Project description:In the present study we examined the effect of hepatocyte-derived exosomes (HDE) on innate immune cells by treating a human monocyte cell line (THP-1 cells) with exosomes collected from the culture medium of acetaminophen (APAP)-treated and untreated primary human hepatocytes and comparing the cytokine response to THP-1 cells treated with mock- and medium-controls. We profiled the gene expression changes observed in the HDE-exposed monocytes prior to LPS stimulation. Findings from our research suggest that HDE may be essential for maintaining immune homeostasis.

Project description:Gene expression profiles of sandwich-cultured primary human hepatocytes exposed to 5 mM and 10 mM acetaminophen were used in a parallelogram approach in order to compare gene expression responses between rat and human using in vitro cellular models, hepatocytes, and between rat in vitro and in vivo. Experiment Overall Design: Samples were retrieved from acetaminophen treated human hepatocyte cultures from 5 individuals (5 biological replicates). Experiment Overall Design: Human hepatocytes from each replicate were treated with 0, 5, and 10 mM acetaminophen for 24 h. Experiment Overall Design: This resulted in (3x5) 15 dual channel arrays on which control (0 mM acetaminophen) and reference samples (0, 5, and 10 mM acetaminophen) were hybridized.

Gene expression data from acetaminophen-induced toxicity in human hepatic in vitro systems and clinical liver samples

Project description:In this study we conducted transcriptomics analyses of: (i) liver samples from patients suffering from acetaminophen-induced acute liver failure (n=3) and from healthy livers (n=2) and (ii) hepatic cell systems exposed to acetaminophen, including their respective vehicle controls. The investigated in vitro systems are: HepaRG cells, HepG2 cells and a novel human skinpostnatal stem cell-derived model i.e. human skin-precursors-derived hepatocyte-like cells (hSKP-HPC). Clinical samples were obtained after surgical removal of the explant liver of patients diagnosed with ALF due to acetaminophen intoxication and treated by orthotopic liver transplantation (n=3). Samples from healthy livers were obtained from individuals deceased from brain damage (n=2). Different samples of hSKP, HepG2 and HepaRG were obtained from the same cell batch of each cell system. All cells were exposed for 24 hours to their corresponding sub cytotoxic concentrations (IC10): IC10(hSKP-HPC)=18mM; IC10(HepaRG)=13mM; IC10(HepG2)=2mM. Experiments were conducted in triplicate. This dataset is part of the TransQST collection.
