Project description:During pregnancy, a high-fructose diet (HFrD) exacerbates gestational insulin resistance and is associated with an increased risk of gestational diabetes. The relationship between liver ChREBP and gestational insulin resistance remains elusive. To address this question, we fed pregnant mice either a high-fructose diet or a normal diet. On the 17th day of pregnancy, we extracted liver tissue from the mice and conducted comparative transcriptomic analysis to elucidate the alterations in liver ChREBP during gestational insulin resistance.

Gene expression in the liver, effect of maternal high-fat diet during or prior to pregnancy

Project description:The present study aimed to examine the effect of high-fat diet prior to pregnancy on the liver of mouse offspring. Female C57BL/6J mice were fed a normal chow (15.2% fat by energy) (CTR and CTR-PP groups) or a high-fat chow (31.2% fat by energy) (HFD and HFD-PP groups) for 3−4 weeks and then mated with male C57BL/6J mice fed normal chow. Some mothers continued on the same diet until pups reached 21 days of age (CTR and HFD), and others were fed the different chows from gestational day 0 (CTR-PP and HFD-PP) to determine the effects of a high-fat diet during the pre-pregnancy period in HFD-PP/CTR and HFD/CTR-PP comparisons. RNA sample was taken from liver of 3-week-old mouse prenatally received high-fat diet prior to pregnancy, during pregnancy and lactation, or through prior to and during pregnancy and lactation, while control RNA was taken from control counterpart prenatally received normal diet alone. Comparisons among groups were made by one-color method with normalized data from Cy3 channels for data analysis.

Mouse gestational protein restriction: Newborn offspring liver and hindleg muscle

Project description:Gestational protein restriction is a model for low birth size. We hypothesized that taurine supplementation would protect against changes in newborn liver and muscle caused by a maternal low protein diet. Pregnant mouse dams were subjected to different diet schemes from day 1 of pregnancy until birth. Pups were killed following birth and liver and hindleg skeletal muscle taken out and frozen at -80C until analysis. Diet schemes: Normal Protein (20% casein; NP), Normal Protein + taurine (1% taurine supplementation in water ad libitum; NP+tau), Low Protein (8% casein; LP) and LP+tau The liver and muscle samples were normalized separately.

The Amniotic Fluid Proteome Changes Across Gestation in Humans and Rhesus Macaques

Project description:Amniotic fluid is a complex biological medium that offers mechanical protection and nutrition to the fetus, and also plays a key role in normal fetal growth, organogenesis, and potentially fetal programming. Amniotic fluid is also critically involved in longitudinally shaping the in utero milieu during pregnancy. Yet, the molecular mechanism of action by which amniotic fluid regulates fetal development is ill-defined partly due to an incomplete understanding of the evolving composition of the amniotic fluid proteome. Prior research consisting of cross-sectional studies suggests that the amniotic fluid proteome changes as pregnancy advances, yet longitudinal alterations have not been confirmed because repeated sampling is prohibitive in humans. We therefore performed serial amniocenteses at early, mid, and late gestational time-points within the same pregnancies in a rhesus macaque model. Longitudinally-collected rhesus amniotic fluid samples were paired with gestational-age matched cross-sectional human samples. Utilizing LC-MS/MS isobaric labeling quantitative proteomics, we demonstrate considerable cross-species similarity between the amniotic fluid proteomes and large scale gestational-age associated changes in protein content throughout pregnancy. This is the first study to establish a reference proteomic profile across gestation. This non-human primate model holds promise as a translational platform for amniotic fluid studies and to identify adversely affected pregnancies.

Gene expression in the mouse embryo following early pregnancy exposure to ethanol or nicotine

Project description:The in utero environment is a critical determinant of the immediate and future health of the developing fetus. Two of the most commonly used drugs during pregnancy are alcohol and nicotine. While prolonged gestational exposure to alcohol or nicotine has been associated with a range of adverse outcomes in the offspring, the consequences of exposure during early gestation only are less well understood. Here, we use mouse models of relatively moderate early gestational ethanol or nicotine exposure to profile gene expression in whole male embryos at 9.5 days post coitum using an Illumina microarray.

Project description:Chronic hypertension during pregnancy affects both mother and infant but its pathological mechanisms are unclear. Due to ethical limitations human data is available in term tissue only- so deeper understanding of the vascular changes during early pregnancy are unknown. This study utilises a known rat model of chronic hypertension to study changes in gene expression at an early gestational time point. After 6 days of pregnancy animals were sacrificed and main uterine arteries harvested for RNA-Seq (or age-matched virgin rats as controls).

Placenta-derived exosomes as early biomarker of preeclampsia - Potential role of exosomal miRNAs across gestation.

Project description:Preeclampsia (PE) and its related diseases is one of the most significant pregnancy complications, occurring at an incidence of 3-5% and is responsible for nearly 40% of premature births delivered before 35 weeks. There is a need to develop strategies for early prediction of patients who will develop PE, in order to establish preventive strategies to reduce the prevalence and severity of the disease and their associated aftereffects. Therefore, a prospective cohort of patients were recruited at three time points during pregnancy for each patient (i.e. 11-14 (early gestational age), 22-24 (middle gestational age) and 32-36 (late gestational age) weeks gestation). A retrospective stratified study design was used to characterize miRNAs in exosomes derived from maternal plasma of normal (n=32 per time point) and PE (n=15 per time point) pregnancies.
