Project description:The interactions between tumor cells and the host vasculature and immune cells results in increased net fluid production via enhanced plasma extravasation into the pleural space.Understanding the regulatory mechanism of IL-10 in MPE formation is essential both for deciphering how IL-10 acts in regulating tumor immunity and for discovering key molecular targets for intervention in advanced cancers. We investigate the characteristic of microRNAs (miRNAs) in IL-10 induced murine MPE. Identifing the distinct classes of up-regulated and down-regulated miRNAs during this process.

Project description:Malignant pleural effusion (MPE) is a common medical problem caused by multiple malignancies especially lung cancers and always along with a poor therapeutic outcome.Interlukin-10 (IL-10) promotes the formation and development of malignant pleural effusion (MPE). In the current study, our results showed that IL-10 deficiency decreased the percentage of macrophages in mice MPE and regulated M1/M2 polarization in vivo and vitro. The migration capacity of tumor cells was suppressed, and apoptosis was promoted when co-cultured with MPE macrophages in the absence of IL-10.

MPE-seq, a New Method for the Genome-wide Analysis of Chromatin Structure

Project description:In this study we developed MPE-seq, a method for the genome-wide characterization of chromatin that involves the digestion of nuclei with methidiumpropyl-EDTA-Fe(II) [MPE-Fe(II)] followed by massively parallel sequencing. Like micrococcal nuclease (MNase), MPE-Fe(II) preferentially cleaves the linker DNA between nucleosomes. We also performed MNase-seq as a comparison. We further performed ChIP-seq using chromatin samples obtained by MPE-Fe(II) or MNase digestion of nuclei. Nuclei from J1 mouse embryonic stem cells were treated with MPE-Fe(II) or MNase. The isolated DNA was sequenced by Illumina HiSeq sequencers. Some of the digested chromatin was studied by performing ChIP-seq using antibodies against histone H2B or H3.

We used microarrays to detect gene expression changes in directly ex vivo sorted CNS IL-10+ and IL-10- CD8 T cells from a neurotropic J2.2-V-1-infected mouse.

Project description:Ex vivo generated tolerogenic dendritic cells (tDCs) have a strong therapeutic potential to induce antigen-specific iTreg upon infusion in patients. We previously demonstrated that IL-10 tDC-primed T cells are very suppressive and produce IL-10. Here, we show that the majority of the IL-10+ T cells co-express IFNγ, giving rise to the question whether these cells are proinflammatory or regulatory. Whole genome gene expression analysis revealed a strong regulatory gene profile and a suppressed Th1 gene profile for the IL-10/ IFNγ co-expressing CD4+ T cells.
