Project description:Rats were trained to orally self-administer alcohol in a concurrent, two-lever, free-choice contingency using a modification of the sweet solution fading procedure (O'Dell et al., 2004; Roberts et al., 2000; Vendruscolo et al., 2012). Following acquisition of self-administration, rats were allowed to self-administer unsweetened alcohol (10%) for 4 weeks and were then assigned to two groups matched by levels of responding: one group (dependent group) was exposed to chronic, intermittent ethanol vapors for 4 weeks to induce dependence; the other group (nondependent group) was not exposed to ethanol vapor. After a month of vapor exposure, rats were again tested during acute withdrawal (6-8 hours after removal from the vapor chambers) until stable levels of alcohol intake were achieved. As expected, alcohol vapor-exposed rats self-administered significantly greater amounts of alcohol than control rats not exposed to alcohol vapor during acute withdrawal. Rats were sacrificed during protracted abstinence (3 weeks after the end of alcohol vapor exposure) along with age-matched alcohol naive rats. 96 gene expression profiles (GEP) were obtained from 8 brain regions believed to be relevant in alcoholM-bM-^@M-^Ys reinforcing properties using the Affymetrix RN230.2 platform. Specifically, the following brain regions were microdissected and analyzed from nondependent and dependent alcohol self-administering rats as well as age-matched alcohol naive rats: (a) medial prefrontal cortex (MPF), (b) shell and (c) core NAc sub-regions, (d) central nucleus (CeA) and (e) basolateral nucleus of the amygdala (BLA), (f) dorsolateral and (g) ventral bed nucleus of the stria terminalis (BNST), and (h) ventral tegmental area (VTA).

Project description:Oligodendrocytes (OLs) are glial cells within the central nervous system (CNS) that produce myelin, which coats neuronal axons and enhances fast propagation of neuronal signals. OLs are known to be severely affected by alcohol in fetal alcohol spectrum disorder (FASD) and alcohol use disorder (AUD). In FASD, oligodendrocyte precursor cells have been reported to be highly affected by any dose of alcohol. In AUD, high doses of alcohol can lead to demyelinating complications such as Wernicke-Korsakoff sybdrome. However, relatively little is known about how alcohol at low and high doses affects mature OLs (mOLs). To better understand the impact of concentration-dependent alcohol exposure on mOLs, we isolated and differentiated immature murine OLs into mOLs and exposed them to a moderate or high concentration of alcohol in vitro. RNA was then isolated from the cultures and sequenced using 3’-Tag RNA sequencing. Differential expression across alcohol concentration and control groups was calculated, which was used for gene set enrichment analysis to identify KEGG and GO terms that were significantly changed as a result of alcohol treatment. We found concentration dependent changes in biological pathways related to cell cycle regulation, translation initiation, integrin signaling, FGF2 signaling, inflammation, and myelination. Further, we found that the moderate and high concentrations of alcohol elicited distinct changes in gene expression, indicating that concentration is a critical factor when determining the effects of alcohol on transcriptomic changes.

Project description:In the adult brain, epigenetic control of gene expression has important roles in the processing of neural activity. Emerging evidence suggests that epigenetic regulation is dependent on metabolic state, implicating specific metabolic factors in neural functions that drive behavior. In neurons, histone acetylation is dependent on the metabolite acetyl-CoA that is produced from acetate by chromatin-bound ACSS21. Here, using in vivo stable isotope labeling in mouse, we show that alcohol metabolism rapidly fuels histone acetylation in the brain by direct deposition of alcohol-derived acetyl groups onto histones in an ACSS2-dependent manner. A similar induction was observed with heavy labeled acetate injection in vivo. Injection of labeled alcohol into a pregnant mouse results in incorporation of labeled acetyl groups into gestating fetal brains, indicating that the acetate passes through the placenta. In isolated primary hippocampal neurons ex vivo, extracellular acetate induced learning and memory-related transcriptional programs that were sensitive to ACSS2 inhibition. Strikingly, alcohol-related associative learning requires ACSS2 in vivo. These findings establish a novel and direct link between alcohol metabolism and neuronal ACSS2-dependent histone acetylation in the brain, providing evidence that dynamic acetate release from liver metabolism may travel to the brain for direct epigenetic regulation in neurons.
