Project description:High mobility group (HMG) proteins play an important role in regulation of gene transcription through modulate the structure of DNA. In this study, OsHMGB707, a HMG gene localized in rice drought resistance QTL interval, was isolated and the function on rice stress resistance was identified. Overexpression of OsHMGB707 significantly enhanced the drought resistance of the transgenic rice plants, whereas the OsHMGB707-RNAi transgenic rice plants exhibited slightly decrease in drought stress tolerance. To search the downstream genes regulated by OsHMGB707, we performed microarray analysis of the OsHMGB707-overexpressing, OsHMGB707-RNAi and wild-type plants under both normal conditions using Affymetrix Rice Genome Genechip. 21-day-old plants of the OsHMGB707-overexpressing line OE1, OsHMGB707-RNAi line RNAi1 as well as the wild-type plants were used in the normal condition.

Project description:To explore the effect of stable RNAi on the small RNA (sRNA) population in wheat, we constructed a sRNA library from hexaploid wheat that expresses an RNAi construct under the 35S promoter that targets the endogenous NO APICAL MERISTEM (TaNAM) gene. The presence of this RNAi transgene causes a 40% reduction in expression of the target genes as measured by quantitative RT-PCR and significantly delays senescence and reduces remobilization of N, Fe, and Zn to the grain. RNA was extracted from flag-leaves of transgenic NAM-RNAi plants at 12 days post-anthesis. RNA was collected from a pool of flag-leaves from four plants.

Project description:Mutant maize lines expressing RNAi gene silencing constructs directed to two distinct classes of chromatin associated proteins show heightened sensitivity to UV-B including sunburned necrosis and down regulation of chromatin genes (Casati and Walbot, 2006). We have available transgenic maize expressing an RNAi for these chromatin factors: a subunit of a chromatin remodeling complex R, which is a SWI2/SNF2-like protein containing a SNF2 N and a helicase C domain (CHC101, AI746001); and a methyl-CpG-binding protein (MDB101, AI737448). To better understand the processes that involve chromatin remodeling proteins in UV-B responses, we conducted transcriptome analysis of chromatin remodeling knockdown plants in under two radiation regimes: plants were illuminated with UV-B lamps using fixtures mounted 30 cm above the plants (Phillips, F40UVB 40 W and TL 20 W/12) for 8 h; leaf samples were collected immediately after the radiation treatment. The bulbs were covered with CA filters to exclude wavelengths lower than 280 nm (UV-B). As a control, the bulbs were covered with PE filters to exclude wavelengths lower than 320 nm (minus UV-B). We analyzed genes that are changed upon UV-B irradiation in the transgenic plants and compared to the responses in non transgenic plants. To differentiate between differences due to background specific genes expression, we used non-transgenic sibling as controls. For these experiments, we used Long oligonucleotides arrays from Agilent Technologies. These studies will help to dilucidate how alteration in chromatin remodeling protein proteins affect gene expression, and the transcriptome analysis will help to understand why transgenic plants are hypersensity to UV-B. Cinnamyl Alcohol Dehydrogenase (CAD) is the final enzyme involved in the monolignol biosynthesis pathway. Transgenic poplars were produced using RNAi strategy (pHellsgate 8 vector, Helliwell and Waterhouse, 2003) and exhibited the expected phenotype (red xylem, reduced CAD activity).Biological question (15 lines max):This experiment aims to characterize global gene expression in young xylem of transgenic RNAi-CAD poplars in comparison to WT poplars. Four-month-old poplar plants were inclined at 30° in the greenhouse and sampled after 26 days. Young differentiating xylem originating from the lower side of stems - opposite wood - (ODX) was sampled on each individual tree by scrapping slightly the debarked stem with a scalpel. Samples were immediately flash frozen in liquid nitrogen, ground with mortar and pestle, and total RNAs were extracted from fine ground powder using the QIAGEN miRNeasy kit according to the manufacturer. One Affymetrix slide corresponds to a pool of RNA samples from 2-4 individual trees (WT, RNAi-CAD transgenic lines 2 and 19). Total number of slides = 2 genotypes (WT/RNAi line) x 1 tissue x 2 biological replicates = 4 slides were done.

Project description:Plants deal with cold temperatures via different signal transduction pathways. To elucidate the molecular mechanisms by which HaHB1 conferred tolerance to cold temperatures, we performed an exploratory transcriptome analysis comparing RNAs of 3-week-old transgenic and WT plants. Several transcripts encoding glucanases and chitinases were induced. Notably, there was not a considerable overlap with CBF regulon genes previously described in cold tolerance molecular mechanism. We propose that HaHB1 is involved in plant cold tolerance via the induction of proteins able to stabilize cell membranes and inhibit ice growth.
