more early-maturing rapeseed varieties were planted across China. Unfortunately, the early-maturing rapeseed varieties with low cold tolerance have higher risk of freeze injury in cold winter and spring. Little is known about the molecular mechanisms for coping with different low-temperature stress conditions in rapeseed. In this study, we investigated 47,328 differentially expressed genes (DEGs) of two early-maturing rapeseed varieties with different cold tolerance treated with cold shock at chilling (4°C) and freezing (−4°C) temperatures, as well as chilling and freezing stress following cold acclimation or control conditions. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis indicated that two conserved (the primary metabolism and plant hormone signal transduction) and two novel (plant-pathogen interaction pathway and circadian rhythms pathway) signaling pathways were significantly enriched with differentially-expressed transcripts. Our results provided a foundation for understanding the low-temperature stress response mechanisms of rapeseed. We also propose new ideas and candidate genes for genetic improvement of rapeseed tolerance to cold stresses.

Project description:This study investigates the transcriptome and physiological responses of rapeseed to post-flowering temperature increases, providing valuable insights into the molecular mechanisms underlying rapeseed tolerance to heat stress. Two rapeseed genotypes, Lumen and Solar, were assessed under control and heat stress conditions in field experiments conducted in Valdivia, Chile. Results showed that seed yield and seed number were negatively affected by heat stress, with genotype-specific responses. Lumen exhibited a 9.3% average seed yield reduction, while Solar showed a 28.7% reduction. RNA-seq analysis of siliques and seeds revealed tissue-specific responses to heat stress, with siliques being more sensitive to temperature stress. Hierarchical clustering analysis identified distinct gene clusters reflecting different aspects of heat stress adaptation in siliques, with a role for protein folding in maintaining silique development and seed quality under high temperature conditions. In seeds, three distinct patterns of heat-responsive gene expression were observed, with genes involved in protein folding and response to heat showing genotype-specific expression. Gene coexpression network analysis revealed major modules for rapeseed yield and quality, as well as the trade-off between seed number and seed weight. Overall, this study contributes to understanding the molecular mechanisms underlying rapeseed tolerance to heat stress and can inform crop improvement strategies targeting yield optimization under changing environmental conditions.

Project description:BACKGROUND:Acetolactate synthase (ALS)-inhibiting herbicide tribenuron-methyl (TBM) is an efficient gametocide that can cause rapeseed (Brassica napus L.) to become male sterile and outcrossing. To find the reason the TBM treatment leads to male sterility, an integrated study using cytological, physiological, and transcriptomic methods was conducted. RESULTS:Some temporary symptoms, including the discoloration of young leaves and a short halt of raceme elongation, were observed in the rapeseed plants exposed to TBM at an application rate of 1??g per plant. Both chloroplasts in young leaves and plastids in anthers were deformed. TBM also reduced the leaf photosynthetic rate and the contents of chlorophyll, soluble sugar and pyruvate. Both the tapetal cells and uni-nucleate microspores in the treated plants showed large autophagic vacuoles, and the tissue degenerated quickly. A transcriptomic comparison with the control identified 200 upregulated and 163 downregulated differential expression genes in the small flower buds of the TBM treatment. The genes encoding functionally important proteins, including glucan endo-1,3-beta-glucosidase A6, QUARTET3 (QRT3), ARABIDOPSIS ANTHER 7 (ATA7), non-specific lipid-transfer protein LTP11 and LTP12, histone-lysine N-methyltransferase ATXR6, spermidine coumaroyl-CoA acyltransferase (SCT), and photosystem II reaction centre protein psbB, were downregulated by TBM exposure. Some important genes encoding autophagy-related protein ATG8a and metabolic detoxification related proteins, including DTX1, DTX6, DTX35, cytosolic sulfotransferase SOT12, and six members of glutathione S-transferase, were upregulated. In addition, several genes related to hormone stimulus, such as 1-aminocyclopropane-1-carboxylate synthase 8 (ACS8), ethylene-responsive factor ERF1A, ERF1, ERF71, CRF6, and RAP2-3, were also upregulated. The transcriptional regulation is in accordance with the functional abnormalities of pollen wall formation, lipid metabolism, chloroplast structure, ethylene generation, cell cycle, and tissue autophagy. CONCLUSION:The results suggested that except for ALS, the metabolic pathways related to lipid metabolism, pollen exine formation, photosynthesis and hormone response are associated with male sterility induced by TBM. The results provide new insight into the molecular mechanisms of inducing male sterility by sulfonylurea.
