Project description:In this study, we have utilized a single-cell RNA-Sequencing platform coupled with transgenic lineage tracing in order to follow the progression of different embryonic neural stem cell populations as they progress into adult dormant neural stem cells. We show that adult dormant V-SVZ neural stem cells of different embryonic origins share a common molecular signature and reacquire an embryonic precursor-like state when activated to make new neurons in the adult brain.

Project description:Here we used single cell RNA-sequencing and single cell spatial transcriptomics to characterize the forebrain neural stem cell (NSC) niche under homeostatic and injury conditions. We define the dorsal and lateral ventricular-subventricular zones (V-SVZ) as two distinct neighborhoods, and show that following white matter injury, dorsal NSCs are locally activated to make oligodendrocytes for remyelination. This activation is coincident with a robust increase in transcriptionally-distinct microglia in the dorsal V-SVZ niche. We modeled ligand-receptor interactions within this changing niche and identified two remyelination-associated microglial ligands, IGF1 and OSM, that promote precursor proliferation and oligodendrogenesis in culture. Infusion of either ligand into the lateral ventricles also enhanced oligodendrogenesis, even in the lateral V-SVZ, where NSCs normally make neuroblasts. These data support a model where gliogenesis versus neurogenesis is determined by the local NSC neighborhood and where injury-induced niche alterations promote NSC activation, local oligodendrogenesis, and likely contribute to myelin repair.

Androgen regulation of genes in rat forebrain, prostate, ovary, and skin

Project description:The goal of this study was to identify androgen regulated genes in the forebrain, prostate, ovary, and skin. The results of this study will be used to make comparisons between androgen regulation of gene expression in different androgen-receptor expressing tissues, and will contribute to our understanding of tissue-selective androgen activity. RNA was extracted from tissues castrate male rodents after 16 hours of treatment of either vehicle (Ctrl) or DHT. The 4 tissues of extraction are Forebrain (ForeB.Ctrl & ForeB.DHT), Ovary (Ovary.Ctrl & Ovary.DHT), Prostate (Prost.Ctrl & Prost.DHT), and Skin (Skin.Ctrl & Skin.DHT). There are 3 biological replicates per group for a total of 24 samples.

Project description:AimTo assess whether amplitude-integrated electroencephalography (aEEG) alterations in the newborn period are associated with poor precursor skills of literacy at five years of age in children born preterm.MethodsBetween October 2007 and September 2011 248 preterm infants were eligible for the study at Innsbruck Medical University Hospital. aEEG was analysed for dominating background activity, calculation of the percentage of continuous activity, the Burdjalov scoring system, the minimum, mean and maximum amplitude. At the age of five years, we evaluated preterm born children by the Bielefelder screening (BISC) to assess for early diagnosis of reading problems and weak spelling and classified them as normal performers (n = 64) or poor performers (n = 20). Completion of testing was not possible for one infant.ResultsThe minimum amplitude was significantly lower in the poor BISC performance group as compared to the normal BISC performance group at postnatal week two. The percentage of continuous background activity was significantly higher in infants with normal BISC performance than in infants with poor BISC performance at postnatal week three.ConclusionChildren with poor developed precursor skills of literacy showed alterations in aEEG signals. The aEEG could be useful in further diagnosing preterm infants at risk for developmental complications.
