Project description:). In this study we describe the effect of drought combined or not with S-deficiency on pea productivity, nutrient partitioning between plant parts and seed quality traits. A moderate water stress was applied at the beginning of the reproductive phase, a period during which drought frequently occurs in the field. Sharp differences in the plant and seed characteristic in response to the combined stress compared to individual stresses were observed, highlighting synergistic effects on reproductive processes and antagonistic effects on seed protein composition, which could be explained by changes in the sink strength of the seeds for nitrogen.

Project description:Pea (Pisum sativum) is one of the most abundant and inexpensive alternate sources of protein. Pea proteins have a good distribution of essential amino acids, but they contain relatively low amounts of sulfur-containing amino acids (methionine and cysteine) and tryptophan in comparison to cereal crops. Pea isolate samples from different time-points during fermentation with Aspergillus oryzae, were used for shotgun mass spectrometry (LC-MS/MS) analysis to identify the proteome matrix changes.

Project description:Pea (Pisum sativum L.) is an important source of dietary proteins. Nutrient recycling from leaves contributes to the accumulation of seed proteins and is a pivotal determinant of protein yields in this grain legume. The aim of this study was to unveil the transcriptional regulations occurring in pea leaves before the sharp decrease in chlorophyll breakdown. As a prelude to this study, a time-series analysis of 15N translocation at the whole plant level was performed, which indicated that nitrogen recycling among organs was highly dynamic during this period and varied depending on nitrate availability. Leaves collected on vegetative and reproductive nodes were further analyzed by transcriptomics. The data revealed extensive transcriptome changes in leaves of reproductive nodes during early seed development (from flowering to 14 days after flowering), including an up-regulation of genes encoding transporters, and particularly of sulfate that might sustain sulfur metabolism in leaves of the reproductive part. This developmental period was also characterized by a down-regulation of cell wall-associated genes in leaves of both reproductive and vegetative nodes, reflecting a shift in cell wall structure. Later on, 27 days after flowering, genes potentially switching the metabolism of leaves toward senescence were pinpointed, some of which are related to ribosomal RNA processing, autophagy, or transport systems. Transcription factors differentially regulated in leaves between stages were identified and a gene co-expression network pointed out some of them as potential regulators of the above-mentioned biological processes. The same approach was conducted in Medicago truncatula to identify shared regulations with this wild legume species. Altogether the results give a global view of transcriptional events in leaves of legumes at early reproductive stages and provide a valuable resource of candidate genes that could be targeted by reverse genetics to improve nutrient remobilization and/or delay catabolic processes leading to senescence.

Project description:Pea (Pisum. sativum L.) is a traditional and important edible legume that can be sorted into grain pea and vegetable pea according to their harvested maturely or not. Vegetable pea by eating the fresh seed is becoming more and more popular in recent years. These two type peas display huge variations of the taste and nutrition, but how seed development and nutrition accumulation of grain pea and vegetable pea and their differences at the molecular level remains poorly understood. To understand the genes and gene networks regulate seed development in grain pea and vegetable pea, high throughput RNA-Seq and bioinformatics analysis were used to compare the transcriptomes of vegetable pea and grain pea developing seed. RNA-Seq generated 18.7 G raw data, which was then de novo assembled into 77,273 unigenes with a mean length of 930 bp. Functional annotation of the unigenes was carried out using the nr, Swiss-Prot, COG, GO and KEGG databases. There were 459 and 801 genes showing differentially expressed between vegetable pea and grain pea at early and late seed maturation phases, respectively. Sugar and starch metabolism related genes were dramatically activated during pea seed development. The up-regulated of starch biosynthesis genes could explain the increment of starch content in grain pea then vegetable pea; while up-regulation of sugar metabolism related genes in vegetable pea then grain pea should participate in sugar accumulation and associated with the increase in sweetness of vegetable pea then grain pea. Furthermore, transcription factors were implicated in the seed development regulation in grain pea and vegetable pea. Thus, our results constitute a foundation in support of future efforts for understanding the underlying mechanism that control pea seed development and also serve as a valuable resource for improved pea breeding.

Project description:In angiosperms, the mature seed consists of an embryo (E), a seed coat (SC), and, in many cases, an endosperm. In contrast to knowledge about embryo and endosperm, we have relatively little knowledge of SC, especially at the genomics level. In this study, we analyzed the gene expression during seed development using the panel of cultivated and wild pea genotypes. We report the comprehensive gene expression changes related both to development as well as domestication status. Analysis of seed developmental stages revealed extensive modification of gene expression between wild pea progenitor and cultivated pea crop. A significant difference in gene expression dynamics appeared between early and late developmental stages D1, D2, and D3, D4, D5 in wild pea genotypes, where the expression was increased 3-5-fold and 5-10-fold, respectively. Our work extends knowledge about the role of the seed coat during pea seed development. We described gene expression dynamic resulting in specific metabolic profiles providing new insight into pea domestication.

Project description:Although poplars are good candidates for the phytoremediation of cadmium (Cd)-polluted soils and sulfur is a key player in modulating Cd detoxification and accumulation in plants, the physiological and molecular mechanisms underlying sulfur-mediated Cd accumulation remain largely unknown. To investigate physiological and transcriptomic regulation mechanisms of poplars in response to Cd exposure and different sulphate (S) supply levels, Populus deltoides saplings were exposed to either 0 or 50 µM Cd together with one of low, moderate and high S levels. Cd was accumulated in the leaves of Cd-exposed poplars under moderate S condition, and its accumulation is inhibited by low S but tended to increase by high S. Sulfur nutrition became deficient in Cd-treated leaves, and it was aggravated by low S but improved by high S. Concentrations of cysteine and glutathione were increased in Cd-treated leaves, and the Cd-induced increases were limited by low S but stimulated by high S. In line with the physiological changes, a number of mRNAs and microRNAs (miRNAs) involved in Cd accumulation and sulfur assimilation were identified and the miRNAs-mRNAs networks were dissected. In the networks, miR395 and miR399 members were identified as hub miRNAs and their targets were ATP sulfurylase 3 (ATPS3), and phosphate 2 (PHO2) and beta galactosidase 1 (BGAL1), respectively. These results suggest that Cd-induced changes in Cd accumulation and sulfur assimilation are exacerbated by low S but alleviated by high S supply, and that miRNAs-mRNAs regulatory networks play pivotal roles in sulfur-mediated Cd detoxification and accumulation in poplars.
