Project description:In social insects, workers perform distinct tasks according to the caste they belong to, and workers from different castes differ in their age (nest workers are usually younger than foragers are). The caste shift thus seems inseparable from age, preventing from deciphering the role of labour division and age in regulating individual physiology and ageing rates. We set up an experimental protocol separating age and caste effects by defining four groups of black garden ant (Lasius niger) workers: young foragers (Y.F), old foragers (O.F), young nest workers (Y.NW) and old nest workers (O.NW). Proteomics highlighted differences between individuals according to their age, whereas metabolomics revealed caste-related differences. Our study highlighted that age and caste influence specifically different aspects of the physiology of ant workers.

Project description:Animal longevity widely differs across species, and even individuals from the same species may exhibit different rates of ageing. In different species, the rate at which individuals actually age is related to the level of their social interactions, but this was still not known in ants. In a given colony, ant individuals are close genetic relatives, exhibit very different behaviours and a contrasted lifespan according to the Caste. Such characteristics constitute main advantages to study relationships between sociality and ageing. Therefore, the aim of this study was to characterize differences in the proteome of Lasius niger queens versus that of domestic and foraging workers. Proteomic data were put in relation with the behaviour of individuals from the three Castes. Hence, it was found that sociality correlates with ant longevity, with i) social immunity enabling the queen to mainly invest in soma protection, and ii) marked exposition of workers to the environment and nutrients inducing metabolic pathways that reduce their lifespan.

Project description:Eusocial insects have evolved the capacity to generate adults with distinct morphological, reproductive and behavioural phenotypes from the same genome. Recent studies suggest that RNA editing might enhance the diversity of gene products at the post-transcriptional level, particularly to induce functional changes in the nervous system. Using head samples from the leaf-cutting ant Acromyrmex echinatior, we compare RNA editomes across eusocial castes, identifying ca. 11,000 RNA editing sites in gynes, large workers and small workers. Those editing sites map to 800 genes functionally enriched for neurotransmission, circadian rhythm, temperature response, RNA splicing and carboxylic acid biosynthesis. Most A. echinatior editing sites are species specific, but 8M-bM-^@M-^S23% are conserved across ant subfamilies and likely to have been important for the evolution of eusociality in ants. The level of editing varies for the same site between castes, suggesting that RNA editing might be a general mechanism that shapes caste behaviour in ants. Project description:We used whole-genome fire ant microarrays to examine the molecular basis for division of labor in fire ant workers by comparing foraging and non-foraging workers from monogyne colonies. Fire ant colonies were collected in the field and transported into the lab were they were reared in standard conditions. We created a nesting chamber containing the queen, the brood and workers performing nursing tasks and a foraging area, separated from the nesting chamber and provided with food and water sources. Foraging workers were collected in the foraging area while non-foraging workers were collected in the nesting chamber. Total RNA was isolated from pools of whole workers and processed for microarrays.

Project description:Drone honey bees are the obligate sexual partners of queens, and the availability of healthy, high-quality drones directly affects the queen's fertility and the productivity of her subsequent colony. Yet, our understanding of how stressors affect drone fertility and physiology is presently limited. Like other male Hymenopterans, drones are haploid and are thus expected to be more sensitive to stressors than workers, as suggested by the haploid susceptibility hypothesis. We used quantitative proteomics to investigate protein expression profiles in the hemolymph of topically exposed workers and drones, and we show that drones express surprisingly high levels of putative stress response proteins - even in the negative control groups - relative to workers. These findings suggest that drones may invest in strong constitutive expression of damage-mitigating proteins for a wide range of stressors, potentially to the detriment of launching a more specific response to new stressors. Regardless of the underlying mechanism, the robust expression patterns of proteins involved in stress responses in drones suggests that drone stress tolerance systems are fundamentally rewired relative to workers, and their susceptibility to stress depends on more than simply gene dose or deleterious recessive alleles.

Project description:Senescence is manifested by an increase in oxidative stress and a decline in biological functions with age. In most organisms, body maintenance is traded-off with reproduction. The negative relationship between longevity and fecundity is also evident on the molecular level, yet the proximate mechanisms remain poorly understood. Apparently by-passing this trade-off, social insect queens are both extremely long-lived (up to 30 years in some ants) and highly fecund compared to workers. Here, we study changes in gene expression with age and fecundity in queens to determine how the trade-off between those two traits is reshaped. We analyse tissue-specific gene expression in young founding queens and old, highly fecund queens of the ant Temnothorax rugatulus. More genes altered their expression with age in the fat body than in the brain. Despite strong differences in ovary development, few fecundity genes were differentially expressed. However, many longevity genes involved in well-known pathways or lifespan-associated biological processes changed their expression with age indicating that multiple longevity mechanisms are activated successively throughout a queens’ life. Young queens invested in immunity (i.e. activation of the Toll signalling pathway) and resistance against environmental and physiological stress associated with the founding phase (i.e. down regulation of the TOR pathway), while established older queens up-regulate anti-aging mechanisms (i.e. up-regulation of catalase, superoxide dismutase, heat shock 70 kDa proteins). Finally, we identified a number of candidate genes and pathways, potentially involved in reshaping the fertility-longevity trade-off in social insects, shedding light on how this is achieved on a proximate level.
