Project description:Aroma is an important parameter for table grapes and wines; terpenes are typical compounds in Muscat-type grape cultivars and can be easily perceived by humans because of their low olfactory threshold. Volatile terpenes contribute directly to the aroma character, while glycoside-bound terpenes are potential aromatic compounds and can be changed to their volatile forms via hydrolysis. With gas chromatography-mass spectrometry and a solid-phase microextraction method, an automatic data analysis platform was constructed; terpene compounds were identified and quantified from three table grape cultivars at three stages during berry development, and the raw data were deposited in MetaboLights. Terpene metabolite accumulation profiles are presented in this article for integrative analysis with the transcriptome data and phenotypic data to elucidate the important candidate genes and mechanism for terpene biosynthesis. Our method has applications in the identification and quantification of terpene compounds with very low or trace concentrations.

Project description:A double cropping system has been commercially adopted in subtropical regions in southern China, where there is abundant sunshine and heat resources. In this viticulture system, the first growing season normally starts as a summer cropping cycle; then, the vine is pruned and forced by hydrogen cyanamide, resulting in a second crop in January of the next year. Due to climate differences between the two growing seasons,the transcriptome varies greatly between summer- and winter- grape berries. In this study, we conducted RNA-seq for two table grape varieties (Vitis vinifera cv. Victoria and V. vinifera cv. Muscat Superior) at three developmental stages (pre-veraison, post-veraison and full maturity).The transcriptome between summer- and winter- grape berries were compared.

Project description:Vitis vinifera is widely grown worldwide for making wine and for use as table grapes. Of the existing cultivars, some have a floral and fruity flavour, referred to as a Muscat flavour. It is well-documented that this flavour originates from a series of terpene compounds, but the mechanism of terpene content differences among the Muscat-type cultivars remains unclear. Transcript and terpene metabolite profiles were integrated to elucidate the molecular mechanism of this phenomenon. In this research, three genotypes with different aromatic strengths were investigated by RNA sequencing. A total of 27 fruit samples from three biological replicates were sequenced on Illumina HiSeq 2000 at three stages, corresponding to the veraison; berries had intermediate Brix value and were harvest-ripe. After quality assessment and data clearance, a total of 254.18 Gb of data with more than 97% Q20 bases were obtained, approximately 9.41 Gb data were generated per sample. These results will provide a valuable dataset for the discovery of the mechanism of terpene biosynthesis.

Project description:Monoterpenes are typical aroma components of muscat grape cultivars. However, the molecular mechanism of monoterpene biosynthesis between muscat and non-muscat grape remains unclear. In this study, two F1 hybrids with different aroma profiles from the cross between 'Midknight Beauty' and 'Red Globe' grapes were chosen for comprehensive transcriptome and metabolome analysis to study the unique monoterpenoid accumulation pattern and related candidate genes. The results showed that in the hybrid progeny, the concentrations of most terpenoids differed at different times, and the concentration of most terpenoids increased from the pre-veraison to veraison, followed by a decreasing trend from veraison to maturity. In the F1 generation, the concentration of monoterpenes accumulated in Muscat grapes was significantly higher than that in Neutral grapes. Furthermore, linalool，α-terpineol, and L-α-terpineol may be important odorants that affect the floral characteristics of Muscat grapes because of their high odor activity values (OAVs). Transcriptional analysis identified 3 VvDXS (LOC100249323, LOC100268079, LOC100247834), 1 VvHDS (LOC100257071), 1 VvMVK (LOC100251148), 1 VvHMGR (LOC100265082), and 1 VvGGPPS (LOC100257234) in the MEP and MVA pathways. Meanwhile, 11 TPS genes that may be involved in the regulation of monoterpenes were identified. Among them, VvTPS (LOC100853562) showed consistency with the accumulation of monoterpenes and had a high expression level， MYB (LOC100255488), GATA (LOC100262474), and ERF (LOC100255943, LOC100260401) were selected as candidate transcription factors (TFs). The findings provide new insights for the subsequent functional validation and further investigation of genes related to monoterpene synthesis in Muscat aroma grapes.

Project description:We used Illumina RNA-Seq technology to carry out digital gene expression profiling of Dormant and non dormant buds of Shine Muscat Cultivar.127 pathways were annotated by Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, and metabolic, biosynthesis of secondry metabolite and. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis demonstrated that DEGs were involved in various metabolic processes, including phytohormone metabolism. The newly generated transcriptome and gene expression profiling data provide valuable genetic information for revealing transcriptomic variation during dormant and non dormant period in Grapevine and ultimately helpful in uplifting of table grape industry.

Project description:The experiment was aimed at describing similarities and differences between grapevine flesh and skin ripening at the transcriptome level. Transcriptional profiles throughout flesh and skin ripening were followed in three biological replicas during two consecutive growing seasons in a table grape cultivar ‘Muscat Hamburg’. Berries were weekly picked off bunches (one bunch per vine from each selected vine) from june to august. For each sampling, berries were classified in five ripening stages based on monitoring developmental stages instead of a simple chronological sampling. They corresponded to pre-véraison (P), 50% véraison (V1), 100% véraison (V2), ripe 1 (R1), and ripe 2 R2). P samples were harvested as green, hard berries of at least 15 mm in size, according to touch-assessed hardiness. Véraison berries with half or full coloured skin surface corresponded to V1 and V2 samples. After véraison, ripening berries were classified according to their floatability in solutions with different NaCl densities, as an indication of the internal sugar concentration. At this stage berries were sampled with densities between 110-130 g NaCl/L for R1 and between 130-150 g NaCl/L for R2, which corresponded to commercially ripen table grape berries. Project description:In commercial fruit production, synchronized ripening and stable shelf life are important properties. The loosely clustered or non-bunching muscadine grape has unrealized potential as a disease resistant cash crop, but requires repeated hand harvesting due to its unsynchronized or long or heterogeneous maturation period. Genomic research can be used to identify the developmental and environmental factors that control fruit ripening and postharvest quality. This study coupled the morphological, biochemical, and genetic variations between ‘Carlos’ and ‘Noble’ muscadine grape cultivars with RNA-sequencing analysis during berry maturation. The levels of antioxidants, anthocyanins, and titratable acids varied between the two cultivars during the ripening process. We also identified new genes, pathways, and regulatory networks that modulated berry ripening in muscadine grape. These findings may help develop a large-scale database of the genetic factors of muscadine grape ripening and postharvest profiles and allow the discovery of the factors underlying the ripeness heterogeneity at harvest. These genetic resources may allow us to combine applied and basic research methods in breeding to improve table and wine grape ripening uniformity, quality, stress tolerance, and postharvest handling and storage.
