Project description:Genome wide DNA methylation profiling of paired muscle tissue and cultured myoblast genomic DNA. The Illumina Infinium Methylation450 array was used to obtain DNA methylation profiles across approximately 450,000 CpGs. Samples were male, taken from the Hertfordshire Sarcoepnia Study exension (HSSe), to determine prelimnary associations measures of muscle mass, function and insulin/glucose levels and DNA methylation in muscle tissue. Furthermore, we sought to determine whether DNA methylation in the muscle tissue is conserved in cultured myoblasts from the muscle biopsies and whether association differed between the two tissues.

Epigenome analysis of muscle tissue samples from male participants of the Hertfordshire Sarcopenia Study (HSS) and Hertfordshire Sarcopenia Study extension (HSSe)

Project description:Genome wide DNA methylation profiling of muscle tissue genomic DNA. The Illumina Infinium MethylationEPIC array was used to obtain DNA methylation profiles across approximately 850,000 CpGs. Samples were male, taken from both the Hertfordshire Sarcopenia Study (HSS, n=34) and the Hertfordshire Sarcoepnia Study exension (HSSe, n=44), to determine DNA methylation associations with measures of muscle mass and function. We further aimed to test whether differences in the muscle methylome are associated with sarcopenia in older individuals. We found significant associations between DNA methylation in the muscle tissue and sarcopenia, as well as individual measures of muscle mass and function, enriched around EZH2 binding sites.

Transcriptome analysis of aging muscle progenitors (human myoblasts)

Project description:Although sarcopenia is evident from the fifth decade, mechanisms leading to this phenomenon start earlier, emphasizing the importance of defining biomarkers related to the onset of muscle weakness. To this aim, a transcriptome analysis will be performed on muscle cell cultures (myoblasts) obtained from adult donors of different ages. Any biomarkers identified by this analysis will be confirmed by western blot and immunostatining. We will then assess the role of those biomarkers in muscle aging. Human myoblasts were extracted from the quadriceps of young (15-20 years old) and elderly (>70 years old) healthy subjects . The muscle cell population has been sorted using CD56 MACS beads. The myogenicity and the life span analysis of each muscle cell extract have been determined.
