Project description:Pomegranate skin extract could prevent fatty liver due to high fat diet in adult male Sprague Dawley rat. There are 3 groups of rats, feeding chow diet, high fat diet and high fat diet combined with pomegrante skin extract. After 8 weeks feeding, high fat diet group developped fatty liver but the other two groups still have healthy liver. We used microarrays to detail the global programme of gene expression underlying the fatty liver development and preventive effect of pomegranate skin extract on fatty liver. There are 3 treatment groups and 6 replicates for each group. The 3 treatment groups are control group feeding chow diet, high fat group feeding high fat diet and PE group feeding high fat diet and pomegranate skin extract. Rat livers were collecteded for RNA extraction and hybridization on Affymetrix microarrays.

Transcriptomic Characterization of Inhalation Phosphine Toxicity in Adult Male Sprague-Dawley Rats

Project description:Phosphine (PH3) is a highly toxic, corrosive, flammable, heavier-than-air gas that is a commonly used fumigant. Although the mechanism of toxicity is unclear, PH3 is thought to be a metabolic poison. PH3 exposure induces multi-organ toxicity and no effective antidotes or therapeutics have been identified. To better characterize the mechanism(s) driving PH3-induced toxicity we have performed transcriptomic analysis on conscious adult male Sprague-Dawley rats following whole-body inhalation exposure to phosphine gas at three concentrations. PH3 exposure induced concentration- and time-dependent changes in gene expression across multiple tissues.

Protein Expression in Liver Tissue and Bile of Elderly Sprague-Dawley Rats after Normothermic Machine Perfusion

Project description:Liver are frequently declined for transplantation due to the donor age. It is still unclear how donor age affects the graft quality. Normothermic machine perfusion (NMP) has been proposed as a useful assessment tool prior to transplantation. We aimed to compare the performance of young and elderly rat liver grafts in a small animal NMP model. Grafts from 24 rats were procured, either 3 or 12 months old and perfused for 6 hours at 37°C or stored on ice as a reference group (n=6/group). Livers in both NMP groups cleared lactate and produced bile, with similar pressure, bile production, and pH levels. However, older rat livers showed higher peak transaminase and urea levels. Proteomic analysis revealed differences in protein composition, particularly higher levels of proteins related to oxidoreductase and catalytic activity in older livers. Older age appeared to increase susceptibility to oxidative stress in the livers.
