Project description:Resistance to pyrethroids, the only insecticide approved for bednets, threatens control of the major malaria vector, Anopheles funestus, in Malawi. To improve the management of such resistance countrywide, it is crucial to understand the dynamics and mechanisms driving resistance in the field. In this study the levels of insecticide resistance were determined across the highly endemic densely populated lake and southern agricultural area. Insecticide resistance to pyrethoids was assessed using standardized WHO bioassay methods and resistant mosquitoes were hybridized to susceptible mosquitoes. This microarray analysis revealed the key role of cytochrome P450 genes such as CYP6P9a, CYP6P9b and CYP6M7. However, a significant shift in the over-expression of these CYP450s was detected across a south/north transect, with CYP6M7 more highly over-transcribed in the two northern collection sites and the tandemly duplicated genes, CYP6P9a and CYP6P9b, more greatly over-transcribed in the south.

Project description:Transcription profiles of a field collection of Anopheles gambiae M molecular form (i.e. Anopheles coluzzii) from a population from Cote d'Ivoire (West Africa) were compared to two laboratory strains and to a wild caught field strain (all carbamate insecticide susceptible) to investigate their differences in insecticide resistance. Both carbamate-exposed and carbamate-unexposed Cote d'Ivoire samples were compared to each susceptible strain, and also directly to one another (i.e. exposed vs. unexposed).

Project description:Malaria control relies on insecticides targeting the mosquito vector, but is being increasingly compromised by insecticide resistance. Elevated expression of metabolic enzymes frequently drives resistance. In diploids, gene expression is regulated both in cis, by regulatory sequences on the same chromosome, and by trans acting factors, affecting both alleles equally. Differing levels of transcription can be caused by mutations in cis-regulatory modules, but few cis-regulatory modules controlling the expression of genes that determine insecticide resistance have been identified. Genes potentially under differential cis-regulation between bendiocarb resistant and susceptible Anopheles gambiae strains were identified by counting transcripts produced from maternal and paternal alleles in F1 hybrids of these strains (allelic specific expression). Cis regulatory module sequences controlling gene expression in insecticide resistance relevant tissues such as midgut, Malpighian tubules and legs were predicted using a previously established machine learning method. These predictions included CRM proximal to both genes under differential cis regulation and genes that show consistent differential expression patterns in multiple resistant Anopheles strains.

Project description:Anopheles gambiae mosquitoes play an important role in malaria transmission. In sub-Saharan Africa, the dry season can last several months. The mechanisms for mosquito population to survive through the dry season are poorly understood. One possible mechanism is that adults increase their desiccation tolerance over the dry season. Genetic analyses have shown that inversions 2La, 2Rb, 2Rc, 2Rd and 2Ru are associated with aridity resistance, however little is known about the transcriptional response of genes in response to desiccation. The results of the present study demonstrate that desiccation affects expression of genes associated with several mosquito physiological mechanisms, including those that protect against water loss, but all structural related genes decreased their expression. The identified differentially expressed genes in response to desiccation stress can lay a foundation for better understanding of molecular mechanisms underling dry-season survival of An. gambiae mosquitoes, so it may provide a different option for malaria vector control. Transcriptional profiles of Anopheles gambiae female mosquitoes were exposed to 70% (standard) or 30% (desiccated) RH without any access to sugar and water at 0hr, 18hr or 36hr.

Project description:Identification of genes associated with bendiocarb resistance. Mosquitoes collected as larvae from Nagongera and Kihihi, Uganda. Bendiocarb-resistant and unexposed female mosquitoes selected using standard WHO tube bioassays. RNA was extracted from pools of five individuals identified as An. gambiae s.s. Insecticide-susceptible mosquitoes from the Kisumu strain were included as controls. RNA hybridized in an interwoven loop design to compare four biological replicates each of resistant, unexposed, and laboratory mosquitoes.

Project description:The heavy use of pesticides in intensive agricultural areas often leads to the contamination of neighbouring mosquito larvae breeding sites. Such exposure to complex agrochemical mixtures can affect the tolerance of mosquitoes larvae to insecticides. The objective of this study was to determine whether agrochemical pollutants found in Anopheline larval breeding sites can affect the tolerance of adults to Fludora® Fusion, a novel vector control insecticide formulation combining two insecticides with distinct modes of action. An. gambiae larvae were exposed to a sub-lethal dose of a mixture of agrochemical pesticides representative of a region with an intense agricultural activity in Ivory Coast. Comparative bioassays with Fludora® Fusion and its two insecticide component (deltamethrine and clothianidin) were carried out between adult mosquitoes exposed or not to the agrochemicals at the larval stage. A transcriptomic analysis through RNA sequencing was subsequently performed in larvae and adults in order to highlight the molecular mechanisms underlying the phenotypic changes observed. Bioessais revealed a significant increased tolerance of adult females to clothianidin (2.5-fold) and Fludora® Fusion (2.2-fold) following larval exposure to agrochemicals. No significant increased tolerance to deltamethrin was observed suggesting that pesticide exposure affects the efficacy of the Fludora® Fusion mixture mainly through mechanisms acting on clothianidin tolerance. Transcriptomic analysis revealed the potential of agrochemicals to induce various resistance mechanisms including cuticle proteins, detoxifications activities and altered insecticide sequestration. These results suggest that though Fludora® Fusion has a good potential for vector control, its efficacy may be locally affected by the ecological context. The present study also suggests that, although the complex interactions between the use of agrochemicals and vector control insecticides are difficult to decipher in the field, they still must be considered in the frame of insecticide resistance management programmes.
