Transcriptome analysis among young and senescent fibroblasts

ABSTRACT: We carried out the analysis using human skin fibrobllast HCA2 (MJ90) cells. Cells were indcued to be senescent cells by treatment with RO3306 and Nutlin3a. The transcriptome analysis between young cells (uniduced cells) nd senescent cells revealed lonrf2, a ubiqutin E3 ligase, is one of the genes highly expressed in senescnet cells.

Project description:For senescence induction, HCA2 humn skin fibroblasts were synchronized at G2 phase by treatment with 9 uM of RO3306 (Roche) for 24 hrs, treated with 9 uM of RO3306 and 5 uM of Nutlin3a (Sigma-Aldrich) for 8 hrs, and then with 5 uM of nutlin3a for 18 hrs. Cells were then treated with 100 nM of BI-2536 for 9 days to eliminate proliferating cells, and cultured in normal media for 9 days. The senescent and non-senescent (young) cells were collected, washed with PBS, snap-frozen, and analyzed with a iMPAQT method.

Bulk RNA-seq of Normal and Doxorubicin-Induced Senescent HCA2 cells

Project description:Purpose: Use RNA-seq strategy to characterize the PD-L1 correlated genes in sorted PD-L1 negative and positive doxorubicin-induced senescent and normal human foreskin fibroblasts HCA2 cells to identify the upstream transcriptional network for regulating heterogeneous PD-L1 expression in senescent cells. Methods: Early passaged normal HCA2 cells and senescent HCA2 cells induced by 24 hours of 100 nM doxorubicin treatment were utilized for the preparation of bulk RNA-seq libraries. Among the senescent cells, PD-L1 positive and negative cells were separated by FACSorting.

Circulating senescent myeloid cells drive blood brain barrier breakdown and neurodegeneration

Project description:Neurodegenerative diseases (ND) are characterized by a progressive loss of neuronal function. Mechanisms of ND pathogenesis are incompletely understood hampering the development of effective therapies. Langerhans cell histiocytosis (LCH) is an inflammatory neoplastic disorder caused by hematopoietic progenitors expressing MAPK activating mutations that differentiate into senescent dendritic cells that drive formation of lesions. Some patients with systemic LCH subsequently develop progressive and incurable neurodegeneration (LCH-ND). Here, we show that LCH-ND is caused by hematopoietic cells that are clonal with systemic LCH lesion histiocytes. Strikingly, we discovered that circulating BRAFV600E+ myeloid cells cause the breakdown of the blood-brain barrier (BBB), enabling migration of into the brain where they differentiate into senescent, inflammatory macrophages that accumulate in the brainstem and cerebellum. Blocking MAPK activity and senescence programs synergistically reduced parenchymal infiltration, neuroinflammation and neurologic damage in preclinical LCH-ND. MAPK activation in circulating myeloid cells represents a novel and targetable mechanism of ND.

Methods: MEFs were isolated from p16-CreERT2/rtTA3-mKate2/TRE-OSKM-mCherry mice. After the senescence induction through RO3306 and nutlin-3a, treatments with 4-OHT for 9 days and doxycycline for 12 days were performed to induce OSKM expression. The doxycycline treated MEFs and untreated MEFs were collected by trypsinization and directly used for scRNA library preparation through droplet system.
