Project description:The blood brain barrier (BBB) acts both a physical as well as a metabolic barrier isolating the Central Nervous System (CNS) from systemic circulation, blocking intravascular toxin entry and promoting toxic substance elimination to blood stream. The BBB is formed by four main cellular elements: endothelial cells, astrocyte end-feet, microglial cells, and pericytes. There is strong evidence that astrocytes are able to regulate the phenotypes of endothelial cells by directly contacting or secreting a range of chemical agents, resulting in BBB permeability changes and so on. Conversely, endothelium-derived leukemia inhibitory factor (LIF) has been shown to induce astrocytic differentiation, from studies in normal condition of cell culture. However, in pathological conditions, the specific regulatory mechanism that endothelial cells exert on astrocytes is still less known. In our recently study, a transwell-based, endothelial-astrocyte co-culture model was established to investigate this regulatory relationship in bacterial infection. An extraintestinal pathogenic Escherichia. coli strain was used to infect human brain microvascular endothelial cells (hBMEC) planted in the upper chamber, astrocytes co-cultured in the bottom well were actived without direct contact with bacteria or endothelial cell. The transcription levels of some chemokines and cytokines such as TNF-α and CXCL10 were significantly upregulated. We hypothesized that the secreted proteins from endothelial cells is the main cause of astrocyte activation and the culture supernatant of hBMEC infected by E.coli was obtained to perform the mass spectrometry analysis. In summary, the proteomic identification results showed that 2,692 common proteins were identified in the two groups of control and infected hBMEC, including 140 differentially expressed proteins. The bioinformatics analysis indicated that differentially expressed proteins are mainly classified into structural molecular activity and constituent of ribosome. KEGG pathway enrichment analysis showed that many proteins are involved in focal adhesion as well as a bunch of signaling pathways, PI3K-Akt, Rap1, Ras, MAPK and NF-kappaB signaling pathways, for example. The present study laid an important foundation for the subsequent study of secreted proteins of hBMEC in bacterial infections.

Project description:To understand the mechanism by which MBG increases permeability of human brain microvascular endothelial cell (HBMEC) monolayers, the primary cultures were treated with 10nM MBG for 12 h. RNA was isolated and analysed for gene expression on Agilent one color microarrays. Triplicate wells of HBMEC were treated with 10 nM MBG in DMSO or with just the drug vehicle for 12 h. RNA was isolated from each well and used to reverse transcribe and generate Cy3-labelled cRNA. Microarrays were hybridized, washed and scanned. Data were normalized and assessed for differential gene expression between MBG treated and control HBMEC.

Project description:Increase in frequency and size of wildfire raises a significant public health concern due to rapid climate change. Epidemiological studies have shown that wildfire smoke exposure can increase the risk of neurological disorders. Our study aims to understand how wildfire smoke particles affect brain endothelial cells using both primary human brain microvascular endothelial cells (HBMEC) and an immortalized human cerebral microvascular endothelial cell line (hCMEC/D3). HBMEC and hCMEC/D3 were exposed to different levels of smoldering eucalyptus smoke particles (10, 30, 50 ug/mL) for 24 hr. Supernatants were collected for LDH and ELISA array, cell lysates were collected for RNAseq, and cells were fixed and stained for tight junction marker (Zonula Occludens-1, ZO-1) via ICC. Our study found that treatment to smoldering eucalyptus particles can increase inflammatory cytokine production dose-dependently in HBMECs and hCMEC/D3. Moreover, RNAseq analyses resulted in dose-dependent changes in HBMEC and hCMEC/D3 from wood smoke treatment that could be mediated via AhR and Nrf2 pathways.

Project description:Exosomes are small vesicles with a diameter of approximately 30-140nm. This double-layer phospholipid membrane encapsulates a variety of proteins, lipids and nucleic acids, which are produced and released by cells under specific conditions, participating in a large scale of regulatory processes. There is accumulating evidence indicating that exosomes also play an indispensable regulatory function in the central nervous system under physiological and pathological conditions, such as promoting communication between various cells in the blood-brain barrier. In our study, we attempted to obtain exosomes of human brain microvascular endothelial cells (hBMEC) infected by an extraintestinal pathogenic Escherichia. coli strain as well as control cells, and mass spectrometry analysis was performed to study the role of encapsulated proteome in central nervous system inflammation caused by bacterial infection. The particle size analysis results show that the size of the exosomes we isolated is mainly distributed between 30-200nm. Tetraspanin TSG101, CD9, and cytoplasmic protein Alix that we selected as positive markers for exosomes were successfully detected, while the negative marker, the endoplasmic reticulum-specific expression protein GRP94, was not found. In summary, the proteomic identification results showed that 1162 common proteins were identified in the two groups of exosomes from control and infected hBMEC, including 59 differentially expressed proteins. (Proteins with expression change more than 2 times or less than 0.5 times and P value less than 0.05 are regarded as differentially expressed proteins). The bioinformatics analysis indicated that differentially expressed proteins are mainly classified into structural molecular activity and constituent of ribosome. KEGG pathway enrichment analysis showed that many proteins are involved in ribosome-related pathways, pathogenic E. coli infections, and PI3K-Akt Signal pathways, etc. The present study laid an important foundation for the subsequent study of exosomal proteins in bacterial infections.

Project description:Cerebral malaria is a severe complication of Plasmodium falciparum infection characterized by the loss of blood-brain barrier (BBB) integrity, which is associated with brain swelling and mortality in patients. P. falciparum-infected red blood cells and in!ammatory cytokines, like tumor necrosis factor alpha (TNF-a), have been implicated in the development of cerebral malaria, but it is still unclear how they contribute to the loss of BBB integrity. Here, a combination of transcriptomic analysis and cellular assays detecting changes in barrier integrity and endothelial activation were used to distinguish between the effects of P. falciparum and TNF-a on a human brain microvascular endothelial cell (HBMEC) line and in primary human brain microvascular endothelial cells. We observed that while TNF-a induced high levels of endothelial activation, it only caused a small increase in HBMEC permeability. Conversely, P. falciparum-infected red blood cells (iRBCLs) led to a strong increase in HBMEC permeability that was not mediated by cell death. Distinct transcriptomic pro"les of TNF-a and P. falciparum in HBMECs con"rm the differential effects of these stimuli, with the parasite preferentially inducing an endoplasmic reticulum stress response. Our results establish that there are fundamental differences in the responses induced by TNF-a and P. falciparum on brain endothelial cells and suggest that parasite-induced signaling is a major component driving the disruption of the BBB during cerebral malaria, proposing a potential target for much needed therapeutics.

A cDNA microarray imprinted with 2156 genes representing about 98% of Streptococcus suis serotype 2 genome was used for transcriptome analysis . For two-sample (reference vs. test) microarray hybridization, Two-condition experiment, control vs contact with hBMEC 3 h, four replicates (two biological replicates, two technical replicates) at each condition.
