Project description:A mouse parabiotic model supports the existence of transferable factor(s) in blood that rescue(s) age-related degenerative phenotypes, however, whether it prevents endothelial cell senescence linked to common cardio-cerebral vascular diseases, requires to be inspected. In this study, plasmas donated from 27 young (23.04 ± 4.86 years, young human plasma, YHP) and 26 elderly (55.04 ± 5.42 years, elderly human plasma, EHP) healthy volunteers were used to culture primary human umbilical endothelial cells (HUVEC). When comparing with EHP, we found that the YHP reduced the percentages of SABG-stained cells and prohibited the expressions of senescent markers, including p16, p21, and p53 in HUVEC. Functionally, the YHP decreased the expression of ICAM-1, a marker for endothelial cell activation and increased abilities in tube formation and mobility at late passages that represent an enhanced angiogenic function. Validated by both RNA sequencing and Real-time PCR in HUVECs from all three umbilical cords, nineteen up-regulated and fifteen down-regulated genes were observed in YHP-treated HUVEC compared with group treated with the EHP. Among 34 genes, the expressions of MCM10, DHCR24, WBP1, UNC5B, and CYP26B1 were reversed by the YHP and we showed that these reversed expressions delayed senescence of both HUVEC and human coronary artery endothelial cell (HCAEC) through p16- and/or p21-assocated signaling. The five genes are relevant to maintain or reinstate the ability for proliferation, anti-apoptosis, and endothelial cell related functions, such as angiogenesis and anti-atherosclerosis. Thus, our study suggests that YHP restores biological processes in senescent endothelial cell through multiple pathways.

Project description:Laminar shear stress due to constant blood flow is known to play a critical role in maintaining vascular health. In contrast, endothelial cell senescence appears to be closely associated with the incidence of vascular disorder. In an attempt to identify functional biomarkers for age-related vascular health/disease, the present study investigated differential gene expression of young and senescent human umbilical vein endothelial cells (HUVECs) under static and laminar shear stress. We used a cDNA microarray method to compare gene expression profiles of young and senescent HUVECs under static and laminar shear stress conditions. Experiment Overall Design: Senescent cells were prepared by continuous subculture in vitro, and a cone-and-plate device was used to impose laminar shear stress onto cells. Young and senescent cells were exposed to laminar shear stress or maintained under static conditions. Total mRNA was extracted and gene expression profiles were analyzed by cDNA microarray.

Project description:Proton pump inhibitors (PPIs) are among the most frequently prescribed drugs, especially in older people. Although these drugs are usually considered safe, recent evidence suggests that high dose and/or long term use of PPIs may have several detrimental effects, including increased risk of adverse cardiovascular events. The impact of PPI in the aging host environment still need to be characterized. Aged tissues, including vascular tissues, accumulate senescent cells that can communicate with their environment by secreting a myriad of cytokines and growth factors. Human coronary artery endothelial cells (HCAECs) provide an excellent model system to study â??in vitroâ?? most aspects of cardiovascular function and disease related to cellular senescence. The purpose of this study is thus to investigate the in vitro effects of two well-known PPIs (Omeprazole and Lansoprazole) on endothelial gene expression in senescent e non-senescent HCAECs. We used a cDNA microarray method to compare gene expression profiles of young and senescent HCAECs treated with omeprazole and lansoprazole. Young cells were cultured in medium supplemented with vehicle (CTRL) or 100μM PPIs (Omeprazole or Lansoprazole) and grown for subsequent passages until they evidenced senescence-associated phenotypes. Total mRNA was extracted and gene expression profiles were analyzed in senescent endothelial cells (P12) compared to the non-senescent cells (P6) in both untreated (CTRL) and PPI-treated groups by cDNA microarray. All experiments were performed in triplicate for each treatment group.

Project description:Heart failure with preserved ejection fraction (HFpEF) is characterised by a distinct but poorly understood in skeletal muscle pathology. In a rat model ZSF1, we identify impaired myonuclear accretion as a mechanism for ablated myofiber growth in HFpEF following resistance exercise intervention. Muscle vascular or mitochondrial dysfunction, or impaired protein synthesis signalling, do not appear as primary limiting mechanisms, and cardiac therapies do not attenuate skeletal muscle pathology. However, we identify that acute caloric restriction rejuvenates myofiber growth in HFpEF during resistance exercise intervention.
