Project description:Many insects are associated with endosymbionts that influence the feeding, reproduction, and distribution of their hosts. Although the small green mirid, Nesidiocoris tenuis (Reuter) (Hemiptera: Miridae), a zoophytophagous predator that feeds on plants as well as arthropods, is a globally important biological control agent, its microbiome has not been sufficiently studied. In the present study, we assessed the microbiome variation in 96 N. tenuis individuals from 14 locations throughout Japan, based on amplicon sequencing of the 16S ribosomal RNA gene. Nine major bacteria associated with N. tenuis were identified: Rickettsia, two strains of Wolbachia, Spiroplasma, Providencia, Serratia, Pseudochrobactrum, Lactococcus, and Stenotrophomonas. Additionally, a diagnostic PCR analysis for three typical insect reproductive manipulators, Rickettsia, Wolbachia, and Spiroplasma, was performed on a larger sample size (n = 360) of N. tenuis individuals; the most prevalent symbiont was Rickettsia (69.7%), followed by Wolbachia (39.2%) and Spiroplasma (6.1%). Although some symbionts were co-infected, their prevalence did not exhibit any specific tendency, such as a high frequency in specific infection combinations. The infection frequency of Rickettsia was significantly correlated with latitude and temperature, while that of Wolbachia and Spiroplasma was significantly correlated with host plants. The predominance of these bacteria and the absence of obligate symbionts suggested that the N. tenuis microbiome is typical for predatory arthropods rather than sap-feeding insects. Rickettsia and Wolbachia were vertically transmitted rather than horizontally transmitted from the prey. The functional validation of each symbiont would be warranted to develop N. tenuis as a biological control agent.

Poly(A) mRNA was isolated from 20 µl total RNA using Oligo (dT) magnetic beads and then was broken into short fragments (about 200bp) in the presence of fragmentation buffer at 94 °C for 5 min. The mRNA samples were used to construct the cDNA libray with mRNA-Seq assay.The libraries were sequenced using HiSeq4000 (Illumina) in paired-end mode, creating reads with a length of 150 bp.

Project description:Nesidiocoris tenuis (Reuter) (Hemiptera: Miridae) is an effective predator of multiple pests of vegetable crops, such as thrips, mites, aphids, whiteflies, leafminers. It is mass-reared and released for augmentative biocontrol programs mainly aimed at controlling whiteflies and Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) in greenhouses and open field. We evaluated the lethal and sublethal toxicity upon N. tenuis adults of label doses of three insecticides (pyriproxyfen, spirotetramat, cypermethrin) and seven fungicides (benomyl, chlorothalonil, copper oxychloride, cyazofamid, fluopicolide + propamocarb hydrochloride (FPH), penconazol, trifloxystrobin) commonly used in various crops. Two exposure routes were tested: (i) contact with dry residues of insecticides or fungicides on tomato sprouts and (ii) multiple exposure to these chemicals via topical sprays on adults which foraged on treated sprouts; and fed on treated eggs of Ephestia kuehniella (Zeller) (Lepidoptera: Pyralidae) simultaneously. Mortality and reproductive capacity were investigated as indicators of lethal and sublethal effects on N. tenius. The tested insecticides and fungicides were all classified as harmless when predator was exposed only to the dry residues of each. However, the multiple exposure to either cypermethrin, benomyl, chlorothalonil, copper oxychloride or trifloxystrobin caused significant mortality of N. tenuis adults. Cypermethrin also significantly reduced its reproductive capacity. Interestingly, N. tenuis produced a higher number of progeny when exposed to fungicides penconazol and FPH in both exposure scenarios. Overall, findings suggest that the two insecticides, pyriproxyfen and spirotetramat but not cypermethrin, and all tested fungicides can be considered compatible with N. tenuis.

Project description:The Rio Grande Valley is one of the most productive agricultural areas in the U.S and is located in the southernmost part of Texas. In October 2013, we detected an exotic plant bug, Nesidiocoris tenuis Reuter (Hemiptera: Miridae: Bryocorinae) occurring in the region. Nesidiocoris tenuis has zoophytophagous habits; however, in the absence of insect prey, it feeds on its plant hosts. After its morphological and genetic identification, this study monitored the population of N. tenuis in its introduction phase in commercial fields and corroborated its establishment in research fields for three years. Populations of N. tenuis were high during the fall and low during winter. This study found that N. tenuis populations were higher in tomato fields as compared to adjacent pepper, okra, and squash fields, indicating its host preferences during the introduction phase. Recurrent population growth patterns suggest that N. tenuis was established in Rio Grande Valley with permanent populations in tomato fields. In addition, N. tenuis populations were affected by tomato cultivar selection and by plastic mulch color. The presence of N. tenuis could establish a new trophic insect relationship for vegetable production. However, it is unknown if the presence of N. tenuis may help to control pests of economic importance, such as whiteflies in cotton, or become a pest on sesame, an emerging crop.

Project description:Zoophytophagous predators provide biocontrol services in various major crops of modern horticulture due to the combination of its predatory capacity and the induction of plant defenses derived from its phytophagy. However, under certain conditions of prey scarcity, these natural enemies can inflict plant damage. Exploitation of genetic variation and subsequent selective breeding on foraging traits is a potential alternative to overcome this inconvenience. In this study, we quantified the genetic variation of phytophagy and zoophagy of Nesidiocoristenuis (Reuter) (Hemiptera: Miridae), a zoophytophagous predator widely used in tomato crops to suppress key pests. We compared nine isofemale lines on their capacity to produce necrotic rings and wilting on tomato plants as a proxy for phytophagy, as well as their efficacy to prey on Ephestia kuehniella Zeller (Lepidoptera: Pyralidae) eggs, as a proxy for zoophagy. Differences between isofemale lines in phytophagy and zoophagy indicated a genetic basis. Variation found in the zoophagy levels was larger than that in phytophagy levels. Our results showed that there is a genetic basis for the variation observed in the feeding behavior of isofemale lines of N.tenuis, highlighting the potential importance of selective breeding for such traits of biocontrol interest.
