Transcriptional response of Enterococcus faecali to the glycosylated bacteriocin glycocin F

Project description:Glycocin F (GccF) is a bacteriocin produced by Lactobacillus plantarum which causes susceptible cells to enter bacteriostasis within minutes of exposure. GccF is diglycosylated with two N-acetylglucosamine (GlcNAc) moieties and is active against strains of Lb. plantarum, as well as potential pathogens such as Enterococcus faecalis and E. faecium. Its mechanism of action, however, is unknown. To better understand how GccF inhibits growth, we carried out RNA sequencing (RNA-seq) on E. faecalis strain JH2-2 exposed to different concentrations of GccF for different lengths of time. Here we demonstrate that free GlcNAc both protects E. faecalis from the effects of GccF, as well as pre-sensitises it to GccF. Transcriptional analysis showed that exposure to free GlcNAc resulted in up-regulation of a GlcNAc-specific phosphotransferase system (PTS) transporter, which is a putative GccF receptor. Time course analysis showed over 100 genes were up- or down-regulated within 10 minutes of exposure, a number that increased to over 300 genes after 40 minutes. A large number of these genes were found to be regulated by the alternative sigma factor 54, which was shown to not be required for susceptibility to GccF. Interestingly, genes encoding the V-type ATPases and sodium:proton antiporters were found to be up-regulated at all times and all concentrations of GccF tested. This suggests an indirect response to GccF, possibly due to low-level permeabilization of the cell membrane.

Project description:To further investigate the homeostatic response of E. faecalis to Fe exposure, we examine the whole-genome transcriptional response of wild-type (WT) exposed to non toxic Fe excess. This experiment correspond the work titled Transcriptomic response of Enterococcus faecalis to iron excess (work in preparation) A four chip study using total RNA recovered from four separate wild-type cultures of Enterococcus faecalis OG1RF, two controls samples (N medium growth) and two iron samples (N medium gowth with 0.5 mM Fe-NTA). Each chip measures the expression level of 3,114 genome genes from Enterococcus faecalis strain V583 (A7980-00-01).

Global transcriptional response to copper of Enterococcus faecalis wild-type and copY mutant

Project description:We examined the whole-genome transcriptional response of E. faecalis wild-type (WT) and copy mutant cells exposed to non toxic Cu excess. The analysis was focused in those genes which changed significantly their expression levels in the Cu-treated cells when compared to their untreated counterparts. The results indicated that after 3 h at 0.5 mM CuSO4, WT cells exhibited a significant increment of Cu content and the induction of three genes of cop operon along with other 142 genes. The comparison with the copY mutant exposed to Cu showed an extensive overlapping of transcriptional changes suggesting that other transcription factors may be mediating the response to Cu. Transcriptional profiling of intestinal epithelial cells in response to Enterococcus gallinarum MRx0518

Project description:In this study we describe the immunostimulatory properties of Enterococcus gallinarum MRx0518, a candidate live biotherapeutic agent with anti-tumorigenic properties. Here we demonstrate that strain MRx0518 elicits a strong pro-inflammatory response in intestinal epithelial cells. Treatments with metabolically active cells, inactivated cells or culture supernatants were found to induce distinct host responses. Microarray analysis was used to investigate and compare changes in the gene expression of intestinal epithelial cells in response to live MRx0518, heat-killed MRx0518 and MRx0518 culture supernatant.

Caenorhabditis elegans infected with Enterococcus

Project description:Young adult N2 Caenorhabditis elegans were infected with Enterococcus faecalis or Enterococcus faecium for 8 h to determine the transcriptional host response to each enterococcal species. Analysis of differential gene expression in C. elegans young adults exposed to four different bacteria: heat-killed Escherichia coli strain OP50 (control), wild-type E. faecalis MMH594, wild-type E. faecium E007, or Bacillus subtilis PY79 (sigF::kan). Samples were analyzed at 8 hours after exposure to the different bacteria. These studies identified C. elegans genes induced by pathogen infection. Brain-heart infusion agar plates (10 ug/ml kanamycin) were used.
