Project description:We characterised tumour macrophages and tumour microglia using flow cytometry. We subsequently isolated tumour macrophages and tumour microglia, using flow-cell sorting in a mouse glioblastoma model and performed RNA-sequencing to identify: unique cell surface markers, upregulation of pro or anti-inflammatory genes and important functional pathways to overall determine the functional roles of tumour macrophages and tumour microglia.

Project description:Both established glioma cells lines U87MG and U373 were used for studying their interactions in the indirect co-cultures. Eventhough both being derived from the glioma tissue, those two cell lines prove morphologically and physiologically disctinct. Therefore their intre-cellular interactions were examined on gene exprexssion level in vitro to observe whether those co-cultures could make for a suitable in vitro cell model mimicking the in vivo glioma tumour heterogeneity.

Glioblastoma intra-tumour transcriptomics

Project description:Gene expression microarray profiling on glioblastoma intra-tumour regions, where the study hypothesis states that the infiltrative tumour margin harbours a distinct transcriptomic profile from all non-infiltrative tumour regions. Data is from three patients (patient 9, 14 and 15) where regions 1-4 per patient were obtained from non-infiltrative intra-tumour regions, and region 5 was obtained from the infiltrative margin. All patients underwent craniotomy with intra-operative image guidance and visualization of 5ALA induced fluorescence to obtain infiltrative margin biopsies.

Proteomic profiling of glioblastoma tumour tissue

Project description:Using HiRIEF LC-MS/MS, we analysed 10 GBM tumour tissue samples ran in one TMT10 set. The set consisted of 7 primary tumours and 3 non-matched recurrent tumours. One primary tumour was highly necrotic.
