Project description:Malaria infection induces complex and diverse immune responses, including impairment of dendritic cell (DC) function and immune suppression that may contribute to the low vaccination antibody titers to some antigens in endemic populations. To elucidate the mechanisms underlying host-parasite interaction, we performed a genetic screen during early Plasmodium yoelii infection and identified a large number of interacting host and parasite genes/loci after trans-species expression quantitative trait loci (Ts-eQTL) analysis. We next investigated a host E3 ubiquitin ligase gene (march1) that was clustered with interferon stimulated genes. March1 can inhibit MAVS/STING induced IFN-I signaling and reverse inhibition of viral replication mediated by MAVS in vitro. However, in malaria-infected hosts, deficiency of march1 activates IFN signaling inhibitors such as SOCS1, SOCS3, and TRIM24, leading to reduced early (24h) serum IFN-I levels. Increased CD86+ DC populations and elevated levels of IFN-? and IL-10 produced by T cells day 4 post infection protect infected march1-/- mice. Malaria lysate stimulate MACRH1 expression, which reduces CD86+ DC cells and impairs T cell activation. This study reveals previous unknown functions of MARCH1 in innate response to malaria infections and provides potential avenues for activating anti-malaria immunity and enhancing vaccine efficacy.

Project description:Malaria continues to pose a significant public health threat, with millions of cases and hundreds of thousands of deaths reported annually, primarily in sub-Saharan Africa. The disease disproportionally affects children under five years of age residing in holoendemic Plasmodium falciparum transmission regions, who account for 94% of the cases and 80% of the mortality. Young children are highly vulnerable to developing life-threatening severe malarial anemia [SMA, hemoglobin (Hb)<5.0 g/dL]. The overall goal of the project was to identify critical gene pathways within the transcriptome that mediate disease severity and then target these specific genes with compounds that elicit expression profiles witnessed in children with milder forms of disease. To achieve this goal, we are investing the following specific aims: 1) Determine how changing temporal dynamics of gene pathways in the Malarial Immunity Transcriptome promote SMA during acute disease; 2) Determine how changes in gene pathways in the Malarial Immunity Transcriptome mediate malarial severity throughout the development of naturally acquired immunity; and 3) Identify immunotherapeutic targets in the Malarial Immunity Transcriptome that can be used to reduce malaria disease severity and improve clinical outcomes in future trials. To successfully complete these aims, we are determining how host profiles impact on acute disease over 14 days. We will utilize Ex vivo samples from the cohort to test the effect of immunotherapeutic compounds on host expression profiles. Accomplishing these goals will have broad reaching translational implications for: (1) identifying at-risk groups, and (2) prioritizing compounds that can be used to improve clinical outcomes in future immunotherapy trials.

Project description:Cerebral Malaria (HCM) is a serious neurological complication caused by Plasmodium falciparum infection. Currently the only treatment for HCM is the provision of anti-malarial drugs; however, such treatment by itself often fails to prevent death or development of neurological sequelae. To identify new potential adjunct treatments for HCM, we performed a non-biased whole brain transcriptomic time-course analysis of anti-malarial drug chemotherapy of murine experimental CM (ECM).

Project description:Chronic activation of inflammatory pathways and suppressed interferon are hallmarks of immunosuppressive tumors. Previous studies have shown that CD11b integrin agonists could enhance anti-tumor immunity through myeloid reprograming, but the underlying mechanisms remain unclear. Herein we find that CD11b agonists alter tumor-associated macrophage (TAM) phenotypes by repressing NF-κB signaling and activating interferon gene expression simultaneously. Repression of NF-κB signaling involves degradation of p65 protein and is context independent. In contrast, CD11b agonism induces STING/STAT1 pathway-mediated interferon gene expression through FAK-mediated mitochondrial dysfunction, with the magnitude of induction dependent on the tumor microenvironment and amplified by cytotoxic therapies. Using tissues from phase I clinical studies, we demonstrate that GB1275 treatment activates STING and STAT1 signaling in TAMs in human tumors. These findings suggest potential mechanism-based therapeutic strategies for CD11b agonists and identify patient populations more likely to benefit.
