Project description:In previous papers we proved that cancer nodules and surrounding normal tissue are governed by different gene master regulators (GMRs) and that expression manipulation of a cancer GMR can selectively destroy cancer cells with minimal effects on the normal ones. GMR is defined as a coding or non-coding transcript whose strictly controled abundance by the cell homeostatic mechanisms regulates most major cell functional pathways. Here, we determined the gene hierarchy in the standard adenocarcinomic human alveolar basal epithelial cell line A549.

Project description:Several reports indicated that miR155 is upregulated in many cancer forms, including lung adenocarcinoma. Studies perfomed in Calin's lab (e.g. Van Roosbroeck et al., Clin Cancer Res, doi: 10.1158/1078-0432.CCR-16-1025) demonstrated that miR-155 induces resistance to multiple chemotherapeutic agents. We hypothesize that the induced resistance is related to the rearrangement of cellular gene hierarchy. In previous papers (Iacobas & Iacobas, Cancer & Oncol Res DOI:10.13189/cor.2017.050301; Iacobas et al., Oncotarget doi: 10.18632/oncotarget.23417) we proved that cancer nodules and surrounding normal tissue are governed by different gene master regulators (GMRs) and that expression manipulation of a cancer GMR can selectively destroy cancer cells with minimal effects on the normal ones. Project description:To gain deeper insight into the mechanism of toxicity, it is important to identify and characterize mRNAs profiles involved in responses to specific classes of toxicants in conjunction with their impact on gene expression levels. However, few reports have described the effects of toxicants on mRNA expression profiles. Taking into account the prominent role of mRNAs in cancer development, progression, cell cycle control, and proliferation-related processes, it is likely that mRNAs are involved in the toxic response induced by carcinogens. Aldehydes are a well-characterized class of human carcinogens. In the present study, we documented the different expression profiles of mRNAs in environmental carcinogen-exposed A549 cells by mRNA microarray analysis. To evaluate the change in mRNA expression levels, human alveolar cells(A549) were exposed to seven lower-molecular-weight saturated aliphatic aldehyde (LSAAs) for 48h. mRNA expression analysis was conducted using a 4x44k human whole genome oligo microarray (Agilent Technologies, USA).

Transcriptional Profile of Mycobacterium tuberculosis Replicating in Human Type II Alveolar Epithelial A549 cell line

Project description:Infection with Mycobacterium tuberculosis (M. tb) is initiated when an aerosol droplet carrying a few bacilli is inhaled into an alveolus. Alveolar epithelial cells (AEC) (type II and type I) are among the first cells encountered by the infecting bacteria and greatly outnumber macrophages in the alveolus. M. tb replicates dramatically (>20,000 fold) in a “non-migrating” compartment in the lung prior to the development of the cell-mediated immune response in aerosol-infected mice (Wolf AJ, 2008). M. tb DNA has been found in type II AEC in autopsied lung tissue of latently infected individuals (Hernandez-Pando R et. al, 2000; Barrios-Payan J et. al 2012), and M. tb-infected AEC in broncho-alveolar lavage (BAL) and in sputum samples from TB patients indicates the infection of these cells in active as well as latent human TB (Eum SY et. al, 2010). M. tb has been shown to infect and replicate in the human type II AEC line, A549, and passaging of M. tb through A549 increases M. tb invasiveness (Bermudez LE et.al, 2002). In this work, we have used DNA microarray analysis to investigate the transcriptome of M. tb replicating in type II AEC (A549) compared to M. tb grown logarithmically in Middlebrook 7H9 broth media in order to identify M. tb adaptations to this intracellular environment as well as M. tb mechanisms/factors contributing to M. tb replication and increased invasiveness in primary infection. The global gene expression of M. tb H37Rv replicating in A549 cells at 72 hr was compared to that of M. tb grown to log phase in Middlebrook 7H9 media.

Long Term Culture of the A549 Cell Line Promotes Differentiation Towards an Alveolar Type II (ATII) Phenotype; A549 Time course.

Project description:Pulmonary research requires models that represent the physiology of alveolar epithelium but concerns with reproducibility, consistency and the technical and ethical challenges of using primary or stem cells has resulted in widespread use of continuous cancer or other immortalized cell lines. The A549 ‘alveolar’ cell line has been available for over four decades but there is an inconsistent view as to its suitability as an appropriate model for primary alveolar type II (ATII) cells. Since most work with A549 cells involves short term culture of proliferating cells, we postulated that culture conditions that reduced proliferation of the cancer cells would promote a more differentiated ATII cell phenotype. We examined A549 cell growth in different media over long term culture and then used microarray analysis to investigate temporal regulation of pathways involved in cell cycle and ATII differentiation; we also made comparisons with gene expression in freshly isolated human ATII cells. Analyses indicated that long term culture in Ham’s F12 resulted in substantial modulation of cell cycle genes to result in a quiescent population of cells with significant up-regulation of autophagic, differentiation and lipidogenic pathways. There were also increased numbers of up- and down-regulated genes shared with primary cells suggesting adoption of ATII characteristics and multilamellar body (MLB) development. Subsequent Oil Red-O staining and Transmission Electron Microscopy confirmed MLB expression in the differentiated A549 cells. This work defines a set of conditions for promoting ATII differentiation characteristics in A549 cells that may be advantageous for studies with this cell line.
