Project description:Objective: To study the physiological role of eosinophils in the GI tract and lung under homeostatic conditions, Method: we analyzed tissue-specific eosinophil gene expression patterns by genome-wide expression microarray analysis using RNA isolated from FACS-sorted eosinophils from the small intestine or lung of naive BALB/cmice (Live eosinophils were identified as DAPI-CCR3+Siglec-F+CD45+CD4-CD8a-CD19-B220-SSChigh cells). Result: There are 513 genes that are differentially expressed by intestinal eosinophils and Lung eosinophils. Project description:As opposed to their well-characterized contributions to inflammatory processes, tissue eosinophils are now also thought to contribute to immune homeostasis at mucosal sites such as the gut. Yet, whether such steady-state eosinophils exist in the lung is currently unclear. In this project, we identified and characterized lung resident eosinophil and also studied what happen to these cell during a mouse model of allergic inflammation (House dust mite (HDM) exact administrations). We compared the transcriptional profile of lung resident eosinophil from naive mice (rEOS ss) or from HDM-treated mice (rEOS i) and of inflammatory eosinophil (iEOS) from HDM-treated mice.

Project description:Background: HAS2 is a member of the gene family encoding hyaluronan synthase 2 (HAS2) which can generate high molecular weight hyaluronan (HMW-HA). Although we previously reported that HAS2 is a novel candidate gene for susceptibility to adult asthma., little is known about whether HAS2 dysfunction affect eosinophilic airway inflammation. Objective: We therefore hypothesized that attenuation of HAS2 will enhance eosinophilic airway inflammation. Methods: C57BL/6 wild type (WT) mice, Has2 heterozygous deficient (Has2+/−) mice were used in eosinophilic airway inflammation model which induced by ovalbumin (OVA). Quantitative reverse transcription polymerase chain reaction (qRT-PCR) was performed to detect Has2 and HA binding protein mRNA expression levels. Lung tissue and lavage fluid (BALF) were analyzed for inflammation and various cytokines and chemokines. Airway resistance was measured using forced oscillation technique. gene expression analyses were also performed to elucidate further pathogenesis. Results: The expression levels of Has2 mRNA was significantly decreased in OVA stimulated Has2+/− (Has2+/−-OVA) mice. Has2+/−-OVA mice also displayed significant reduce of CD44, and TGF-beta1 mRNA expression. BALF eosinophil number, levels of various Th2 cytokines and chemokines in BALF, and airway responsiveness were significantly increased in Has2+/−-OVA mice compared with similarly treated WT mice. ILK Signaling and PKA signaling were downregulated significantly more in Has2+/−-OVA mice compared with similarly treated WT mice. Conclusions: Has2 dysfunction induce more intense allergic eosinophilic airway inflammation and increase of airway hyper responsiveness with impairment of HAS2-CD44-TGF-beta signaling. Modulating HAS2 signaling might provide novel therapeutic targets for intractable bronchial asthma patients.

Project description:Eosinophils are important in fighting parasitic infections and are implicated in the pathogenesis of asthma and allergy. Interleukin-5 (IL-5) is a critical regulator of eosinophil development, controlling proliferation, differentiation and maturation of the lineage. Mice that constitutively express IL-5 have more than 10 fold more eosinophils in the haematopoietic organs than their wild type counterparts. We have identified that much of this expansion is in a population of Siglec-F high eosinophils, which are rare in wild type mice. In this study we assessed transcription in myeloid progenitors, eosinophil precursors and Siglec-F medium and Siglec-F high eosinophils from IL-5 transgenic mice and in doing so have created a useful resource for eosinophil biologists. We have then utilised these populations to construct an eosinophil trajectory based on gene expression and to identify gene sets that are associated with eosinophil lineage progression. Cell cycle genes were significantly associated with the trajectory, and we experimentally demonstrate an increasing trend towards quiescence along the trajectory. Additionally we found gene expression changes associated with constitutive IL-5 signalling in eosinophil progenitors, many of which were not observed in eosinophils.

Project description:Background. Nuclear erythroid 2 p45-related factor 2 (Nrf2) regulates the expression of antioxidant and detoxification genes and are thought to be important in protection against intracellular pathogens. The aim of this study was to determine the role of Nrf2 in the host defense against Mycobacterium avium complex (MAC) infection in mice. Methods. Wild-type mice and Nrf2-deficient mice were infected with MAC via intranasal inoculation with 1x107 CFU of Mycobacterium avium subsp. hominissuis. At 2 months after infection, the bronchoalveolar lavage fluid, lung tissues were collected, then, comprehensive transcriptome analysis in the lungs was performed by RNA-seq. Results. Nrf2-deficient mice were highly susceptible to MAC, compared with wild-type mice. There was no significant changes between genotypes in the level of oxidative stress. In addition, the expression of genes related to Th1 immunity and the proportion of Th1 cells were not changed between genotypes. Comprehensive transcriptome analysis showed that the expression of Nramp1 was much lower in the infected lungs of Nrf2-deficient mice than those of Wild-type mice. The expression of Nramp1 was also much lower in the infected alveolar macrophages of Nrf2-deficient mice than those of Wild-type mice. The result of electron microscopy showed that many infected alveolar macrophages from Nrf2-deficient mice were found to contain a large number of intracellular MAC bacteria compare to alveolar macrophages from wild-type mice, due to inhibition of phagolysosome fusion. Conclusions. This study identified that the Nramp1 regulated by Nrf2 is essential in defining the outcome of MAC disease. Nrf2 is a critical determinant for host resistance to MAC infection as a regulator for the expression of Nramp1.
