Effects of the bovine SLICK1 mutation in PRLR on sweat gland area and FOXA1 abundance and on global gene expression

Project description:The SLICK1 mutation confers thermotolerance to cattle inheriting one or two copies of the gene. Results are unclear as to whether the mutation changes capacity of animals to undergo sweating during heat stress. Accordingly, differences in characteristics of sweat glands between slick and wildtype Holstein heifers was determined. There were no differences in the proportion of skin occupied by sweat glands but sweat glands from slick heifers had higher amounts of immunoreactive FOXA1 than wildtype heifers. FOXA1 is a transcription factor important for sweating. While results do not support the idea that the SLICK1 mutation changes the abundance of sweat glands in skin, it did affect functional properties of sweat glands, as indicated by increased abundance of immunoreactive FOXA1.

Identification of proteins in the plasma associated with heifer infertility

Project description:We carried out an experiment to test the hypothesis that differences in protein abundance due to fertility fitness would be shared between heifers of different breeds. We produced proteome data from the plasma collected from 22 heifers (purebred Angus heifers (n=12), purebred Holstein (n=10) heifers). The protein Alpha-ketoglutarate-dependent dioxygenase FTO was more abundant in the plasma collected from Fertile heifers relative to Sub-fertile counterparts (FDR < 0.05).

Project description:Pu-erh tea has attracted increasing attention worldwide because of its special flavor and health effects, but its impact on composition and function of the gut microbiota remains unclear. The aim of this study was to investigate effects of aqueous extracts of fermented (ripe) and non-fermented (raw) Pu-erh teas on the composition and function of intestinal microbiota of rats with diet-induced obesity. We conducted a comparative metagenomic and metaproteomic investigation of the microbial communities in cecal samples taken from obese rats administrated with or without extracts of raw and ripe Pu-erh tea. By analyzing the composition and diversity of 16S rRNA amplicons and expression profiles of 814 distinct proteins, we found that, despite differences in the chemical compositions of the raw and ripe Pu-erh tea, administration of either at two different doses (0.15 and 0.40 g/Kg body weight), significantly (P<0.05) increased community diversity, and changed the composition of the cecal microbiota by increasing the relative abundances of Firmicutes and decreasing those of Bacteroidetes. Community metabolic processes including sucrose metabolism, glycolysis, syntheses of proteins, rRNA and antibiotics were significantly (P<0.05), or had a tendency (0.10<P<0.05) to be, promoted by enriching relevant enzymes. Furthermore, evidences from population, molecular and metabolic levels have shown that polyphenols of raw Pu-erh tea and their metabolites can promote potentially the growth of Akkermansia municiphila by stimulating the type II and III secretion system protein, elongation factor Tu, and glyceraldehyde-3-phosphate dehydrogenase. This study has provided new evidences for the prebiotic effects of Pu-erh tea.

Project description:<p>Global climate change has led to an increase in release of atmospheric CO2 and average daily temperatures, making heat stress a major challenge in dairy farming. The present study aimed to investigate metabolite changes of Holstein cows at different stages. In this experiment, 10 growing heifers, 10 heifers and 10 lactating cows were selected for the current study. Blood samples were separately collected from the animals through the caudal root vein in April and August. Samples were transferred to a liquid nitrogen tank for metabolomic analysis.</p>

However, underlying mechanistic links between thermal insult in fetal life and postnatal outcomes are not completely understood. Utilizing RNA Sequencing, this study characterized the mammary gland transcriptome of heifers that were gestated under maternal conditions of heat stress or cooling, i.e., in utero heat stressed (HT) vs. in utero cooled (CL). Mammary tissues were collected from three HT and three CL heifers during their first lactation.

Liver samples from bull calves (CT = 5 and HT = 4) were collected at birth while mammary gland samples from heifers (CT = 3 and HT = 3) were collected during their first lactation.
