Project description:As the differentiation state of the cells is affected by epigenetic modification regulation, the abnormal epigenetic modification is an important factor leading to the intratumoral heterogeneity. To further understand of the intratumoral epigenetic heterogeneity, we performed Reduced Representation Bisulfite Sequencing and RNA sequencing for multiple regions within a breast tumor and exhibited the epigenetic maps of different regions. We detected intratumoral heterogeneity-related molecular features and further explored the correlation between tumor heterogeneity and tumor hypoxic microenvironment.

Approximately 81% of EGR1 peaks were within genic regions or nearby promoters, and over 45% EGR1 peaks were in the proximal promoter regions within 1 kb from transcription starting sites.

Project description:FHND004 and FHND008 are newly synthesized epidermal growth factor receptor (EGFR) inhibitor for the treatment of non-small cell lung cancer (NSCLC), they were obtained on the basis of AZD9291. We found that FHND004 and FHND008 had a strong inhibitory effect on the proliferation of multiple myeloma cells in vitro. Genome-wide maps of transcription showed different genes change in Multiple Myeloma cells after 4μM FHND004 or FHND008 intervention.

Project description:We report the application of sequencing technology for profiling of genome-wide chromatin accessible regions in mouse ES cells. We generated genome-wide chromatin-state maps of mouse embryonic stem cells maintained in pluripotent state (2iL) and differentiated for 48 and 96 hours (48 and 96). We find that the large majority of ESRRB motif-cointaining regions are found in 2iL and at 48 hours and that several formative genes contain time-specific oper regions at 48h, but not at earlier or later time-points.
