Project description:Sulphur dioxide (SO2) and its derivatives are widely used as preservatives in food and beverage industries to assure the maintenance of microbiological quality of their products. In this work it is shown, for the first time, the essential role played by the transcriptional regulator Com (ORF YER130c) in S. cerevisiae response and tolerance to SO2, this being the first biological function attributed for this regulator.

Sulphur dioxide evokes large scale reprogramming of the grape berry transcriptome

Project description:Sulphur is used as a food preservative, especially throughout the table grape and wine industries, however there is increasing concern as to the health rises associated with human consumption. Thus, we investigated the transcript abundance changes in grapes treated with sulfur dioxide and other preservative compounds, compared to control treatments. Export quality, red-skinned M-^QCrimson SeedlessM-^R grapes (Vitis vinifera L.) were harvested at commercial maturity from one vineyard in the Swan Valley region of Western Australia. At least 15 kg grapes per treatment were completely immersed in the treatment solution (? 1 min) and allowed to dry on racks before being weighed in to 7 x 2 kg lined export cartons, without sulphur pads. Once packed, cartons were immediately placed in 2 M-0C storage for up to 56 days and once cool, commercial SO2-generating pads were placed into cartons for SO2 treatment. The salicylic acid (SA, 25 mM), methyl jasmonate (MJ, 5mM) and their combination (25 mM SA + 5 mM MJ) were dissolved in 0.5 % (v/v) dimethyl sulphoxide (DMSO). A 0.5 % solution of DMSO was used as a control treatment. An additional, untreated control for sulphur-treated berries was used. Microarrays were performed in duplicate for the 6 treatments, Sulfur dioxide (plus untreated control), SA, MJ, and the combined SAMJ treatment (plus DMSO control) on grape berries after 21 days of treatment post harvest. The results indcate a large scale re-programing of the grape berry transcritpome, similar to that which has been observed for prolonged exposure to harsh oxidative stress conditions.

MSN2-mCherry vs. msn2 microarray

Project description:Transcriptional profiling of Msn2 regulated genes. Both strains are PKAas (TPK1M164G TPK2M147G TPK3M165G) and msn4-delta. One strain has Msn2 C-terminally tagged with mCherry and the other strain is msn2-delta. In this experiment, both strains were exposed to 3 uM 1-NM-PP1, which leads to Msn2 nuclear localization, for 0, 10, 20 and 40 min. The goal was to determine the genes that show a strong upregulation in the presence of Msn2, but no expression change in the absence of Msn2.

Msn2-mediated nucleosome remodeling dynamics

Project description:The aim of this experiment was to determine, using MNAse-Seq, how nucleosomes get remodeled during an Msn2 activation timecourse genome-wide. Diploid strain EY2807/ASH79 with genotype TPK1M164G TPK2M147G TPK3M165G msn4::TRP1/LEU2 MSN2-mCherry NHP6a-iRFP::kanMX was used. This strain is PKAas, so upon addition of the inhibitor 1-NM-PP1, Msn2 translocates to the nucleus and activates gene expression. In this experiment, the diploid strain was exposed to 3 uM 1-NM-PP1 for 0, 5, 10, 20 and 40 min and nucleosome positions determined using by crosslinking, MNAse treatment, nucleosomal DNA purification and paired-end high-throughput sequencing (Illumina). Results from transcriptional profiling of yeast with or without Msn2 expression were also deposited at ArrayExpress under accession number E-MTAB-1945 ( https://www.ebi.ac.uk/arrayexpress/experiments/E-MTAB-1945/ ).
