Project description:To analyze the molecular function of BROWNING of CALLUS1 (BOC1), we performed RNA-seq experiments using callus that had been subcultured for 21 days from Teqing and YIL25, Teqing and the complementation line pCPL-BOC1-Teqing (pCPL), and also Teqing and the overexpression line pOE-BOC1-Teqing (pOE).We identified a total of 700 differentially expressed genes (DEGs), containing 410 DEGs upregulated in the callus of YIL25 versus Teqing, pCPL versus Teqing and downregulated in pOE versus Teqing. Besides, 290 DEGs downregulated in the callus of YIL25 versus Teqing and pCPL versus Teqing and upregulated in pOE versus Teqing. A large number of DEGs associated with stress response and cell death were presented.

Project description:We previously reported that Def (Digestive-organ expansion factor) was a pan-endodermal enriched factor that is essential for the growth of digestive organs in zebrafish using a def mutant line hi429 as model (Chen et al., 2005). To further elucidate Def function, we generated a Def over-expressed zebrafish line, namely Tg (fabp10a:def)-I, in which def expression was under the control of a liver-specific promoter fabp10a. We used microarrays to detail the global programme of gene expression in Tg (fabp10a:def)-I transgenic line compared with wild type zebrafish control, and identified distinct classes of differently regulated genes in Tg (fabp10a:def)-I line.

The transgenic 1D1a mice were generated on the genetic background of the B10.D2(R101) line and expressed in their T cells the α-chain TCR (TCRα) specific to the allogeneic MHC molecule Н2-Kb. To evaluate effects of TCRα transgenesis on the qualitative and quantitative composition of plasma proteins, comparative proteome analysis was performed for both female and male transgenic 1D1a and wild-type B10.D2(R101) mice aged 3-, 6-, and 12-months.

Project description:We used the NGS techonoly to compare the gene’s transcripts at rice whole genome levels of the WT (XS134) and OsPEP1 Ri1 transgenic line in response to the exogenous PEP1 signal. We found that the expgenous PEP1-mediated signal were blocked in OsPEP1 Ri1 transgenic line, but the application of exogenous PEP1 could restore this PEP1-mediated signal to the WT (XS134) level.

Gene expression profiling in leaf and panicles of non-transgenic and HMB4 over-expression line.

Project description:The profiling was conducted with the Rice 3'-Tiling 135k Microarray designed from 31,439 genes deposited at IRGSP, RAP2 database (http://rapdb.lab.nig.ac.jp). In this research, an array of 31,439 rice genes was used to elucidate gene expression in leaf and panicles of non-transgenic and HMB4 over-expression line. The analyses show that transgenic rice induces early flowering due to an enhancement of stress response.
