Comparative Analysis of MiRNA Expression Profiles of Yak Milk-derived Exosomes at Different Altitudes

Project description:Yaks are a rare and unique animal species inhabiting the Qinghai-Tibet Plateau, known for their remarkable resilience. Milk-derived exosomes, tiny vesicles containing various biological molecules, play crucial roles in numerous pathological and physiological processes, including cell growth, development, and immune regulation. This study conducted small RNA sequencing to characterize the miRNA expression profiles of yak milk-derived exosomes from high and low altitudes, aiming to identify differentially expressed miRNAs. Our findings revealed that these differentially expressed miRNAs are involved in mammary gland inflammation, virus infection regulation, and heat stress response. Through GO and KEGG enrichment analyses, we discovered that the target genes of these differentially expressed miRNAs are enriched in signaling pathways related to Th17 cell differentiation and the Ras-MAPK signaling pathway. In conclusion, this research sheds light on the differential expression of miRNAs in yak milk-derived exosomes under varying environmental conditions. This new knowledge provides a theoretical foundation for further exploring yak resilience and the potential role of milk-derived exosomes in disease and immune modulation.

CnbHLH130 positively regulates salt tolerance through CnCBL10-CnCIPK4 mediated SOS pathway in Campeiostachys nutans

Project description:Campeiostachys nutans, a dominant perennial grass in the Qinghai-Tibet Plateau, exhibits high tolerance to salt stress. The Salt Overly Sensitive (SOS) pathway is key to plant salt stress tolerance. However, the pivotal role of the SOS pathway in response to salt stress in C. nutans remains unknown. Here, we identified CnbHLH130 as a novel transcriptional activator of CnCBL10, directly binds to the G-box motif in the promoter. CnbHLH130 responds to salt stress and positively regulate salt tolerance in rice and C. nutans. Interestingly, we found CnCBL10 and CnCIPK4 interact with CnbHLH130 by a Y2H screening assay. The interactions were confirmed by split-luciferase complementation (split-LUC), Pull-down, Co-immunoprecipitation (Co-IP) and bimolecular fluorescence complementation (BiFC) assays. Moreover, CnbHLH130 enhanced the interaction between CnCBL10 and CnCIPK4, which further phosphorylate and activate Na+/H+ antiporter CnSOS1 to exclude excess cytosolic Na+ from cells in the shoots. Genetic evidence showed that CnCBL10, CnCIPK4 and CnbHLH130 coordinately regulates salt tolerance in plants. In summary, this study demonstrated that CnbHLH130 acts as a novel core component and transcriptional activator regulating CnCBL10-CnCIPK4 mediated SOS pathway, thus conferring to the salt tolerance in C. nutans. This work advanced our understandings of how an alpine plant greatly survived in the Qinghai-Tibet Plateau by concise regulation of the SOS pathway in response to salt stress.

Dynamic changes in cellular atlases and communication patterns within yak ovaries across diverse reproductive states unveiled by single-cell analysis [scRNAseq_Yak_Ovary_Pregnancy_2]

Project description:Yaks (Bos grunniens) exhibit exceptional adaptation to the challenging high-altitude environment of the Qinghai-Tibetan plateau, making them the sole bovine species capable of thriving in such exreme conditions. Investigating the cellular and molecular characteristics of yak ovaries across different reproductive states is crucial for gaining insight into their ovarian functions. Herein, the cellular atlases of yak ovaries in different reproductive states were depicted by single-cell RNA-sequencing (scRNA-seq). The cellular atlases of the ovaries were established by identifying specific gene expression patterns of various cell types, including granulosa cells, theca cells, stromal cells, smooth muscle cells, endothelial cells, glial cell, macrophages, natural killer cells, and proliferating cells. The cellular compositions of the ovaries vary among different reproductive states. Moreover, a cell-to-cell communication network was constructed among distinct cell types within the ovary, spanning the three reproductive states.
