Project description:A homologue of the transcriptional repressor ScNrg1 described previously in the budding yeast and Candida albicans NRG1 wich plays an essential role in repressing hyphal development in both Saccharomyces cerevisiae and C. albicans, was found in Ustilago maydis. In S. cerevisiae it regulates a set of stress-responsive genes, also in Cryptococcus neoformans is involved in pathogenesis. In this work, we describe the effect of the absence of the U. maydis NRG1 gene in cell response to acid pH, mannosylerithritol lipids production, and cellular response to several stressful conditions. In U. maydis, NRG1 is required for filamentous growth and appears to be essential to respond to pH changes, and oxidative stress accurately. By comparing gene expression in a wild type strain versus nrg1 mutant strain of the fungus trough RNA_Seq analyses, it turned out to act as transcriptional factor altering the expression of 368 genes (205 up-regulated, 163 downregulated). Most relevant cellular processes affected by NRG1 are osmotic stress pathway, pH response, internal environmental sensor mechanism, represented by the genes Hog1, Rim101, and WCO1 respectively, also, all the genes present in the mannosylerithritol lipids production pathway, even under conditions not favorable for the production of glycolipids. Among previous specific functions described before for this transcriptional regulator as a glucose repressor, it seems to have an important role in metabolic adaptation, cellular transport, cell rescue defense and interaction with the environment.

Project description:The fungal pathogen Ustilago maydis establishes a biotrophic relationship with its host plant maize. Hallmarks of the disease are large plant tumors in which fungal proliferation occurs. Plants have developed various defense pathways to cope with pathogens. We used microarrays to detail the global programme of gene expression during the infection process of Ustilago maydis in its host plant to get insights into the defense programs and the metabolic reprogramming needed to supply the fungus with nutrients. Project description:The sexual cycle of Ustilago maydis includes the succession of a saprophytic haploid yeast stage to a virulent hyphal dikaryotic stage, product of the mating of sexually compatible yeast cells. This dimorphic transition may be replicated in vitro by different means. Recently, it was shown that ethanol induced filamentous growth on solid medium, and we observed that this process also occurred in liquid media. Since the utilization of a six- or a two-carbon source involves different metabolic pathways, we considered that the morphogenetic process might be associated with this alteration. Accordingly, we analyzed the transcriptome of U. maydis grown in glucose or in ethanol using the Illumina RNA Seq. Around 18 million reads were obtained from each treatment. From the 6788 U. maydis genes, 542 were differentially expressed (158 upregulated and 384 downregulated) during incubation with ethanol compared to glucose-grown cells, revealing a noticeable alteration in the metabolism of the fungus through their adaptation to either carbon source. The present phenomenon is a further example of how an alteration in a two-dimensional mechanism (metabolism) can affect a three-dimensional process (cell morphogenesis).

Project description:The role of the Ustilago maydis putative homolog of the transcriptional repressor ScNRG1, previously described in Saccharomyces cerevisiae, Candida albicans and Cryptococcus neoformans, was analyzed by means of its mutation. In S. cerevisiae this gene regulates a set of stress-responsive genes, and in C. neoformans it is involved in pathogenesis. It was observed that the U. maydisNRG1 gene regulates several aspects of the cell response to acid pH, such as the production of mannosyl-erythritol lipids, inhibition of the expression of the siderophore cluster genes, filamentous growth, virulence and oxidative stress. A comparison of the gene expression pattern of the wild type strain versus the nrg1 mutant strain of the fungus, through RNA Seq analyses, showed that this transcriptional factor alters the expression of 368 genes when growing at acid pH (205 up-regulated, 163 down-regulated). The most relevant genes affected by NRG1 were those previously reported as the key ones for particular cellular stress responses, such as HOG1 for osmotic stress and RIM101 for alkaline pH.
