Project description:Since we reasoned that RiNLE1 exerts its functions inside host nuclei, we explored the influence of RiNLE1 on plant gene expression. Therefore, we applied a dexamethasone (DEX) inducible promoter system (Borghi, 2010) to induce RiNLE1 expression (lacking signal peptide) in Medicago roots. As a control we replaced the RiNLE1 coding sequence by a short nonsense sequence.

Project description:Effector-Triggered Immunity (ETI) and Pattern-Triggered Immunity (PTI) are well-defined modes of plant immunity triggered by recognition of pathogen effector proteins and microbe-associated molecular patterns, respectively. While ETI and PTI network extensively share signaling components, the shared components are used in different ways, resulting in distinct network properties in the model plant Arabidopsis: immunity is highly robust against network perturbations in ETI but relatively sensitive in PTI. However, the molecular mechanism how the shared network leads to the different properties is not known. Here we show that sustained MAPK activation compensate salicylic acid (SA) signaling. A 12 DNA microarray study using total RNA from Arabidopsis transgenic plants carrying DEX-inducible MKK4DD in Col or sid2-2 background treated with DEX or control.

IgE Effector Mechanisms, in Concert with Mast Cells, Contribute to Acquired Host Defense against Staphylococcus aureus

Project description:Best known for their roles in allergic diseases, the physiologic function of immunoglobulin E (IgE) and mast cells (the so-called allergy module of immunity) has been enigmatic. Recent evidence shows that allergic reactions can help to protect against the toxicity of venoms. As bacteria are a potent alternative source of toxins, we assessed the potential role of the allergy module in antibacterial host defense. We observed that the immune response against S. aureus skin infection includes specific IgEs and substantially improves systemic host defense against secondary S. aureus infections in mice. This acquired protection depends on functional IgE effector mechanisms and mast cells. Our results reveal the powerful antibacterial potential of the allergy module and therefore a novel physiologic function of IgEs and mast cells.

Effects of ACTH and DEX on gene expression in rat kidneys

Project description:The left kidneys of 8 Sprague-Dawley rats were removed and the rats were left for 2 weeks to recover. After 4 additional days (during which blood pressure measurements were obtained), they were injected daily subcut. with either saline (1ml/kg), adrenocorticotropic hormone (ACTH, 0.2mg/kg) or dexamethasone (DEX, 0.02mg/kg) for 8 days before the right kidney was removed. ACTH and DEX treatment caused hypertension in the rats as illustrated by increased systolic blood pressure. The gene expression patterns of the kidneys were analysed using Affymetrix arrays.
