Transcriptome Profile Analysis Reveals the Regulation Mechanism of Stamen Abortion in Handeliodendron bodinieri（Levl.）Rehd

Project description:To elucidate the molecular mechanism of stamen abortion underlying sex differentiation from transcriptomic aspects in Handeliodendron bodinieri, we use abnormal stamens before and after abortion in female flowers and normal stamen in male flowers at the same stage as control material. Key genes regulating stamen abortion was identified by using RNA-seq technique. The results show that it was abortion science stage 6 during anther development, at the same time, HbGPAT, HbAMS, HbLAP5, HbLAP3 and HbTES were down-regulated , and HbML5 was up-regulated. Therefore, these information will provide theoretical foundation for the conservation, breeding, scientific research and application of Handeliodendron bodinieri.

A distinct immature low-density neutrophil population characterizes acute generalized pustular psoriasis [PBMC]

Project description:Generalized pustular psoriasis (GPP) is a severe disease featured by neutrophilic pustules and enhanced IL-36 inflammatory pathway in skin. Recently, a transcriptomic analysis of PBMCs and neutrophils in MPO-deficient GPP patients has been performed in stable disease state. However, transcriptomic profiling of PBMCs during acute flare of GPP is unclear. Here, we reported a predominant neutrophil signature in GPP PBMC and a marked increase in the CD66+CD16+ low-density neutrophils (LDNs) within the PBMC fraction of acute GPP patients. Transcriptomic and functional analysis of LDNs revealed a hypoinflammatory phenotype yet enhanced release of neutrophil granule proteases, implicating that LDNs might contribute to the IL-36-mediated inflammation in GPP patients.

Comparison of healthy, plaque psoriasis and pustular psoriasis FFPE skin biopsies

Project description:Generalized pustular psoriasis (GPP) is a rare, debilitating, and often life-threatening inflammatory disease characterized by episodic infiltration of neutrophils into the skin, pustule development, and systemic inflammation, which can manifest in the presence or absence of chronic plaque psoriasis (PV). Current treatments are unsatisfactory thus a better understanding the pathogenesis of GPP is warranted. To assess the pathophysiological differences between GPP and PV we performed a gene expression study on formalin-fixed paraffin-embedded biopsies of GPP (n=30) and PV (n=12) lesions and healthy control (n=20) skin. Compared with healthy skin, GPP lesions yielded 365 and PV 898 differentially expressed genes respectively, with 190 upregulated in both diseases. We detected higher expression of IL-1 and IL-36 cytokines in GPP lesions compared with PV, and this occurred proximal to neutrophils. We show both activated neutrophils and isolated neutrophil proteases can activate IL-36. Diverging from the Th1/Th17 pathophysiology of PV, significantly fewer IL23A, IL17A, IFNG, CXCL9, CXCL10 and MX1 transcripts were detected in GPP lesions. Our data indicate a level of sustained activation of IL-1 and IL-36 in GPP, inducing neutrophil chemokine expression, infiltration and pustule formation, suggesting that the IL-1 and IL-36 inflammatory axes are the main drivers of disease pathology in GPP.

Transcriptional analysis of epithelial cells in grapefruit peel

Project description:Epithelial cells (EC) lining the most inner layers of secretory glands in Citrus peel are hypothesized to be the specialized cells that synthesize citrus essential oil. The major biosynthetic pathway(s) for essential oil are therefore postulated to be specifically and highly active in EC; transcripts that are involved in the pathway(s) are expected to be highly up-regulated in the cells as well. We performed cell-specific transcriptional analysis using GeneChip Citrus Genome Arrays to probe the global gene expression in EC during initial stage of essential oil biosynthesis and to identify groups of highly expressed genes in the EC. Grapefruits of two different sizes (diameters at 28 mm and 41 mm), at which points where biosynthesis of essential oil entering its active phase, were selected for the purpose of this experiment. For comparison, we selected parenchyma cells (PC), the non-oil-biosynthesizing cells, as the control cell type. Grapefruit peels were fixed, embedded in paraffin, sectioned and mounted on slides prior to cells isolation using laser microdissection and pressure catapulting. RNA was extracted from the isolated cells and then used for microarray hybridization. Transcriptional analysis were carried out by comparing transcript profiles between different cell types of the same time points, and same cell type at the different time points.

Project description:We find that two SAM synthases in C. elegans, SAMS-1 and SAMS-3/4, contribute differently to modification of H3K4me3, gene expression and survival in the heat stress response. Our data showing differential SAM provisioning to distinct groups of methylated targets based on SAM synthase source suggests that determining how methyl donors are provided will broaden insight into 1CC functions in aging and stress.

A comprehensive microarray analysis of Enterococcus faecium in response to vancomycin in combination with the essential oil components carvacrol and cuminaldehyde

Project description:This study aims to determine the global gene expression in vancomycin resistant Enterococcus faecium (VRE) in response to a novel essential oil-vancomycin combination, and the individual components (vancomycin, carvacrol and cuminaldehyde) to help determine the mechanism of action of this antimicrobial formulation. This formulation increases the susceptibility of VRE to vancomycin and the array provides data on the synergistic mechanism of action. Five conditions (1. Control; 2. Carvacrol, 1.98 mM; 3. Cuminaldehyde, 4.20 mM; 4. Vancomycin, 0.031 mg/l; 5. Combination, 1.98 mM Carvacrol, 4.2 mM Cuminaldehyde, 0.031 mg/l vancomycin) all with 1% DMSO were tested in triplicate with a 60 minute exposure time before extraction.
