Project description:The interaction between cancer and stroma plays a key role in tumor progression. Inactivation of p53 is often observed in stromal cells surrounding in cancer, suggesting that p53 in fibroblasts is involved in tumor progression. To elucidate the mechanism by which p53 loss in fibroblasts effect on proliferation and invasiveness of cancer cells, we performed comprehensive expression profiling analyses between p53 knockdown and control fibroblasts using GeneChip Human Genome U133 plus 2.0 arrays. Intact human fetal lung normal diploid fibroblasts TIG-7 cells and TIG-7 cells infected with lentiviruses for the expression of shRNA against p53 were used for RNA extraction and hybridization on Affymetrix microarrays.

RNA-seq profiling of ITPKC overexpression in normal human lung fibroblasts

Project description:Idiopathic pulmonary fibrosis (IPF) is characterized by pathogenic accumulation of scar tissue in the lung. Many of the processes implicated in fibrosis also occur following pneuomonectomy, but fibrotic changes are absent. We aimed to characterize differences between the responses to pneumonectomy and bleomycin, a fibrosis-inducing drug. We identified Itpkc as a gene uniquely up-regulated following pneumonectomy and identified it to have anti-fibrotic effects in cell culture. We then used RNA-sequencing on normal human lung fibroblasts overexpressing ITPKC or control to gain a deeper understanding of its role.

Project description:Patients with Lymphangioleiomyomatosis (LAM) develop nodules of cells within the lung parenchyma. These lesions contain both TSC2-/- LAM cells, and wild type fibroblasts. We investigated the effect of TSC2-/- cells on the transcriptional profile of both normal fibroblasts and fibroblasts isolated from LAM patient tissue (LAFs) using Affymetrix Gene Chip arrays, to identify upregulated pathways which could contribute to LAM pathology. We used Affymetrix microarrays to detail the changes in the global profile of gene expression in fibroblasts following co-culture with 621-101 cells, and identified upregulation of several chemokines, indicating that the fibroblasts had adopted an inflammatory phenotype.
