Project description:Exploitation of the immune system has emerged as an important therapeutic strategy for acute lymphoblastic leukemia (ALL). However, the mechanisms of immune evasion during leukemia progression remain poorly understood. We sought to understand the role of calcineurin in ALL and observed that knocking-down calcineurin B (CnB) in leukemia cells dramatically prolonged survival in immune-competent but not immune-deficient recipients. Furthermore, immune-competent recipients were protected from challenge with leukemia if they were first immunized with CnB-deficient leukemia, suggesting robust adaptive immunity. In the bone marrow, recipients of CnB-deficient leukemia had expansion of T cell populations as compared to controls. Gene expression analyses of leukemia cells extracted from bone marrow identified Cn-dependent significant changes in the expression of immuno-regulatory genes. We confirmed increased secretion of IL-12 from CnB-deficient leukemia cells, sufficient to induce T cell activation ex vivo, an effect that was abolished when IL-12 was neutralized. Strikingly, recombinant IL-12 prolonged survival of mice challenged with highly aggressive B-ALL. Moreover, gene expression analyses from children with ALL demonstrate prolonged survival in those with higher expression of either IL-12A or IL-12B. These data suggest that leukemia cells are dependent upon calcineurin for immune evasion by restricting the regulation of pro-inflammatory genes, particularly IL-12.

Project description:CNIs drastically modify the Treg specific transcriptional program in vivo in an IL-2 dependent manner CD4+CD25+FOXP3+ regulatory T cells (Tregs) constitute a heterogeneous lymphocyte subpopulation essential for establishing peripheral immune tolerance. Interleukin-2 (IL-2) plays a critical role in maintaining immune homeostasis promoting optimal development, survival and function of Tregs. Because of their suppressive properties, manipulation of Tregs represents a clear target to modulate immune responses in transplantation and autoimmunity. However, the specific effects of adjunctive immunosuppressive drugs to the maintenance and function of the different Treg subsets remain unclear. Calcineurin inhibitors (CNIs), which are the mainstay immunosuppressants in transplantation, are the most effective agents in controlling alloreactive effector T cells, but also hamper Treg activity. In this study we sought to investigate the differential effects of CNIs on the homeostasis of Treg subsets, and the extent to which these effects are dependent on the overall availability of IL-2.

Project description:The intrinsic role of endogenous IL-17A in spontaneous intestinal tumorigenesis has not been addressed previously to our knowledge. Ablation of IL-17A significantly reduced tumor development in mice bearing a heterozygote mutation in the adenomatous polyposis coli (APC) gene (Apc(Min/+) mice). There was also a decrease in inflammatory cytokines and proinflammatory mediators, reduced infiltration of lymphocytes including T cells, and preservation of intestinal architecture and the presence of APC protein in intestinal epithelial cells. Interestingly, IL-17A ablation also corrected immunological abnormalities such as splenomegaly and thymic atrophy in Apc(Min/+) mice. CD4 T cells from Apc(Min/+) mice showed hyperproliferative potential in vitro and in vivo and increased levels of IL-17A and IL-10. The effector CD4 T cells from Apc(Min/+) mice were more resistant to regulatory T cell-mediated suppression. Finally, these CD4 T cells induced colitis in immunodeficient mice upon adoptive transfer, whereas the ablation of IL-17A in CD4 T cells in Apc(Min/+) mice completely abolished this pathogenic potential in vivo. Taken together, our results show that CD4 T cell-derived IL-17A promotes spontaneous intestinal tumorigenesis with altered functions of CD4 T cells in Apc(Min/+) mice.

Project description:Engineered cytokine-based approaches for immunotherapy of cancer are poised to enter the clinic, with IL-12 being at the forefront. However, little is known about potential mechanisms of resistance to cytokine therapies. We found that orthotopic murine lung tumors were resistant to systemically delivered IL-12 fused to murine serum albumin (MSA, IL12-MSA) due to low IL-12R expression on tumor-reactive CD8+ T cells. IL2-MSA increased binding of IL12-MSA by tumor-reactive CD8+ T cells, and combined administration of IL12-MSA and IL2-MSA led to enhanced tumor-reactive CD8+ T cell effector differentiation, decreased numbers of tumor-infiltrating CD4+ regulatory T (Treg) cells, and increased survival of lung tumor-bearing mice. Predictably, the combination of IL-2 and IL-12 at therapeutic doses led to significant dose-limiting toxicity. Administering IL-12 and IL-2 analogs with preferential binding to cells expressing IL12rb1 and CD25, respectively, led to a significant extension of survival in mice with lung tumors while abrogating dose-limiting toxicity. These findings suggest that IL-12 and IL-2 represent a rational approach to combination cytokine therapy whose dose-limiting toxicity can be overcome with engineered cytokine variants.

Project description:Gene expression was quantified in crz1 delete S. cerevisae cells over a time course following calcineurin activation by CaCl2. Cells were pretreated with buffer or the calcineurin inhibitor FK506 to allow the identification of calcineurin-regulated genes. A strain with a calcineurin-resistant mutation in the transcription factor Hcm1 was also examined. We show that calcineurin activation leads to downregulation of a many clusters of cell cycle-regulated genes, including those expressed at the G1/S and G2/M transitions.

Project description:We have analyzed how interaction of calcineurin with perinuclear signalosome AKAP6 affects the expression profile of transcription factors involved in cAMP/calcium signaling. We have found that calcineurin interacts with AKAP6 in calcium-dependent manner and by using anchoring disruptors, we demonstrated the importance of this interaction for depolarization-induced outgrowth of hippocampal neurons.

Project description:Following the initial demonstration of Helicobacter pyloriâ??s pathogenic potential, evidence has been accumulated that H. pylori is the leading cause of gastric ulcers, carcinoma and lymphoma3. Cholesterol is a physiological constituent of membranes critical for their biophysical properties, but is stigmatised as mediating detrimental effects in obesity and cardiovascular disease. Since H. pylori is auxotrophic for cholesterol, we explored the assimilation of cholesterol by H. pylori upon infection. Here we show that H. pylori follows a cholesterol gradient and extracts the lipid from plasma membranes of epithelial cells for subsequent glycosylation. Cholesterol promotes phagocytosis of H. pylori by antigen-presenting cells such as macrophages and dendritic cells and enhances antigen-specific T cell responses. Consistently, cholesterol-rich diet during bacterial challenge leads to a reduction of the H. pylori burden in the stomach. Intrinsic a-glycosylation of cholesterol abrogates phagocytosis of H. pylori and subsequent T cell activation.
