Project description:We generated whole genome expression profiles from a homogeneous population of purified pacemaker neurons (ventral Lateral Neurons, LNvs) from wild type and clock mutant Drosophila. The study identifes a group of genes whose expression is highly enriched in LNvs compared to other neurons; and a second group of genes rhythmically expressed in LNvs in a clock-dependent manner. Drosophila larval brains (L3 stage) were taken from a 12:12hr light entrainment regime and dissected at ZT-3 or ZT-15. Larvae contained either Pdf-Gal4:UAS-GFP transgenes or Pdf-RFP to label the 8 LNvs pacemaker neurons per brain, allowing purification by flow cytometry. Brains were dissociated and LNvs purified by FACS, followed by RNA amplification and hybridization on Affymetrix microarrays.

Project description:The cellular composition of the brain and how it is affected by starvation, remains largely unknown. Here we introduce a single-cell transcriptome atlas of the entire Drosophila melanogaster first instar larval brain. We first assigned cell type identity based on the expression of previously characterized marker genes, allowing us to distinguish five major groups: neural progenitors cells, differentiated neurons, glial cells, undifferentiated neurons as well as non-neural cells corresponding to organs and structures located adjacent to the brain. All major classes were further subdivided into multiple subtypes based on cluster analysis, revealing critical biological features of various cell types. Moreover, we included two different feeding conditions: normal fed versus starved. After starvation, the transcriptional profile of several cell clusters were altered, while the overall composition of the brain remains unaffected. Intriguingly, different cell clusters show very distinct responses to starvation, suggesting the presence of cell-specific programs for nutrition availability. Establishing a single-cell transcriptome atlas of the larval brain provides a powerful tool to explore cell diversity, assess genetic profiles of neurogenic, neuronal and glial cell types. The analysis of neurotransmitters, neuropeptides and their respective receptors may further open the doors for functional studies.

Project description:The brain plays a key role in energy homeostasis, detecting circulating hormones from peripheral organs, nutrients, and metabolites, and integrating this information to control food intake and energy expenditure. However, the signals mediating communication between peripheral organs and brain are largely unknown. Here, we show that a group of neurons in the Drosophila larval brain expressing the adiponectin receptor (AdipoR) control systemic growth and metabolism. We identify glucose-regulated protein 78 (Grp78) as a circulating ligand for AdipoR. Grp78 is produced by fat cells in response to dietary sugar and modulates the activity of AdipoR-positive neurons. The terpenoid juvenile hormone (JH) serves as an effector for brain AdipoR signaling, reducing the levels of insulin signaling in peripheral organs. In conclusion, we identify a neuroendocrine axis whereby AdipoR neurons control systemic insulin responses by modulating peripheral JH function.

Project description:The larval brain of Ciona intestinalis has similar architecture to that of vertebrates, but is only composed of approximately 330 cells. Transgenic embryos that carried Ci-beta-tubulin(promoter)::Kaede exhibited robust Kaede expression in the larval brain. Kaede-expressing cells were isolated, and their transcriptome was compared with that of cells that did not express Kaede using an oligonucleotide-based microarray. Our analysis identified 565 candidate genes that were preferentially expressed in the larval brain, 77 of which have previously been reported to be brain-related. The 565 genes included transcription factors, such as Otx, en, Pax3/7, Prop-A, Lhx1, Six3/6, Unc4-A, FoxC, and DMRT1; and signal transduction molecules, such as FGF4/5/6, Hedgehog1, Hedgehog2, patched, Fringe1, and Dkk3. Nearly 30 of the identified genes coded for receptors for neurotransmitters, neuropeptides or hormone pepetides. In addition, 15 genes encoded neuropeptides and hormone peptides, five of which were novel. Our catalog of genes that are expressed in the Ciona larval brain provides a foundation for future studies exploring the complex gene regulatory networks that mediate chordate brain development and function. Project description:Male and female reproductive behaviors in Drosophila melanogaster are vastly different, but the neurons that express sex-specifically spliced fruitless transcripts (fru P1) underlie these behaviors in both sexes. How this set of neurons can generate such different behaviors between the two sexes is an unresolved question. A particular challenge is that fru P1-expressing neurons comprise only 2-5% of the adult nervous system, and so studies of adult head tissue or whole brain may not reveal crucial differences. Translating Ribosome Affinity Purification (TRAP) identifies the actively translated pool of mRNAs from fru P1-expressing neurons allowing a sensitive, cell-type-specific assay. Genes with TRAP mRNAs that are detected in both sexes are four times as likely to be male biased (1,210) as female biased (331) in fru P1-expressing neurons. This suggests a potential mechanism to generate dimorphism in behavior. The male-biased genes may direct male behaviors by establishing cell fate in a similar context of gene expression observed in females. These results suggest a possible global mechanism for how distinct behaviors can arise from a shared set of neurons.

Project description:A challenge in systems biology is to understand the gene regulatory networks that connect early cellular specification to terminal differentiation of specific cell types. In neurogenesis, neural specification has been well studied, but the link between the proneural transcriptional regulators of specification and the genes that must be activated to construct differentiated neurons is obscure. High resolution temporal profiling of gene expression reveals the events downstream of Atonal (Ato) proneural gene function in Drosophila sensory neurons. Unexpectedly, many differentiation genes are activated soon after specification, even before cell cycle exit and overt neuronal differentiation. Prominent among them are genes required for construction of the ciliary dendrite. Ato activates differentiation both directly and indirectly via several intermediate transcriptional regulators, including Rfx and a new Forkhead family factor. Our analysis of these factors and their regulation provides insight into how proneural factors regulate neuronal subtype differentiation. Investigating the molecular mechanisms of peripheral nervous sytem development in Drosophila melanogaster. Affymetrix Drosophila version 2.0 chips used to measure gene expression from GFP+ and GFP- cells from embryos expressing GFP under the control of the atonal gene enhancer in both wild type and mutant embryos.
