Project description:Extracellular vesicles (EVs) comprise a novel mechanism for cellular communication by transferring protein and nucleic acid cargo between cells. By utilizing a sequential filtration protocol, we were able to effectively separate microvesicles (MVs), exosomes and membrane-unbound ribonuclear protein complexes (RNPs) released from EGFR- (Acquaviva, 2011) and PDGFR-driven (Jun, 2018) genetically engineered mouse glioblastoma (GMB) cell lines. Regardless of the oncogenic driver of the tumor, different species of RNA partition into different extracellular compartments, with the mRNA in MVs most closely representing the cellular transcriptome, while miRNAs and tRNA fragments segregate into exosomes and RNPs, respectively. This EV RNA profile, combined with the cognate proteomics data, also serves as a reference for future GBM biomarker discovery.

Project description:Cancer extracellular vesicles (EVs) are highly heterogeneous, which impedes our understanding of their function as intercellular communication agents and biomarkers. To deconstruct this heterogeneity, we analyzed extracellular RNAs (exRNAs) and extracellular proteins (exPTNs) from size fractionation of large, medium, and small EVs and ribonucleoprotein complexes (RNPs) from mouse glioblastoma cells by RNA sequencing and quantitative proteomics. mRNA from medium-sized EVs most closely reflects the cellular transcriptome, whereas small EV exRNA is enriched in small non-coding RNAs and RNPs contain precisely processed tRNA fragments. The exPTN composition of EVs and RNPs reveals that they are closely related by vesicle type, independent of their cellular origin, and single EV analysis reveals that small EVs are less heterogeneous in their protein content than larger ones. We provide a foundation for better understanding of segregation of macromolecules in glioma EVs through a catalog of diverse exRNAs and exPTNs.

Glioma proteome organization under oncolytic adenoviral infection

Project description:Glioblastoma multiforme (GBM) is the most common and aggressive type of malignant glioma. Oncolytic adenoviruses are being modified to exploit the aberrant expression of proteins in tumor cells to enhance tumor tropism and glioma-selective replication. E1A mutant adenovirus Delta-24-RGD has shown favorable toxicity profile and remarkable efficacy in a first-in-human phase I clinical trial. However, the comprehensive modulation of glioma metabolism in response to Delta-24-RGD infection is poorly understood. Integrating mass spectrometry based-quantitative proteomics, physical and functional interaction data, and biochemical approaches, we conducted a cell-wide study of cytosolic, nuclear, and secreted glioma proteomes throughout the early time course of Delta-24-RGD infection.

Project description:We used microarrays to select the genes associated glioma patients survival. This study aimed to define genes associated with survival. Expression profiling was performed on 50 gliomas. A gene classifier was developed. Fifty glioma patients were selected for RNA extraction and hybridization on Affymetrix microarrays.
