Project description:Transcriptome analysis of monocytes directly exposed to cell-to-cell contact with Natural Killer (NK) cells or separated by a transwell membrane and their subsequent monocyte-derived dendritic cells. The role of Natural Killer (NK) cells in the early differentiation of monocytes into dendritic cells (DCs) is poorly understood. Their interaction is thought to be restricted to a bilateral cross talk of soluble cytokines. However, many authors have shown that NK cells effect over myeloid cells is dependent on direct cell-to-cell contact. In order to understand what determines a major effect of NK cells over monocytes and their derived DCs, total RNA samples from purified monocytes and monocyte-derived DCs, exposed to direct contact with NK cells or separated by a transwell membrane were amplified and hybridized to the Affymetrix Human GeneChip 2.0 ST array. Analyses were performed using BRB-ArrayTools version 4.5.0, developed by Dr. Richard Simon and the BRB-ArrayTools Development Team. We found that compared to untouched monocytes, cell-to-cell contact with NK cells modified the expression of 283 genes whereas contact through a transwell membrane modified the expression of 35 genes. After differentiation and compared to DCs derived from untouched monocytes, DCs derived from monocytes allowed to direct cell-to-cell contact with NK cells had the expression of 128 genes modified whereas DCs derived from monocytes primed through a transwell membrane had only 2 genes modified. Our results suggested that cell-to-cell interactions with NK cells are important to imprint a stable transcriptional program in monocytes lasting even after their further differentiation into DCs.

Natural killer cells trigger differentiation of monocytes into dendritic cells.

Project description:Circulating monocytes can differentiate into dendritic cells (moDCs), which are potent inducers of adaptive immune responses. Previous reports show that granulocyte macrophage-colony-stimulating factor (GM-CSF) and interleukin-4 induce monocyte differentiation into moDCs in vitro, but little is known about the physiological requirements that initiate moDC differentiation in vivo. Here we show that a unique natural killer (NK) cell subset (CD3(-)CD56(bright)) that accumulates in lymph nodes and chronically inflamed tissues triggers CD14(+) monocytes to differentiate into potent T-helper-1 (T(H)1) promoting DC. This process requires direct contact of monocytes with NK cells and is mediated by GM-CSF and CD154 derived from NK cells. It is noteworthy that synovial fluid (SF) from patients with rheumatoid arthritis (RA) and psoriatic arthritis (PsA), but not osteoarthritis (OA), induces monocytes to differentiate into DC. However, this process occurs only in the presence of NK cells. We propose that NK cells play a role in the maintenance of T(H)1-mediated inflammatory diseases such as RA by providing a local milieu for monocytes to differentiate into DC.

Project description:Blood genomic profiling has been applied to disorders of the blood and various organ systems including brain to elucidate disease mechanisms and identify surrogate disease markers. Since most studies have not examined specific cell types, we performed a preliminary genomic survey of major blood cell types from normal individuals using microarrays. CD4+ T cells, CD8+ T cells, CD19+ B cells, CD56+ natural killer cells, and CD14+ monocytes were negatively selected using the RosetteSep antibody cocktail, while polymorphonuclear leukocytes were separated with density gradient media. Genes differentially expressed by each cell type were identified. To demonstrate the potential use of such cell subtype-specific genomic expression data, a number of the major genes previously reported to be regulated in ischemic stroke, migraine, and Tourette syndrome are shown to be associated with distinct cell populations in blood. These specific gene expression, cell-type-related profiles will need to be confirmed in larger data sets and could be used to study these and many other neurological diseases. This study includes data from major blood cell types from normal individuals. These cell types include CD4+ T cells, CD8+ T cells, CD19+ B cells, CD56+ natural killer cells, CD14+ monocytes and PMN (polymorphonuclear cells). The data were hybridized on 3 separate batches of gene chips.
