Project description:Androgens have a strong effect against skeletal muscles to increase muscle mass and strength. However, a molecular mechanism of AR action on muscle strength is not clear. To identify the target genes of AR in skeletal muscle, we generated myofiber specific ARKO using HSA-Cre and AR flox mice (cARKO). Nine-week-old female control and cARKO mice were treated with or without DHT for 4 weeks. After euthenization, gastrocunemius muscle were collected and total RNA were extracted.

PRR14 mediates mechanotransduction and regulates myofiber identity via MEF2C in skeletal muscle

Project description:Sarcopenia, characterized by the loss of muscle mass, strength, and function, predisposes adverse outcomes and its mechanism is waiting to reveal. Here, we report decrease of PRR14, a nuclear protein, in skeletal muscle results in sarcopenia. Genetically, genome-wide association studies (GWAS) identified multiple single nucleotide polymorphisms (SNPs) in PRR14 locus associated with body mass index (BMI) and total body lean mass, which indicated its association with sarcopenia; Specific knockout of skeletal muscle Prr14 in mice confirmed the causal effect; Biochemical analysis and high-throughput sequencing, including both transcriptome and approaches for the study of the epigenome (CUT&Tag sequencing and ATAC sequencing), revealed that Prr14 was required for myofiber homeostasis in skeletal muscle: Prr14 loss altered chromatin structure and reduced Mef2c activity, which in combination resulted in failure of maintaining myofiber identity and therefore sarcopenia. Our findings demonstrate that PRR14 orchestrates critical epigenetic changes and transcription factor activity to maintain myofiber identity, thereby providing novel therapeutic avenues for skeletal muscle pathologies associated with dysregulation of these mechanisms.

Project description:Skeletal muscles are complex organs consisting of multiple tissues, including connective tissue, blood vessels, nerves and multinucleated myofibers. Single-nucleus transcriptomics analyses have revealed distinctive myonuclear populations related to myofiber type and to specialized regions, including neuromuscular and myotendinous junctions. However, the use of tissue dissociation methods prevented access to global spatial organization. Here we applied RNA tomography (TOMOseq), a spatial transcriptomics approach, to explore gene expression differences along murine tibialis anterior muscles.

Project description:TEI-SARM2 is a novel selective androgen receptor modulator. To assess the effects of TEI-SARM2 on skeletal muscle, we generated myofiber specific ARKO using HSA-Cre and AR flox mice (mARKO). Male mARKO and littermate control (ARL2/Y) mice underwent ORX or not. Nine days after ORX, the mice were treated with TEI-SARM2 (10 mg/kg weekly, p.o.) or vehicle (weekly, p.o.) for another 2 weeks. After euthanasia, the bulbocavernosus (BC) muscles were collected and total RNA was extracted.

Myofiber androgen receptor increases muscle strength mediated by a skeletal muscle splicing variant of Mylk4.

Project description:Androgens have a robust effect on skeletal muscles to increase muscle mass and strength. The molecular mechanism of androgen/androgen receptor (AR) action on muscle strength is still not well known, especially for the regulation of sarcomeric genes. In this study, we generated androgen-induced hypertrophic model mice, myofiber-specific androgen receptor knockout (cARKO) mice supplemented with dihydrotestosterone (DHT). DHT treatment increased grip strength in control mice but not in cARKO mice. Transcriptome analysis by RNA-seq, using skeletal muscles obtained from control and cARKO mice treated with or without DHT, identified a fast-type muscle-specific novel splicing variant of Myosin light-chain kinase 4 (Mylk4) as a target of AR in skeletal muscles. Mylk4 knockout mice exhibited decreased maximum isometric torque of plantar flexion and passive stiffness of myofibers due to reduced phosphorylation of Myomesin 1 protein. This study suggests that androgen-induced skeletal muscle strength is mediated with Mylk4 and Myomesin 1 axis.
