Project description:Hsa_circ_0084443 expression level is down-regulated during normal skin wound healing and higher level of hsa_circ_0084443 was found in chronic non-healing diabetic foot ulcers compared to normal wounds. However, the biological function of hsa_circ_0084443 in epidermal keratinocytes during wound repair has not been studied. To study the genes regulated by hsa_circ_0084443, we transfected siRNA targeting hsa_circ_0084443 diagnostic junction into human primary epidermal keratinocytes to knockdown hsa_circ_0084443 expression. We performed a global transcriptome analysis of keratinocytes upon knockdown of hsa_circ_0084443 using Affymetrix arrays.

Project description:Recent study has revealed that long non-coding RNAs (lncRNAs) perform as important regulators of cellular physiology and pathology, which makes them promising therapeutic and diagnostic entities. We found lncRNA WAKMAR1 is significantly down-regulated in wound-edge keratinocytes from venous ulcer and diabetic foot ulcer compared to the normal wounds. Interleukin-4 and interleukin-13 cytokines effect on epidermal keratinocytes

Project description:Analysis of cultured epidermal keratinocytes treated with interleukin-4 (IL-4) and interleukin-13 (IL-13). IL-4 and IL-13 are up-regulated in atopic dermatitis. Results provide insight into the role of IL-4 and IL-13 cytokines in the pathogenesis of atopic dermatitis. Analysis of epidermal keratinocytes transfected with dual oxidase 1 (DUOX1) siRNA knockdown before treatment with IL-4 and IL-13. DUOX1 is one of the NOX family members of NADPH oxidases whose primary function is ROS generation. Results provide insight into the role of the incraesed expression of DUOX1 in IL-4/IL-13-treated NHEK for IL4/IL13 signaling. IL-4 and IL-13 induced gene expression in human epidermal keratinocytes (NHEK) was measured at 48 hours. Gene expression in NHEK tranfected with 10 nM DUOX1 siRNA followed by treatment with 100 ng/ml IL-4 and 100 ng/ml IL-13 was measured at 48 hours. Three independent experiments were performed using different strains for each experiment.

Expression data from primary human keratinocytes upon control or HERC6 knockdown using siRNA and stimulated in vitro with or without cGAMP or Poly(I:C)

Project description:HERC6 is an Interferon stimulated gene and expressed by epidermal keratinocytes, yet its specific role in the skin remains unclear. In this investigation, we employed bulk RNA-sequencing to explore the broader impact of HERC6 in response to sensing of cytosolic DNA mimic, cGAMP or RNA agonist, TLR3. We observed an elevated number of differentially expressed genes in keratinocytes following HERC6 knockdown, and among them, several were associated with the Interferon signaling pathway, such as OAS1, IFIT3, and STAT2, showing increased expression in HERC6 knockdown keratinocytes when stimulated with cGAMP. Additionally, our findings revealed a dysregulation in the HIPPO pathway-related genes, particularly LATS1/2, in HERC6 knockdown keratinocytes. Utilizing bulk RNA-sequencing, we identified enriched Gene Ontology terms associated with the Interferon pathway and cytosolic sensing of foreign DNA in HERC6 knockdown keratinocytes, suggesting HERC6 as a negative regulator of Interferon signaling upon DNA sensing but not RNA. This data was instrumental in elucidating the regulatory functions of HERC6 in keratinocytes during the sensing of exogenous DNA or RNA mimics.

To study the genes regulated by cASH1L, we transfected siRNA targeting the diagnostic junctions of cASH1L into human primary epidermal keratinocytes to knockdown cASH1L expression. We performed a global transcriptome analysis of keratinocytes upon circRNA knockdown using Affymetrix arrays.
