Project description:Cucurbitacin C (CuC), a novel analogue of triterpenoids cucurbitacins, confers a bitter taste in cucumber. Genes and signaling pathways responsive for biosynthesis of CuC have been identified in the recent years. In the present study, we explored the anti-cancer effects of CuC against human cancers in vitro and in vivo. CuC inhibited proliferation and clonogenic potential of multiple cancer cells in a dose dependent manner. Low dose CuC treatment induced cell cycle arrest at G1 or G2/M stage in different cancer lines, whereas high dose treatment of CuC caused apoptosis in cancer cells. PI3K-Akt signaling pathway was found to be one of the major pathways involved in CuC-induced cell growth arrest and apoptosis by RNA-Seq and western blotting. Mechanistic dissection further confirmed that CuC effectively inhibited the Akt signaling by inhibition of Akt phosphorylation at Ser473. In vivo CuC treatment (0.1 mg/kg body weight) effectively inhibited growth of cancer cells derived xenograft tumors in athymic nude mice and caused significant apoptosis. Our findings for the first time demonstrated the potential therapeutic significance of CuC against human cancers.

Project description:Streptomyces has the largest repertoire of natural product biosynthetic gene clusters (BGCs), yet developing a universal engineering strategy for each Streptomyces species is challenging. Given that some Streptomyces species have larger BGC repertoires than others, we hypothesized that a set of genes co-evolved with BGCs to support biosynthetic proficiency must exist in those strains, and that their identification may provide universal strategies to improve the productivity of other strains. We show here that genes co-evolved with natural product BGCs in Streptomyces can be identified by phylogenomics analysis. Among the 597 genes that co-evolved with polyketide BGCs, 11 genes in the “coenzyme” category have been examined, including a gene cluster encoding for the co-factor pyrroloquinoline quinone (PQQ). When the pqq gene cluster was engineered into 11 Streptomyces strains, it enhanced production of 16,385 metabolites, including 36 known natural products with up to 40-fold improvement and several activated silent gene clusters. This study provides a new engineering strategy for improving polyketide production and discovering new biosynthetic gene clusters.

Project description:Natural products represent a rich source for antibiotics addressing versatile cellular targets. The deconvolution of their targets via chemical proteomics is often challenged by the introduction of large photocrosslinkers. Here we select elegaphenone, a largely uncharacterized natural product antibiotic bearing a native benzophenone core scaffold, for affinity-based protein profiling (AfBPP) in Gram-positive and Gram-negative bacteria. This study utilizes the alkynylated natural product scaffold as a probe to uncover intriguing biological interactions with the transcriptional regulator AlgP. Furthermore, proteome profiling of a Pseudomonas aeruginosa AlgP transposon mutant revealed unique insights into the mode of action. Elegaphenone enhanced the killing of intracellular P. aeruginosa in macrophages exposed to sub-inhibitory concentrations of the fluoroquinolone antibiotic norfloxacin.

A resilient mutualistic interaction between cucumber mosaic virus and its natural host Arabidopsis halleri to adapt to an environmental change

Project description:A pseudometallophyte Arabidopsis halleri is frequently found to be infected with cucumber mosaic virus (CMV) in its natural habitat. The purpose of this study is to elucidate the effect of CMV infection on its natural hosts. The CMV(Ho) strain isolated from A. halleri was inoculated into clonal A. halleri plants that were obtained from runners of mother plants, and the pathosystem consisting of CMV(Ho) and its natural host A. halleri was established.In low heavy metal environment, the CMV(Ho) infection caused growth retardation in the above-ground part (stems and leaves) of host plants, and thereby conferred strong drought tolerance on host plants. On the other hand, in high heavy metal environment, which simulates a natural habitat for A halleri, the CMV(Ho) infection did not cause any symptoms to host plants and conferred mild drought tolerance. And the result of transcriptome analysis suggests that CMV(Ho) is recognized as a symbiont rather than a pathogen by its host plant. These results indicate a resilient mutualistic interaction between CMV(Ho) and its natural host A. halleri to adapt to an environmental change.
