Project description:Purpose: The goals of this study are to compare the transcriptome profilings (RNA-seq) of CD45 positive immune cells in the control, polyIC, anti-PD-L1 and combination groups. Methods: All the CD45 positive immune cells of the mouse liver in control, polyIC, anti-PD-L1 and combination groups at day 24 were isolated, after hydrodynamic trasfection of c-Myc and N-ras at day 0. Then the mRNA profiles were generated by sequencing, in triplicate, using Illumina HiSeq 4000. RNA-seq generated raw data were aligned to the GRCm38 mouse reference genome using Star program (2.3.0). Gene differential expression analysis was performed using Cuffdiff to obtain the expression levels of genes in each sample.Gene Set Enrichment Analysis (GSEA) analysis was performed for pathway analysis online (software.broadinstitute.org/gsea). Results: By comparing to the control group, the number of differentially expressed genes and the overlaps in other groups were listed by Venn diagram. There were a total of 1654 differentially expressed genes specifically in the combination group. Gene set enrichment analysis (GSEA) of these 1654 genes demonstrated that the immune cells in the combination groups were particularly associated with adaptive and innate immune system and the progresses of antigen processing and presentations. The key genes involved in interferon-g (IFg) signaling pathway and some key genes, as indicators of anti-tumor cytotoxic function, were listed by heatmap. Conclusions: The RNA-seq data further confirmed that combination of polyIC and PD-L1 Ab dramatically enhanced the anti-tumor immunity in the liver, especially the adaptive immune response.

Project description:Side population (SP) cells are highly enriched in stem and progenitor cells. CD45+and CD45- SP cells were found at all developmental lung stages with the highest frequency of cells present at embryonic day 17.5 (E17.5), In order to clarify the role of these cells in lung development, we used oligonucleotide microarrays to evaluate their gene expression profiles. To do this, oligonucleotide gene arrays were performed in triplicate on RNA derived from CD45+ and CD45-SP cells, and CD45+ and CD45- non-SP populations (main population; MP) isolated from the E17.5 lung.

Analysis of differential genetic expression by dietary intervention using moderate-carbohydrate restriction diet (MCD) using a transgenic liver cancer model developed by a simple hydrodynamic transfection method.

Project description:We performed survival analysis of control and MCD groups, and explored underlying tumor suppression mechanisms after dietary intervention, focused on alterations in the energy-dependent signaling pathways, histone modifications, and global gene expression differences on cDNA microarray study. Five- week- old male C57BL6 mice were randomly divided into two groups and fed control diet (control group, LabDiet, Brentwood, MO, USA) or moderate restriced carbohydrate diet formula (MCD, Treat group) in a specific pathogen free zone. All procedures were approved by the institutional animal use and care committee. Following a preliminary feeding of each diet formula for two week, HrasG12V / shp53 / GFP4 gene containing transposon vector were injected into mouse tail vein by hydrodynamic injection method. After 4 weeks of diet supplementation, all mice were sacrificed. Mouse liver tissue was excised for microarray analysis.

Comparison of gene expression profile among four groups of mice that developed under the different combination of genes and environments

Project description:To investigate the mechanisms underlying the emergence of schizophrenic phenotypes, especially in sucrose fed Glo1 heterozygote mice, we conducted a transcriptome analysis of PFC regions using microarray experiments among the four groups of mice. The coagulation factor V, which is an essential participant in the processes of production of fibrin from fibrinogen, ranked seventh on the list of transcripts that exhibited more than doubled expression levels in sucrose fed Glo1 heterozygote mice compared with the remaining three groups. This result supports our idea that angiopathy with fibrrin deposition is a novel phenotype in schizphrenia.
