Project description:The objective of the present study is to identify early miRNAomic biomarkers of response to a hypocaloric mediterranean diet (RESMENA study). The RESMENA study (“REducción del Síndrome Metabólico en Navarra”) is a Spanish, randomized controlled and longitudinal trial over a six-month period that aimed to improve the parameters related with Metabolic Syndrome, including body fat composition, biochemistry, inflammation, or oxidative stress. The study was approved by the Research Ethics Committee of the University of Navarra (ref.065/2009). All the participants (n=96) were Caucasian and gave written informed consent for participation in agreement with the Declaration of Helsinki (www.clinicaltrials.gov; NTC01087086). Participants were categorized into “High Responders” (HR), when weight loss was >8%, and “Low Responders” (LR) when weight loss was <8%. White blood cells were separated from whole blood by centrifugation at 1,100 g at 4ºC for 15 min. Total RNA from WBCs isolated before the nutritional intervention was extracted using TRIzol reagent. miRNAs from white blood cells of 6 LR and 5 HR were sequenced using Illumina’s miRNA-Seq following the standardized protocol.

Mediterranean Diet and Weight Loss: Targeting the Bile Acid/Gut Microbiome Axis to Reduce Colorectal Cancer

Project description:A Mediterranean Diet (MedDiet), a largely plant-based dietary pattern, is relevant to CRC prevention and microbial production of anti-cancer metabolites in observational studies. A MedDiet can shift BA metabolism as shown in primates and when combined with calorie restriction, shows superior adherence and weight control in humans, given its palatability. To date, no studies have tested in an RCT the effects of a MedDiet alone (MedA), WL through lifestyle intervention (WL-A) or a calorie-restricted MedDiet for WL (WL-Med) on the BA-gut microbiome axis and its relevance to CRC prevention among AAs. A multidisciplinary team combining expertise in psychology, nutrition, microbiology, molecular cell biology, computational biology, medicine and biostatistics, proposes to conduct a four-arm RCT in which 232 obese AAs, 45-75 years old complete one of the following 6-month interventions: Med-A, weight stable; WL-A, calorie restriction with no diet pattern change; WLMed; or Control. The investigators will use samples and data collected at baseline, mid-study (month-3) and post-intervention to compare the effects of the interventions on 1) Concentration and composition of circulating and fecal BAs; 2) Gut microbiota and metabolic function; and 3) Gene expression profiles of exfoliated intestinal epithelial cells.

Project description:Genome wide DNA methylation profiling of white blood cells in obese patients under two different diets (The American Heart Association's Diet and RESMENA diet (Zulet MA et al., Nutr Hosp. 2011 Jan-Feb;26(1):16-26. )) in order to reduce weight. The Illumina infinium HumanMethylation450 beadchip kit was used, which assayed 450,000 potential methylation sites. Bisulphite converted DNA from the 48 samples were hybridised to the Illumina Infinium 450 Human Methylation Beadchip.

Prospective analyses of white adipose tissue gene expression in relation to long-term body weight changes

Project description:The aim of this study was to investigate transcriptome signatures in human subcutaneous white adipose tissue (sWAT) before and after long-term weight changes and assess their predictive value for body weight change. Long-term body weight change in adult women associates with altered sWAT transcriptome. Expression of genes associated with inflammation, insulin response, adipogenesis and lipogenesis are linked to weight loss. However, other pathways such as protein homeostasis not only associate with but also predict future weight gain suggesting that intrinsic factors in sWAT impact tissue expansion.

Project description:Genome wide expression profiling of white blood cells in obese patients under two different diets (The American Heart Association's Diet and RESMENA diet (Zulet MA et al., Nutr Hosp. 2011 Jan-Feb;26(1):16-26. )) in order to reduce weight. The Illumina HumanHT-12 v4 Expression BeadChip Kit was used, which assayed 47,000 probes.A total of 24 RNA was purified from white blood cells using a TRIZOL RNA Isolation Protocol (Life technologies) and 500 ng of starting material were used as input for the Illumina TotalPrep Amplification Kit. Next, The Illumina GenomeStudio Gene Expression Software Module (v 1.9.0, Illumina, USA) was used to extract the signal intensities of each gene expression probe, which were then further preprocessed using the lumi package of Bioconductor in R. The preprocessing included background correction of the gene expression data, followed by log2-transformation and quantile normalization. It also automatically removes the control probes, leaving only the regular probes. We keep probes that are expressed in at least one individual according to a detection p-values of 5%. Assays containg SNPs within the probe were removed.
