Structural and functional dissection of Pacinian corpuscle reveals a multicellular mechanism of touch detection independent of the outer core

Project description:Lamellated corpuscles are rapidly adapting mechanoreceptors that detect transient touch and high-frequency vibration in many vertebrates. It the prevailing model, these properties are determined by outer core, which acts as a mechanical filter that limits static and low-frequency stimuli from reaching afferent terminal—the sole site of touch detection in corpuscles. Using focused ion beam scanning electron microscopy and single corpuscle electrophysiology, we determine the detailed architecture of corpuscular components and reveal their contribution to touch detection. We show that outer core is dispensable for rapid adaptation and frequency tuning. Instead, these properties are encoded within inner core, composed of gap junction-coupled syncytium formed by lamellar Schwann cells (LCS) around afferent terminal. By acting as additional touch sensors, LSCs potentiate mechanosensitivity of the terminal, which detects touch via fast-inactivating ion channels. Our findings support a model in which lamellated corpuscle function is mediated by a bicellular mechanism within inner core.

Architecture and function of lamellar cells in Meissner corpuscle

Project description:Meissner corpuscles detect transient touch and vibration in glabrous skin, but their ultrastructure and mechanism of function are poorly understood. The Meissner corpuscle sensory core contains the terminal neurite(s) of the mechanoreceptor afferent surrounded by Schwann cell-derived lamellar cells. The afferent is thought to be the sole touch-sensing element within the corpuscle, whereas the role of lamellar cells is unclear. Here, we present a high-resolution three-dimensional ultrastructure of an avian Meissner (Grandry) corpuscle in intact skin acquired using the enhanced focused ion beam scanning electron microscopy (FIB-SEM), followed by machine learning based segmentation and reconstruction. We show that the afferent splits within the corpuscle into several disk-shaped endings layered between flattened lamellar cells. Each lamellar cell contains numerous exocytotic dense core vesicles and forms large-area contacts with adjacent afferent disks. Together with single-corpuscle RNA sequencing and electrophysiology, our FIB-SEM data reveal lamellar cells as secretory sensors of touch. We developed a method for direct electrophysiological recordings of afferent activity from an individual corpuscle in the skin and show that activation of a single mechanosensitive lamellar cell is sufficient to trigger action potentials in the afferent. These results reveal comprehensive Meissner (Grandry) corpuscle architecture and demonstrate a dual sensory mechanism comprised of a mechanoreceptor afferent and lamellar cells.

A MYC2, MYC3, MYC4-dependent transcription factor network regulates touch responses in plants

Project description:Plants are continuously exposed to environmental triggers, including mechanical stimulation. Our results demonstrate that jasmonic acid (JA)-signalling not only plays a key role in touch-induced developmental changes, but also in the very early gene expression changes. Using multi-omics, we show that the JA-activated transcription factors MYC2/MYC3/MYC4 co-regulate touch-induced gene expression of 266 genes. MYC2/3/4 particularly activate top touch-induced genes, which peak around 25 minutes and then rapidly decline by 40-60 minutes. ChIP-seq shows that MYC2 dynamically binds hundreds of touch-induced promoters within 25 minutes. Furthermore, promoter activation assays confirm that MYC2 directly activates touch-induced promoters. By combining these data, we identified a core MYC2/3/4-dependent ‘touch regulon’, containing many previously-unknown MYC2 targets like bHLH19 and ERF109. bHLH19 can in turn directly activate the ORA47 promoter, indicating that MYC2/3/4 initiate a hierarchical network of downstream transcription factors. Finally, hormone profiling shows that the rapid touch-induced accumulation of JA/JA-isoleucine is directly controlled by MYC2/3/4 in a positive amplification loop regulating JA-biosynthesis genes.

The star-nosed mole reveals clues to the molecular basis of mammalian touch.

Project description:Little is known about the molecular mechanisms underlying mammalian touch transduction. To identify novel candidate transducers, we examined the molecular and cellular basis of touch in one of the most sensitive tactile organs in the animal kingdom, the star of the star-nosed mole. Our findings demonstrate that the trigeminal ganglia innervating the star are enriched in tactile-sensitive neurons, resulting in a higher proportion of light touch fibers and lower proportion of nociceptors compared to the dorsal root ganglia innervating the rest of the body. We exploit this difference using transcriptome analysis of the star-nosed mole sensory ganglia to identify novel candidate mammalian touch and pain transducers. The most enriched candidates are also expressed in mouse somatosesensory ganglia, suggesting they may mediate transduction in diverse species and are not unique to moles. These findings highlight the utility of examining diverse and specialized species to address fundamental questions in mammalian biology.
